The 13th meet- ing on Bacterial Genetics and Ecology (BAGECO) intends to provide a forum for discussing all the aspects related to such microbial continuities along changing environments including large and small-scale gradients, chemoclines and environmental transitions. Biotic gradients, like those in the animal gut, and abiotic ones, where physicochemical variations occur, as well as the combina- tion of the two in relation to any complex ecosystems will be central focuses of the conference. We are pleased to welcome some of the most renowned scientists in microbial ecology to discuss all these aspects in relation to the most complex environments of earth including, among others, soil, sediments, the symbiotic ecosystems associated to plant, animals and humans, the aquatic ecosystems and the role of microbes and their metabolism in the protection of the environ- ment. The conference debate will be also discussing the advancement on our understanding of the role of gene flow and on how passing from empirical approaches to dissect microbial community functions to establishing the theories that rule out the microbial world and finally the functioning of earth. The hierarchical taxonomic system of the phylum Proteobacteria began in 1809 when the genus Polyangium was first reported and has been generally adopted from 2001 based on the road map of Bergey’s Manual of Systematic Bacteriology. Until February 2018, the phylum Proteobacteria consisted of eight classes, 44 orders, 120 families, and more than 1,000 genera. Proteobacteria species isolated from various environments in Korea have been reported since 1999, and 644 species have been approved as of February 2018. In this study, all novel Proteobacteria species from Korean environments were affiliated with four classes, 25 orders, 65 families, and 261 genera. A total of 304 species belonged to the class Alphaproteobacteria, 257 species to the class Gammaproteobacteria, 82 species to the class Betaproteobacteria, and one species to the class Epsilonproteobacteria. The predominant orders were Rhodobacterales, Sphingomonadales, Burkholderiales, Lysobacterales and Alteromonadales. The most diverse and greatest number of novel Proteobacteria species were isolated from marine environments. Proteobacteria species were isolated from the whole territory of Korea, with especially large numbers from the regions of Chungnam/Daejeon, Gyeonggi/Seoul/Incheon, and Jeonnam/Gwangju.

    ABSTRACT TRACING HYDROCARBON CONTAMINATION THROUGH HYPERALKALINE ENVIRONMENTS IN THE CALUMET REGION OF SOUTHEASTERN CHICAGO Kathryn Quesnell, MS Department of Geology and Environmental Geosciences Northern Illinois University, 2016 Melissa Lenczewski, Director The Calumet region of Southeastern Chicago was once known for industrialization, which left pollution as its legacy. Disposal of slag and other industrial wastes occurred in nearby wetlands in attempt to create areas suitable for future development. The waste creates an unpredictable, heterogeneous geology and a unique hyperalkaline environment. Upgradient to the field site is a former coking facility, where coke, creosote, and coal weather openly on the ground. Hydrocarbons weather into characteristic polycyclic aromatic hydrocarbons (PAHs), which can be used to create a fingerprint and correlate them to their original parent compound. This investigation identified PAHs present in the nearby surface and groundwaters through use of gas chromatography/mass spectrometry (GC/MS), as well as investigated the relationship between the alkaline environment and the organic contamination. PAH ratio analysis suggests that the organic contamination is not mobile in the groundwater, and instead originated from the air. 16S rDNA profiling suggests that some microbial communities are influenced more by pH, and some are influenced more by the hydrocarbon pollution. BIOLOG Ecoplates revealed that most communities have the ability to metabolize ring structures similar to the shape of PAHs. Analysis with bioinformatics using PICRUSt demonstrates that each community has microbes thought to be capable of hydrocarbon utilization. The field site, as well as nearby areas, are targets for habitat remediation and recreational development. In order for these remediation efforts to be successful, it is vital to understand the geochemistry, weathering, microbiology, and distribution of known contaminants.

