The Isolation, Genetic Characterisation And
The isolation, genetic characterisation and biological activity of a South African Phthorimaea operculella granulovirus (PhopGV-SA) for the control of the Potato Tuber Moth, Phthorimaea operculella (Zeller) A thesis submitted in fulfilment of the requirements for the degree of MASTER OF SCIENCE At RHODES UNIVERSITY By MICHAEL DAVID JUKES February 2015 i Abstract The potato tuber moth, Phthorimaea operculella (Zeller), is a major pest of potato crops worldwide causing significant damage to both field and stored tubers. The current control method in South Africa involves chemical insecticides, however, there is growing concern on the health and environmental risks of their use. The development of novel biopesticide based control methods may offer a potential solution for the future of insecticides. In this study a baculovirus was successfully isolated from a laboratory population of P. operculella. Transmission electron micrographs revealed granulovirus-like particles. DNA was extracted from recovered occlusion bodies and used for the PCR amplification of the lef-8, lef- 9, granulin and egt genes. Sequence data was obtained and submitted to BLAST identifying the virus as a South African isolate of Phthorimaea operculella granulovirus (PhopGV-SA). Phylogenetic analysis of the lef-8, lef-9 and granulin amino acid sequences grouped the South African isolate with PhopGV-1346. Comparison of egt sequence data identified PhopGV-SA as a type II egt gene. A phylogenetic analysis of egt amino acid sequences grouped all type II genes, including PhopGV-SA, into a separate clade from types I, III, IV and V. These findings suggest that type II may represent the prototype structure for this gene with the evolution of types I, III and IV a result of large internal deletion events and subsequent divergence. -

Miles de familias, en las zonas andinas, dependen directamente de esta actividad. Por esta razón, este cultivo es una importante fuente de ingreso para las comunida des rurales y la economía nacional. Diferentes plagas y enfermedades afectan el cultivo de la papa en el Ecuador, causando pérdidas económicas y consecuentes problemas sociales en decenas de miles de familias (Fig. 1). El complejo de las polillas de la papa (Lepidoptera: Gelechiidae) es uno de los grupos que causan mayores daños tanto en campo como en almacenamiento. Las principales especies de polillas de la papa son: Phthorimaea operculella (Zeller), Tecia solanivora (Povolny) y Symmetrischema plaesiosema (Turner). Fotografías : Álvaro R. Dichotomous Key to Gelechiid Larvae
KEY TO SEPARATE NATIVE AND QUARANTINE SIGNIFICANT SPECIES OF GELECHIIDAE FEEDING ON SOLANACEAE IN FLORIDA AND THE GULF COAST REGION Steven Passoa and Jim Young Passoa (2007) illustrated identification characters of some exotic Lepidoptera of quarantine significance to the southern United States. As pointed out by Hodges (1998), little information exists on the immature stages of the Gelechioidea and very few vouchers document these life cycles. This lack of material prevented Passoa (2007) from treating the family Gelechiidae at the species level. Here we present a key to separate some native and quarantine significant species of Gelechiidae feeding on Solanaceae in Florida and the Gulf Coast Region. Identification of the pest species is often difficult; in particular, there is a need to distinguish Tuta absoluta from Keiferia lycopersicella (Povolny 1975: 392). Larval terminology follows Stehr (1987). The definition of "outer tooth" (called an extra tooth by Benander 1937) follows Passoa (1985). This key is made for late instar larvae; early instars may not be identifiable. Characters used in the key were taken from Capps (1946), Keifer (1936, 1937), Povolny (1973), Schnitzler et al. (2012), USDA (1973) and Weisman (1986). A few discrepancies in the literature are discussed in the caveat section below. The characters in the couplets are ordered from the head to abdomen for ease of viewing. The number of teeth on the mandible is subject to wear, and the last (fifth) tooth is especially small and easily missed if worn. Typically, gelechiid larvae are recognized by having the following combination of characters: a trisetose prespiracular group, L1 and L2 closely spaced below the spiracle on A3-6, a bisetose SV group on A1, a trisetose SV group on A3-6, and A9 with the D and SD setae each on a separate pinaculum (Capps 1946). -

Hoy: Myths of Managing Resistance 385 INTRODUCTION TO THE SYMPOSIUM, THE MYTHS OF MANAGING RESISTANCE MARJORIE A. HOY Entomology & Nematology Department, University of Florida, Gainesville, FL 32611-0620 Resistance to pesticides in arthropod pests is a serious and increasingly perplexing problem in Florida, the USA, and the world. Resistance to pesticides already has cre- ated significant economic, ecological, and public health problems in agricultural, household and garden, and medical/veterinary pest management programs. Exten- sive research has been conducted on diverse aspects of pesticide resistance, and we have learned much during the past 40 years. However, to some degree, much of the discussion about ‘resistance management’ has been based on ‘myths’. As an organizer of this symposium, one of my goals was to stress that managing resistance is a formi- dable task that will remain a perpetual pest management dilemma, because resis- tance is a fundamental survival response to stress by arthropods. Five papers were presented in this symposium at the 1994 annual meeting of the Florida Entomological Society, but one manuscript regarding the response by indus- try to resistance could not be published in this series. In the first paper, Gary Leibee and John Capinera assess the impact of resistance to pesticides in Florida and cite examples of resistance that limit pest management options. Julie Scott describes what we currently know about the molecular genetics of ar- thropod resistance to pesticides. The number of genes identified, and the diversity of their effects on the physiology of arthropods, verify that resistance is a normal re- sponse to diverse environmental stresses. -

LEPIDOPTERA (MOTHS AND BUTTERFLIES) AT INVERNESS RIDGE IN CENTRAL COASTAL CALIFORNIA AND THEIR RECOVERY FOLLOWING A WILDFIRE J. A. Powell Essig Museum of Entomology, University of California, Berkeley, CA 94720 Abstract.— In numbers of species, Lepidoptera (butterflies and moths) make up the largest group of plant-feeding animals in North America. Caterpillars of nearly all species feed on plants, and most of them are specialists on one or a few kinds of plants. Therefore they are liable to be severely affected by wildfires, and secondarily, their parasites and predators, including birds, bats, lizards, and rodents, suffer losses of a major food resource. In October 1995, a wildfire swept over part of The Point Reyes National Seashore, burning more than 12,300 acres (5,000 hectares) of public and private land, following a fire-free period of several decades. I tracked survival and recolonization by moths and butterflies during the subsequent five seasons. I made daytime searches for adults and caterpillars approximately monthly from March through October and collected blacklight trap samples, mostly in May and September-October. More than 650 species of Lepidoptera have been recorded in the Inverness Ridge area, and about 375 of them were recorded during the post-fire survey, including larvae of 31% of them. Plants in a Bishop pine forest higher on the ridge, where the fire was most intense, accumulated their caterpillar faunas slowly, while Lepidoptera feeding on plants typical of riparian woods in the lower canyons reestablished sooner and more completely. Recolonization varied markedly among different plant species, and the species richness gradually increased, in marked contrast to generalizations about effects of fire on arthropods derived from fire management of grasslands.
