Professor Legesse Negash (Advisor)_________________ ______________ 4. ______________________ (Chairman) ________________ _____________ i Propagation and management practices of coffee (Coffea arabica L.) at the coffee nursery site of Lalo kebele, Didu Wereda, Ilu-Aba-Bora Zone, Oromia Regional State BY BEDILU TEKLE Addis Ababa University, 2018 Abstract Coffee is the major agricultural export crop, providing currently 35% of Ethiopia‟s foreign exchange earnings, down from 65% a decade ago because of the slump in coffee prices since the mid-1990„s. this research work was conducted by identifying and providing the necessary information about the propagation and management practices of coffee on nursery site of Lalo Kebele, Didu Wereda, Ilu-Aba-Bora Zone, Oromia Regional State. Data were collected through semi-structured interviews, discussions with the farmers and/or development agents, and field observations. The study found that a combination of forest soil, top soil and compost promoted growth of coffee seedlings. According to the present study, sound fertilization of seedling provides the following advantages: (1) Production of healthy coffee seedlings and subsequently productive coffee plants; (2) Uniform growth; (3) Fast growth of seedlings; and, (4) High post-planting survival rate. Application of commercial fertilizer to seedlings enhanced the growth of these, especially the application of DAP. Sowing coffee seeds at a depth of 1 cm with the grooved side placed downwards and embryo tip upwards improved seed germination. Covering the seed bed with 3-5 cm mulch enhanced higher germination. Transplanting germinants to a freshly prepared nursery bed, followed by watering at two days interval, helped speed up hypocotyls emergence and subsequent seedling development. Further, providing moderate overhead shade and watering to the developing seedlings twice a week until these produced up to four leaves were found to be mandatory for a high quality seedling production. Keeping nursery hygiene and controlling seedling disease are also major activities at Lalo coffee nursery. Sowing mature, freshly collected and clean coffee seeds after soaking these in cold water for 24 hours hastened germination. It was observed that hardening is generally done by reducing the amount of watering about one month before the date of planting. J u ly 1987 2 To Joyce and Kwaku for their continuous love, care and patient endurance, during the many hours my attention was taken away from them in the course of this work, and to all those who care and sympathise with the Underdevelopment of the developing World. 3 ABSTRACT Mature apple trees were sprayed using a Knapsack mistblower fitted with 3 different nozzle systems, one of which provided an induced electrostatic charge on the spray droplets. Spray distribution was assessed by examining fluorescent tracer deposits on Kromekote cards which had been positioned in different parts of the tree canopy. Subsequently, bioassays with codling moth (neonate larvae) and formulations of cypermethrin examined the bio­ logical effect of deposits. Small uncharged droplets (30-60ym VMD) sprayed at low volume (3.0 - A.8 ml/tree) with spinning disc gave the least deposit but with an electrostatic charge, deposition was significantly improved on the outer canopy. Intermediate results were achieved when sprays were applied from an air-shear nozzle at 27A.0 ml/tree. Directing spray from both sides of the tree improved spray distribution with all three nozzles. Leaf Area Index (LAI) of the apple varieties used in the experi­ ments was estimated in relation to tree height, and was used to calculate the volumes of spray needed to improve coverage of the canopy.

Musa,1 and Dike M.C.2 Abstract: The life cycle of the Khapra beetle, Trogoderma granarium Everts (Coleoptera: Dermestidae) on stored groundnut was studied under fluctuating laboratory conditions of 25±5°C and 70±5% relative humidity. There were five larval instars and the total development time from egg to adult ranged from 37 to 40 days with an average of 37.95 days. The duration of each developmental stage was egg: 6.05 days; first instar: 3.8 days; second instar: 4.7 days; third instar: 5.6 days; fourth instar: 6.2 days; fifth instar: 6.8 days and pupa: 4.8 days. Females had an average fecundity of 80.2 eggs. Mean adult longevity was 12.4 days. Key words: Development time, fecundity, life cycle, Trogoderma granarium. Coffee belongs to the genus Coffea on the family Rubiaceae and it was believed to be originated from tropical Africa. Coffea species were grouped into four sections (Agrocoffea, Paracoffea, Mascarocoffea,Eucoffea). The two most important commercial species are Coffea arabica and Coffea canephora. All the species are diploid with 2n=2x=22 chromosomes except C arabica which is tatraploid with 2n=4x=44. Evaluation of the genetic diversity and available resources with the genus is an important step in coffee breeding. A variety of techniques like morphological, biochemical and genetic markers had been used to measure genetic variation of Coffea species. Coffea arabica has been found to have low polymorphic compared to other species. Coffee is prone to lot of diseases infestation. Two most prominent of them are coffee berry disease and coffee leaf rust which impaired photosynthesis, premature defoliation and reduced folia initiation. Coffee especially C arabica and C canephora are susceptible to insect pest. The most important insect pests are leaf miner and coffee stem borer.

The development of an improved regeneration system, based on high frequency somatic embryogenesis from leaf expiants, passing through multiplication of embryogénie callus in liquid medium, is described. This method can contribute to the obtaining of high protoplast yields and offers perspectives for use in genetic transformation systems of coffee. Several factors affecting protoplast isolation, electroporation and regeneration were studied. This system appeared to be appropriate for transient expression studiesbut , due to difficulties with protoplast regeneration, less promising for achieving stable expression. Further expression studies were performed using particle gun bombardment on different tissues of several coffee genotypes. Best results were obtained using in vitro cultured leaves of Coffea arabica and plasmids carrying the EFla-Al promoter of Arabidopsis thaliana. The effect of tungsten particles on callus induction and the fate of GUS-expressing cells after bombardement on leaves has been described, and the consequences for their use are discussed. Studies onfive selective agents showed best prospects of the herbicide glufosinate for detection of stably transformed coffee tissue.
