Identification and Characterisation of an Old Yellow Enzyme (OYE) - NamA from Listeria monocytogenes by Majid Abdullah Bamaga A thesis submitted in partial fulfilment of the requirements of Edinburgh Napier University, for the award of Doctor of Philosophy September, 2014 Abstract The food-borne pathogene Listeria monocytogenes has been considered a significant threat to human health worldwide. It mainly infects individuals suffering insuffecint immunity such as pregnant women. During pregnancy, L. monocytogenes is capable of causing a serious damage to the mother and the fetus. It can spread to different organs including the placenta via adaptation to interacellular lifestyle. To maintain pregnancy, the levels of the hormones progesterone and β-estradiol increase and reduction in hormone levels was proposed to be associated with fetal death and abortion. The objectives of this project therefore were to investigate the role of pregnancy hormones on the growth and virulence of L. monocytogenes, and to identify bacterial genes with possible roles in binding to pregnancy hormones. It was obsereved that the growth of L. monocytogenes in the presence of progesterone under anaerobic condition was affected by the action of the hormone and the effect was dose/time-dependent of exposure as increasing concentrations showed greater effect on the bacterial growth. Interestingly, bacterial growth was restored within 24 h of exposure to the hormone. In parallel, a Tn917-LTV3 insertion library was constructed and a number of mutants isolated that had reduced growth in the presence of β-estradiol were identified. However, reduction in growth was not microbiologically significant. Furthermore, bioinformatics analysis was performed to identify listerial genes with possible role in hormones degradation. -

Sofos Doreene Hyatt ABSTRACT OF THESIS DETECTION AND MOLECULAR CHARACTERIZATION OF LISTERIA MONOCYTOGENES AND OTHER LISTERIA SPECIES IN THE PROCESSING PLANT ENVIRONMENT Listeria monocytogenes is the causative agent of listeriosis, a severe foodborne disease associated with a high case fatality rate. To prevent product contamination with L. monocytogenes, it is crucial to understand Listeria contamination patterns in the food processing plant environment. The aim of this study was to monitor Listeria contamination patterns for two years in six small or very small ready-to-eat (RTE) meat processing plants using a routine combined cultural and molecular typing program. Each of the six plants enrolled in the study were visited on a bi-monthly basis for a two-year period where samples were collected, microbiologically analyzed for Listeria and isolates from positive samples were characterized by molecular subtyping. Year one of the project focused only on non-food contact environmental samples within each plant, and year two focused again on non-food contact environmental samples as well as food contact surfaces and finished RTE meat product samples from participating plants. Between year one and year two of sampling, we conducted an in-plant training session ii involving all employees at each plant. -

It is an opportunistic intracellular pathogenic microorganism that has become crucial reason for human food borne infections worldwide. It causes Listeriosis, the disease that can be serious and fatal to human and animals. Listeria outbreaks are often linked to dairy products, raw vegetables, raw meat and smoked fish, raw milk. The most effected country by Listeriosis is United States. CDC estimated that 1600 people get Listeriosis annually and regarding 260 die. It additionally contributes to negative economic impact because of the value of surveillance, investigation, treatment and prevention of sickness. The analysis of food products for presence of pathogenic microorganisms is one among the fundamental steps to regulate safety of food. This article intends to review the status of its introduction, characteristics, outbreaks, symptoms, prevention and treatment, more importantly to controlling the Listeriosis and its safety measures. Roberts Professor of Food Science Head of the Department *Signatures are on file in the Graduate School ii ABSTRACT A longitudinal survey of non-food-contact surfaces in a commercial mushroom slicing and packaging facility was conducted to determine the prevalence, distribution, and potential routes of transmission of L. monocytogenes. Samples were taken at 3 sampling occasions over a 13- month period. Multi-virulence-locus sequence typing (MVLST) was used to identify persistent and transient L. monocytogenes clones. The longitudinal study demonstrated that L. monocytogenes was present in 18.8% of samples. A trench drain and a wet porous concrete floor were identified as harborage sites for L. monocytogenes. MVLST identified 4 different virulence types (VTs) of L. monocytogenes in the facility: VT11, VT107, VT105, and VT56. Of these clones, VT11 predominated and was persistent at 2 sampling sites during all sampling periods over 13 months.
