Wash. 47(1), 1980, p. 85-94 Two New Nematode Genera, Safianema (Anguinidae) and Discotylenchus (Tylenchidae), with Descriptions of Three New Species MOHAMMAD RAFIQ SIDDIQI Commonwealth Institute of Helminthology, St. Albans, Hertfordshire, England ABSTRACT: Safianema gen.n. is proposed under Anguininae, family Anguinidae. It differs from Di- tylenchus in having esophageal glands forming a long diverticulum over the intestine. It is compared with Pseudhalenchus whose diagnosis is amended. Safianema lutonense gen.n., sp.n. is described from peaty soil under oak in Luton, England. Safianema anchilisposomum (Tarjan, 1958) comb.n., S. damnatum (Massey, 1966) comb.n., and S. hylobii (Massey, 1966) comb.n. are proposed for species previously in Pseudhalenchus. Discotylenchus gen.n. is close to Filenchus but has a strongly tapering lip region with a distinct disc at the apex. Two new species of this genus are described—D. discretus (type species) from apple and cabbage soils at Damascus, Syria, and D. attenuatus from bush soil at Ibadan, Nigeria. From peaty soil underneath an oak tree at Luton Hoo, Luton, Bedfordshire, England, Safianema lutonense gen.n., sp.n. was collected by my daughter Safia Fatima Siddiqi; the new genus is named after her and the name is neuter in gender. Discotylenchus gen.n. is proposed under Tylenchidae for two new species, D. discretus (type species) and D. attenuatus, described below. The families Anguinidae and Tylenchidae were defined and differentiated from each other by Siddiqi (1971). Thus the genera Ditylenchus Filipjev, 1936 and Tylenchus Bastian, 1865 which were associated together under Tylenchidae for a long time were separated into two families, the former genus belonging to Anguinidae and the latter to Tylenchidae. -

Because there isn’t any research of plant parasitic nematodes on Z. zizyphus, authors were encouraged to work on it. Nematodes isolated from the soil samples by whitehead method (1965) and permanent slides were prepared. Among the species Filenchus aquilonius is redescribed for the first time from Southern Khorasan province.F. aquilonius is characterized by lip region rounded, not offset, with fine annuls; four incisures in lateral line; Stylet moderately developed, 10-11.8 µm long with rounded knobs; Hemizonid immediately in front of excretory pore; Deirids at the level of excretory pore; Spermatheca an axial chamber and offset pouch; Tail about 120-157 µm, tapering gradually to a pointed terminus. For molecular identification the large subunit expansion segments of D2/D3 were performed for F. aquilonius to examine the phylogenetic relationships with other Tylenchids. DNA sequence data revealed that F. aquilonius had closet phylogenetic affinity withIrantylenchus vicinus as a sister group and with other Filenchus species for this region and placed them in one clade with 100% for bootstap value support. -

CARTA, SHIGUANG LI, ANDREA M. SKANTAR, AND GEORGE NEWCOMBE Abstract: During a long-term, large network study of the ecology of plant endophytes in native habitats, various nematodes have been found. Two poplar species, Populus angustifolia (narrowleaf cottonwood) and Populus trichocarpa (black cottonwood), are important ecological and genomic models now used in ongoing plant–pathogen–endophyte interaction studies. In this study, two different aphe- lenchid nematodes within surface-sterilized healthy leaves of these two Populus spp. in northwestern North America were discovered. Nematodes were identified and characterized microscopically and molecularly with 28S ribosomal RNA (rRNA) and 18S rRNA molecular markers. From P. angustifolia, Aphelenchoides saprophilus was inferred to be closest to another population of A. saprophilus among sequenced taxa in the 18S tree. From P. trichocarpa, Laimaphelenchus heidelbergi had a 28S sequence only 1 bp different from that of a Portuguese population, and 1 bp different from the original Australian type population. The 28S and 18S rRNA trees of Aphelenchoides and Laima- phelenchus species indicated L. heidelbergi failed to cluster with three other Laimaphelenchus species, including the type species of the genus. Therefore, we support a conservative definition of the genus Laimaphelenchus, and consider these populations to belong to Aphelenchoides, amended as Aphelenchoides heidelbergi n. comb. This is the first report of these nematode species from within aboveground leaves. The presence of these fungal-feeding nematodes can affect the balance of endophytic fungi, which are important determinants of plant health. -

Summary -A polymerase chain reaction - restriction fragment polymorphism (PCR-RFLP) analysis was used for the discri mination of isolates of Bursaphelenchus nematode. The isolares of B. xylophilus examined originared from Japan, the United Stares, China, and Canada and the B. mucronatus isolates from Japan, China, and France. Ribosomal DNA containing the 5.8S gene, the internai transcribed spacer region 1 and 2, and partial regions of 18S and 28S gene were amplified by PCR. Digestion of the amplified products of each nematode isolate with twelve restriction endonucleases and examination of resulting RFLP data by cluster analysis revealed a significant gap between B. xylophllus and B. mucronatus. Among the B. xylophilus isolares examined, Japanese pathogenic, Chinese and US isolates were ail identical, whereas Japanese non-pathogenic isolares were slightly distinct and Canadian isolates formed a separate cluster. Among the B. mucronalUS isolates, two Japanese isolares were very similar to each other and another Japanèse and one Chinese isolare were identical to each other. The DNA sequence data revealed 98 differences (nucleotide substitutions or gaps) in 884 bp investigated between B. xylophilus isolare and B. mucronmus isolate; DNA sequence data of Aphelenchus avenae and Aphelenchoides fragariae differed not only from those of Bursaphelenchus nematodes, but also from each other. -
