  • Yield Response of Yam (Dioscorea Rotundata Poir.) to Inoculation with Azotobacter and Nitrogen Chemical Fertilization in the Caribbean Region of Colombia
RIA Vol. 47 n.º 1 Abril 2021, Argentina Received March 28th 2019// Accepted July 29th 2019 // Published online July 14th 2021 Yield response of yam (Dioscorea rotundata Poir.) to inoculation with Azotobacter and nitrogen chemical fertilization in the Caribbean region of Colombia SÁNCHEZ, D.B.1; LUNA, L.L.1; ESPITIA, A.A.1; CADENA, J.1 ABSTRACT In previous studies, we collected roots and soils associated with yam crops (Dioscorea rotundata Poir.) along the Caribbean region of Colombia from which several strains of Azotobacter chroococcum and Azotobacter vinelandii were identified, which in laboratory and nursery studies showed growth promotion activity in yam.In this research, we obtained from Agrosavia two of these strains (A. chroococcum DBC12, A. vinelandii DBC9) and evaluated their effect on yam yields and tuber quality under field conditions, in combination with four ni- trogen levels (0, 50, 75 and 100% of the recommended N fertilization dose). In a first instance, in vitro tests confirmed the N fixation capacity and the 3NH production of both bacterial strains, while under field conditions, the highest yields were obtained from treatments that combined the individual inoculation with strains A. ch- roococcum DBC12 or A. vinelandii DBC9, with 50% of the recommended N fertilization level. On the other hand, yam tubers were classified according to market quality, and the same treatments induced the production of higher yields of first category or export-type tubers, suggesting that these two bacterial strains were also able to improve tuber quality. From this study it was concluded that A. chroococcum DBC12, A. vinelandii DBC9 strains have the potential for replacing up to 50% of the recommended N fertilization dose and present potential as possible bio-inoculants, which could be an alternative for reducing levels of chemical nitrogen fertilization, thus contributing to a more sustainable and competitive yam culture.

Patel1 and S. Bhatt2 1Department of Biotechnology, Mehsana Urban Institute of Sciences, Mehsana-Gozariya Highway, Ganpat University-384012, Gujarat, India, 1Department of Life Sciences, Hemchandracharya North Gujarat University, Patan-384265, Gujarat, India *Author for Correspondence ABSTRACT The North Gujarat region is geographically having varied terrestrial ecosystem which is comprised of semiarid to arid and highly arid saline soil types. It also has tropical dry deciduous Forest patches. Thus, considering these ecoclimatic conditions, it is thought to have different diazotrophic populations which might be explored for potential biofertilizer to improve crop productivity in the region. In this context, the long-lasting challenges in the development of effective methods to know the types of microorganisms present in soils, and to determine functions which the microbes perform in situ. Total 68 free living nitrogen fixing bacteria having ability to grow on N2 free media were categorized in different group and identified at species level by ARDRA analysis and 16S r-RNA sequences. One isolate is closely related to uncultured bacteria based on complete sequence analysis. Our result indicated that higher diversity of free-living nitrogen-fixers was mainly affected by the physico-chemical properties of the soils and plant species, especially the changes of nutrient. Keywords: Free Living Nitrogen Fixing Bacteria, 16S r-RNA Sequencing, Diversity INTRODUCTION Symbiotic systems represent the most significant biological N-source for many terrestrial ecosystems (Kennedy and Islam, 2001).

Prof. Dr. Eckhard George 2. Dr. Silke Ruppel Tag der mündlichen Prüfung: 25.11.2009 ii Abstract Plant growth promoting bacteria (PGPB) are ubiquitous in both plant root and shoot, and are important contributors to the nitrogen-input of plants exerting their positive effects on plant growth directly or indirectly through different mechanisms. The present work focuses on a) the isolation of PGPB, which promotes the growth of different plant cultures and controls plant diseases caused by Fusarium species, b) the prospects of PGPB to solve plant nutritional problems, c) developing new molecular methods for the assessment of their diversity and activity. A total of 780 bacterial strains were isolated from root, rhizosphere and phyllosphere of wheat grown in soil Syrdarya and were tested for their ability to promote the growth of other plants resulting in several universal PGPB strains. Contributions of PGPB to plant nutrition were investigated with wheat, and several vegetable plants such as cucumber, tomato, paprika, and cauliflower on quartz-sand substrates. Bacterial colonization, plant dry weight, and N concentrations in plants were measured. Bacterial inoculation effects on plant N nutrition in cucumber and tomato plants were determined by exposing bacterial and non bacterial plants to two, low and high NH4+NO3- supply.
