Penicillin's Discovery and Antibiotic Resistance

Mariya Lobanovska* and Giulia Pilla* Sir William Dunn School of Pathology, University of Oxford, United Kingdom Undoubtedly, the discovery of penicillin is one of the greatest milestones in modern medicine. 2016 marks the 75th anniversary of the first systemic administration of penicillin in humans, and is therefore an occasion to reflect upon the extraordinary impact that penicillin has had on the lives of millions of people since. This perspective presents a historical account of the discovery of the wonder drug, describes the biological nature of penicillin, and considers lessons that can be learned from the golden era of antibiotic research, which took place between the 1940s and 1960s. Looking back at the history of penicillin might help us to relive this journey to find new treatments and antimicrobial agents.This is particularly relevant today as the emergence of multiple drug resistant bacteria poses a global threat, and joint efforts are needed to combat the rise and spread of resistance. This is Peggy Dillon of History Associates Incorporated. I am speaking with Dr. Joseph Burchenal for the NCI oral history interview series. We are at his home in Darien, Connecticut. Today is January 26, 2001. JB: Good morning. PD: I would like to talk with you today about your work in cancer chemotherapy in general and also your involvement with the National Cancer Institute in particular. Before we get into your career overall and NCI, I'd like to ask you about your background. If you could just tell me a little bit about your upbringing and education . I understand that you were interested in chemistry from a very early age. JB: I was born in Milford, Delaware, but I lived in Wilmington, Delaware, for the first part of my life. My mother died when I was three, in childbirth. So I lived with my father in Wilmington. He was a lawyer. I went to school at Tower Hill School. The majority of my friends in the school had fathers or mothers–fathers mainly–who worked for Dupont. They were very likely to, so that there was a lot of interest in chemistry there. In fact, it was the same way on the street where we lived, where the fathers were mostly chemists. But my interest in chemistry, I guess, started when I was eight years old and I had a cousin who was eleven who was given a Chemcraft set and he used to show us all of the interesting things that he could do with it. -

CASTLE EORGE MINOT was born in Boston, Massachusetts, on Decem- G ber 2, 1885, the eldest of three sons of Dr. James Jackson and Elizabeth Frances (Whitney) Minot. His ancestors had been successful in business and professional careers in Boston. His father was a private practitioner and for many years a clinical teacher of medicine as a member of the staff of the Massachusetts General Hospital. In the second half of the nineteenth century his great uncle, Francis Minot, became the third Hersey Pro- fessor of the Theory and Practice of Physic at Harvard; and his cousin, Charles Sedgwick Minot, a distinguished anatomist, was Professor of Histology there in the early years of the twentieth century. George Minot's grandmother was the daughter of Dr. James Jackson, the second Hersey Professor and a cofounder with John Collins Warren of the Massachusetts General Hos- pital, which opened its doors in 1821. Thus his forebears, like those of other Boston medical families, were influential par- ticipants in the activities of the Harvard Medical School and its affiliated teaching hospital. George was regarded by his physician-father as a delicate child who required physical protection and nourishing food. -

FREDERIC MIDDLEBROOK RICHARDS August 19, 1925–January 11, 2009 Elected to the NAS, 1971 Frederic M. Richards (who preferred to be called Fred) is remembered as a guiding influence in 20th century protein chemistry and structural biology. In a landmark 1957 exper- iment, he discovered that the proteolytic enzyme subtilisin cuts the protein ribonuclease into two inactive fragments which, when mixed, associate spontaneously and restore activity completely. This single experiment at once estab- lished the field of protein molecular recognition, showing, for example, how peptide hormones might activate their protein receptors. Later, Fred pioneered computational approaches for analyzing and interpreting protein struc- tures that remain in widespread use today. He was the founding chair of the Department of Molecular Biophysics and Biochemistry at Yale, which became the world’s By Robert L. Baldwin and George D. Rose preeminent department of molecular biophysics soon after its inception. He was an outspoken critic against the social forces that erode time and trivialize achievement. He served the scientific community at large in many influential roles, and his work garnered many of the most prestigious awards. Fred was a natural leader with a charismatic, fun-loving personality. He cherished life and reveled in ideas, conversation, the sheer joy of doing, and, especially, sailing. For Fred, a quintessential part of the full life was to spend at least 4 to 6 weeks a year sailing. -

Lichtman, M.D.* Department of Medicine and the James P. Wilmot Cancer Institute, University of Rochester Medical Center, Rochester, NY, USA ABSTRACT Alfred Nobel was one of the most successful chemists, inventors, entrepreneurs, and businessmen of the late nineteenth century. In a decision later in life, he rewrote his will to leave virtually all his fortune to establish prizes for persons of any nationality who made the most compelling achievement for the benefit of mankind in the fields of chemistry, physics, physiology or medicine, literature, and peace among nations. The prizes were first awarded in 1901, five years after his death. In considering his choice of prizes, it may be pertinent that he used the principles of chemistry and physics in his inventions and he had a lifelong devotion to science, he suffered and died from severe coronary and cerebral atherosclerosis, and he was a bibliophile, an author, and mingled with the literati of Paris. His interest in harmony among nations may have derived from the effects of the applications of his inventions in warfare (“merchant of death”) and his friendship with a leader in the movement to bring peace to nations of Europe. After some controversy, including Nobel’s citizenship, the mechanisms to choose the laureates and make four of the awards were developed by a foundation established in Stockholm; the choice of the laureate for promoting harmony among nations was assigned to the Norwegian Storting, another controversy. -
