This plant has been used in traditional medicine in different cultures as a hallucinatory, analgesic, aphrodisiac, nematicide, sleep inducer, and muscle relaxant, as well as a treatment for rheumatism, asthma, and inflammation. The flowers, fruits, stems, and roots of the plant are used, and different chemical compounds have been identified, such as alkaloids, volatile compounds (mainly terpenes), coumarins, flavonoids, steroids, and hydrocarbons. The concentration of the different compounds varies according to the biotic and abiotic factors to which the plant is exposed.

D. discolor remains nested within the Section Dutra, refuting earlier suggestions as intermediary or ancestral role. But the divergence of D. discolor from the remaining taxa within the section Dutra as a clearly supported subclade (BS 100) may be suggestive of the subdivision of Section Dutra into two subsections. This subdivision of section Dutra however remains to be appraised using cladistical studies incorporating both molecular and morphological data.

It has been recorded as a new alien species to the vascular flora ofBosnia and Herzegovina. In Bosnia and Herzegovina, D. innoxia is reported from two localities in the city of Mostar where the species has escaped cultivation and established small populations in surrounding ruderal habitats including waste land. The species is toxic to animals and humans. The paper presents a short morphological characteristic and photographs as well as the distribution of the species in the Bosnia and Herzegovina. The four genera comprising the family Goetzeaceae and the monotypic families Duckeodendraceae, Nolanaceae, and Sclerophylaceae, often recognized in traditional classifications, are shown to be included in Solanaceae. The current results corroborate previous studies that identify a monophyletic subfamily Solanoideae and the more inclusive “x = 12” clade, which includes Nicotiana and the Australian tribe Anthocercideae. These results also provide greater resolution among lineages within Solanoideae, confirming Jaltomata as sister to Solanum and identifying a clade comprised primarily of tribes Capsiceae (Capsicum and Lycianthes) and Physaleae.

    Poczai and Hyvönen SpringerPlus 2013, 2:459 http://www.springerplus.com/content/2/1/459 a SpringerOpen Journal SHORT REPORT Open Access Discovery of novel plastid phenylalanine (trnF) pseudogenes defines a distinctive clade in Solanaceae Péter Poczai* and Jaakko Hyvönen Abstract Background: The plastome of embryophytes is known for its high degree of conservation in size, structure, gene content and linear order of genes. The duplication of entire tRNA genes or their arrangement in a tandem array composed by multiple pseudogene copies is extremely rare in the plastome. Pseudogene repeats of the trnF gene have rarely been described from the chloroplast genome of angiosperms. Findings: We report the discovery of duplicated copies of the original phenylalanine (trnFGAA) gene in Solanaceae that are specific to a larger clade within the Solanoideae subfamily. The pseudogene copies are composed of several highly structured motifs that are partial residues or entire parts of the anticodon, T- and D-domains of the original trnF gene. Conclusions: The Pseudosolanoid clade consists of 29 genera and includes many economically important plants such as potato, tomato, eggplant and pepper. AMER Botany and Microbiology Department, Faculty of Science, Cairo University, Giza 12613, Egypt *Author for correspondence: Email: [email protected] / [email protected] Abstract Datura stramonium (Solanaceae) is an annual weed found in most temperate and subtropical regions of the world. The taxonomic identity of this species is still under debate since Linnaeus first published the name. Early on, many varieties and forms were recognized. In Egypt, D. stramonium presents in two forms: the white-flowered ‘stramonium’ form and violet- flowered ‘tatula’ form. Some authors treated D. stramonium and D. tatula as two distinct species, while others included D. tatula within D. stramonium as either a variety or forma. The present study aimed to elucidate the taxonomic identity of both the white ‘stramonium’ and the violet ‘tatula’ forms based on morphological, palynological and cytogenetic studies extended to karyotyping. A taxonomic study of these forms was carried out using 75 morphological and pollen characters. Significant morphological differences were observed; the most important ones were flower and stem colour, in addition to flower and fruit dimensions. The anatomical examination of juvenile-fruit in acropetal transverse sections, revealed the gradual displacement of parietal placentation at the fruit base to axile at the fruit apex. Pollen of both forms showed no significant differences.

Recently, we published a tissue-culture based transformation protocol for D. stramonium that enables more thorough functional genomics studies of this plant. However, the tissue culture process can lead to undesirable phenotypic and genomic consequences independent of the transgene used. Here, we have assembled and annotated a draft genome of D. stramonium with a focus on tropane alkaloid biosynthetic genes. We then use mRNA sequencing and genome resequencing of transformants to characterize changes following tissue culture. Results: Our draft assembly conforms to the expected 2 gigabasepair haploid genome size of this plant and achieved a BUSCO score of 94.7% complete, single-copy genes. The repetitive content of the genome is 61%, with Gypsy-type retrotransposons accounting for half of this.

Abstract: The genus Datura (Solanaceae) contains nine species of medicinal plants that have held both curative utility and cultural significance throughout history. This genus’ particular bioactivity results from the enormous diversity of alkaloids it contains, making it a valuable study organism for many disciplines. Although Datura contains mostly tropane alkaloids (such as hyoscyamine and scopolamine), indole, beta-carboline, and pyrrolidine alkaloids have also been identified. The tools available to explore specialized metabolism in plants have undergone remarkable advances over the past couple of decades and provide renewed opportunities for discoveries of new compounds and the genetic basis for their biosynthesis. This review provides a comprehensive overview of studies on the alkaloids of Datura that focuses on three questions: How do we find and identify alkaloids? Where do alkaloids come from? What factors affect their presence and abundance? We also address pitfalls and relevant questions applicable to natural products and metabolomics researchers. With both careful perspectives and new advances in instrumentation, the pace of alkaloid discovery—from not just Datura—has the potential to accelerate dramatically in the near future.