Cells of strain CJ661T were white-coloured, Gram-staining-negative, non-motile, aerobic and rod-shaped. Strain CJ661T grew optimally at 30 C and pH 7.0. The analysis of 16S rRNA gene sequence of strain CJ661T showed that it belongs to the genus Ramlibacter within the family Comamonadaceae and was most closely related to Ramlibacter ginsenosidimutans KCTC 22276T (98.1 %), followed by Ramlibacter henchirensis DSM 14656T (97.1 %). DNA–DNA relatedness levels of strain CJ661T were 40.6 % to R. ginsenosidimutans KCTC 22276T and 25.0 % to R. henchirensis DSM 14656T. The major isoprenoid quinone was ubiquinone (Q-8). The predominant polar lipids were phosphatidylethanolamine, diphosphatidylglycerol and phosphatidylglycerol. The T major cellular fatty acids of strain CJ661 were summed feature 3 (C16 : 1 !6c and/or C16 : 1 !7c), C16 : 0 and summed feature 8 (C18 : 1 !7c and/or C18 : 1 !6c). The G+C content of the genomic DNA was 65.4 mol%. On the basis polyphasic taxonomic data, strain CJ661T represents a novel species in the genus Ramlibacter, for which name Ramlibacter alkalitolerans sp. nov. is proposed; the type strain is CJ661T (=KACC 19305T=JCM 32081T). The genus Ramlibacter was introduced by Heulin et al. [1], (Qiagen). The 16S rRNA gene sequence was determined at and belongs to the family Comamonadaceae in the class Solgent (Daejeon, Korea) using the BigDye Terminator Cycle Betaproteobacteria.

However, compositional and functional responses of bacterial endophyte communities towards agricultural practices are still poorly understood. Hence, we analyzed the influence of fertilizer application and mowing frequency on bacterial endophytes in three agriculturally important grass species. For this purpose, we examined bacterial endophytic communities in aerial plant parts of Dactylis glomerata L., Festuca rubra L., and Lolium perenne L. by pyrotag sequencing of bacterial 16S rRNA genes over two consecutive years. Although management regimes influenced endophyte communities, observed responses were grass species-specific. This might be attributed to several bacteria specifically associated with a single grass species. We further predicted functional profiles from obtained 16S rRNA data. These profiles revealed that predicted abundances of genes involved in plant growth promotion or nitrogen metabolism differed between grass species and between management regimes. Moreover, structural and functional community patterns showed no correlation to each other indicating that plant species-specific selection of endophytes is driven by functional rather than phylogenetic traits. The unique combination of 16S rRNA data and functional profiles provided a holistic picture of compositional and functional responses of bacterial endophytes in agricultural relevant grass species towards management practices. Endophytic bacteria comprising various genera have been detected in a wide range of plant species1. Beneficial endophytic bacteria can promote plant growth and/or resistance to diseases and environmental stresses by a variety of mechanisms.

    _________________________________________ Microbiology of Serpentine Hot Springs, Alaska MSU-233 J979111K013 Dr. Timothy McDermott and Dana Skorupa Montana State University, Bozeman MT August 2011 1 Results & Discussion Water quality assessment: Coliform counts Water samples were examined with the aim being to assess microbial diversity and water quality as potentially impacted by human activity associated with the bathhouse and bunkhouse structures. Coliform counts and microbial phylogenetic diversity of Serpentine Creek waters upstream, downstream, and immediately surrounding and adjacent to the structures were examined. Though quite variable, total coliform (TC) counts were elevated in the four sampling sites most closely adjacent to the bath house and bunk house (structures) (Fig. 7, sites 38, 39, 40, 43), averaging 175± 160 . 100 ml-1 for all four sites. TC counts averaged 30 ± 10 . 100 ml-1 in the creek waters upstream of the structures, which are not likely impacted by human-associated activity (Fig. 7, sites 7 and 42) (Table 9). TC levels at site 8 were of interest because, while it is located up-drainage from the structures, it is part of a drainage system downstream of a very large beaver dam complex and thus could contribute background TC counts from indigenous mammals that visit or inhabit the area. Site 8 TC counts were similar to sites 7 and 42, and thus did not appear to be a significant source of TCs. Fecal coliform (FC) counts were relatively low at all sites and again variable. FCs at sites 38, 39, 40, 43 surrounding the structures averaged 3.8 ± 4.3 . 100 ml-1 (Table 9) as compared to 2.0 ± 1.4 .

Changes in maize genetics (G) and environmental (E) conditions increased productivity under high-synthetic- input conditions. However, novel selective pressures on the rhizobiome may have incurred undesirable trade- offs in organic agroecosystems, where plants obtain nutrients via microbially mediated processes including mineralization of organic matter. Using twelve maize genotypes representing an evolutionary transect (teosintes, landraces, inbred parents of modern elite germplasm, and modern hybrids) and two agricultural soils with contrasting long- term management, we integrated analyses of rhizobiome community structure, potential microbe-microbe interactions, and N-cycling functional genes to better understand the impacts of maize evo- lution and soil management legacy on rhizobiome recruitment. We show complex shifts in rhizobiome communities during directed evolution of maize (defined as the transition from teosinte to modern hybrids), with a larger effect of domestication (teosinte to landraces) than modern breeding (inbreds to hybrids) on rhizobiome structure and greater impacts of modern breeding on po- tential microbe- microbe interactions.
