This is due to disruption of electron transfer from mutant POR to microsomal cytochrome P450 (CYP) enzymes that play a key role in glucocorticoid and sex steroid synthesis. POR also transfers electrons to all major drug- metabolizing CYP enzymes, including CYP3A4 that inactivates glucocorticoid and oestrogens. However, whether ORD results in impairment of in vivo drug metabolism has never been studied. Design: We studied an adult patient with ORD due to homozygous POR A287P, the most frequent POR mutation in Caucasians, and her clinically unaffected, heterozygous mother. The patient had received standard dose oestrogen replacement from 17 until 37 years of age when it was stopped after she developed breast cancer. Methods: Both subjects underwent in vivo cocktail phenotyping comprising the oral administration of caffeine, tolbutamide, omeprazole, dextromethorphan hydrobromide and midazolam to assess the five major drug-metabolizing CYP enzymes. -

All unreported estimates were added to the official data, as reported by the Syrian Arab Republic to the United Nation’s Food and Agriculture Organization (FAO). Total reconstructed catch for 1950-2010 was around 170,000 t, which is 78% more than the amount reported by Syria to the FAO as their national catch. The unreported components added over 74,000 t of unreported catches, of which 38,600 t were artisanal landings, 16,000 t industrial landings, over 4,000 t recreational catches, 3,000 t subsistence catches and around 12,000 t were discards. -

Box 1346 Ann Arbor, Ml 48106-1346 Abstract The cytochromes P450 (CYPs) are membrane bound proteins that collectively carry out a wide range of biological oxidations important in the metabolism of steroids, ecosanoids and xenobiotics including anticancer agents. A method of CYP separation and analysis has been developed using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and nano-scale liquid chromatography-electrospray-tandem mass spectrometry (LC-ESI-MS/MS) for the direct identification of multiple CYPs found in rat liver, human tissues and tumours. Endoplasmic reticulum from selected tissues was prepared as microsomes using ultracentrifugation and subjected to SDS-PAGE. -

These specimens were sampled monthly during landings of coastal fi sheries. Among the whole samples, 85 individuals were not sexually identifi ed. The sexed specimens were composed of 260 males (46.84 %) and 295 (53.15 %). The monthly sex ratio showed signifi cant differences between males and females. Females outnumbered males for sizes between 130 and 199 mm, while males outnumbered females from the 200 mm size up. Sexes combined, the length-weight relationship had a negative allometry. The reproduction period of the common pandora starts in April and ends in August. The gonado- somatic index (GSI) reaches the highest values in June for males and in July for females. -

CYP enzymes catalyze oxidative transformation leading to activation or inactivation of many endogenous and exogenous chemicals, with consequences for normal physiology and disease processes. Many CYPs potentially have roles in developmental specification, and many chemicals that cause developmental abnormalities are substrates for CYPs. Here we identify and annotate the full suite of CYP genes in zebrafish, compare these to the human CYP gene complement, and determine the expression of CYP genes during normal development. Results: Zebrafish have a total of 94 CYP genes, distributed among 18 gene families found also in mammals. There are 32 genes in CYP families 5 to 51, most of which are direct orthologs of human CYPs that are involved in endogenous functions including synthesis or inactivation of regulatory molecules. The high degree of sequence similarity suggests conservation of enzyme activities for these CYPs, confirmed in reports for some steroidogenic enzymes (e.g. CYP19, aromatase; CYP11A, P450scc; CYP17, steroid 17a-hydroxylase), and the CYP26 retinoic acid hydroxylases. Complexity is much greater in gene families 1, 2, and 3, which include CYPs prominent in metabolism of drugs and pollutants, as well as of endogenous substrates. -

Most cases are caused by 21-hydroxylase deficiency (21OHD). The systemic consequences of imbalanced steroid hormone biosynthesis due to severe 21OHD remains poorly understood. Therefore, we developed a zebrafish model for 21OHD, which focuses on the impairment of glucocorticoid biosynthesis. A single 21-hydroxylase gene (cyp21a2) is annotated in the zebrafish genome based on sequence homology. Our in silico analysis of the 21-hydroxylase (Cyp21a2) protein sequence suggests a sufficient degree of similarity for the usage of zebrafish cyp21a2 to model aspects of human 21OHD in vivo. We determined the spatiotemporal expression patterns of cyp21a2 by whole-mount in situ hybridization and reverse transcription polymerase chain reaction throughout early development. -

In current study, two hypotheses of Tamoxifen consumption in breast cancer cell line (MCF7) were investigated. First, the effect of Tamoxifen on genes expression prole at transcriptome level was evaluated between the control and treated samples. Second, due to the fact that Tamoxifen is known as a mutagenic factor, there may be an association between the alterations of genetic variants and Tamoxifen treatment, which can impact on the drug response. Methods In current study, the whole-transcriptome (RNA-seq) dataset of four investigations (19 samples) were derived from European Bioinformatics Institute (EBI). At transcriptome level, the effect of Tamoxifen was investigated on gene expression prole between control and treatment samples.
