It can spread to different organs including the placenta via adaptation to interacellular lifestyle. To maintain pregnancy, the levels of the hormones progesterone and β-estradiol increase and reduction in hormone levels was proposed to be associated with fetal death and abortion. The objectives of this project therefore were to investigate the role of pregnancy hormones on the growth and virulence of L. monocytogenes, and to identify bacterial genes with possible roles in binding to pregnancy hormones. It was obsereved that the growth of L. monocytogenes in the presence of progesterone under anaerobic condition was affected by the action of the hormone and the effect was dose/time-dependent of exposure as increasing concentrations showed greater effect on the bacterial growth. Interestingly, bacterial growth was restored within 24 h of exposure to the hormone. In parallel, a Tn917-LTV3 insertion library was constructed and a number of mutants isolated that had reduced growth in the presence of β-estradiol were identified. However, reduction in growth was not microbiologically significant. Furthermore, bioinformatics analysis was performed to identify listerial genes with possible role in hormones degradation.

Year one of the project focused only on non-food contact environmental samples within each plant, and year two focused again on non-food contact environmental samples as well as food contact surfaces and finished RTE meat product samples from participating plants. Between year one and year two of sampling, we conducted an in-plant training session ii involving all employees at each plant.

This thesis investigated the culturable microbiota of select imported, RTE foods to see whether the existing bacterial microflora could inactivate, inhibit the growth and/or cause a reduction in the virulence of L. monocytogenes. Among all the foods tested (dried apple slices, cumin seeds, date fruits, fennel seeds, pistachios, pollen, raisins and seaweed), the date fruit microbiota displayed the most promise for harbouring antagonistic properties against L. monocytogenes. Of the 191 isolates recovered from five different date fruits, 36 (19%) produced zones of inhibition against L. monocytogenes that ranged from 0.3 to 5.8 mm. The inhibitory strains were all identified as Bacillus spp. Among those Bacillus spp. that were tested for their ability to inhibit PrfA, all caused a significant reduction in the activation of the PrfA protein (p-value < 0.05). In addition, the anti-Listeria compound(s) produced by B. altitudinis DS11 were found to be proteinaceous in nature, acid and alkali-tolerant and resistant to temperature treatments up to 100oC.

D. Cornell University 2017 Bacillales, an order of Gram-positive bacteria, are commonly isolated from dairy foods and at various points along the dairy value chain. Three families of Bacillales are analyzed in this work: (i) Listeriaceae (represented by Listeria monocytogenes), (ii) Paenibacillaceae (represented by Paenibacillus), and (iii) Bacillaceae (represented by the Bacillus cereus group). These families impact both food safety and food quality. Most Listeriaceae are non- pathogenic, but L. monocytogenes has one of the highest mortality rates of foodborne pathogens. Listeria spp. are often reported in food processing environments. Here, 4,430 environmental samples were collected from 9 small cheese-processing facilities and tested for Listeria and L. monocytogenes. Prevalence varied by processing facility, but across all facilities, 6.03 and 1.35% of samples were positive for L. monocytogenes and other Listeria spp., respectively. Each of these families contains strains capable of growth at refrigeration temperatures. To more broadly understand milk spoilage bacteria, genetic analyses were performed on 28 Paenibacillus and 23 B. cereus group isolates. While no specific genes were significantly associated with cold-growing Paenibacillus, the growth variation and vast genetic data introduced in this study provide a strong foundation for the development of detection strategies.
