Introduction George Palade shared the 1974 Nobel Prize with Albert Claude and Christian de Duve for their pioneering work in the characterization of organelles interrelated by the process of secretion in mammalian cells and tissues. These three scholars established the modern field of cell biology and the tools of cell fractionation and thin section transmission electron microscopy. It was Palade’s genius in particular that revealed the organization of the secretory pathway. He discovered the ribosome and showed that it was poised on the surface of the endoplasmic reticulum (ER) where it engaged in the vectorial translocation of newly synthesized secretory polypeptides (1). And in a most elegant and technically challenging investigation, his group employed radioactive amino acids in a pulse-chase regimen to show by autoradiograpic exposure of thin sections on a photographic emulsion that secretory proteins progress in sequence from the ER through the Golgi apparatus into secretory granules, which then discharge their cargo by membrane fusion at the cell surface (1). He documented the role of vesicles as carriers of cargo between compartments and he formulated the hypothesis that membranes template their own production rather than form by a process of de novo biogenesis (1). As a university student I was ignorant of the important developments in cell biology; however, I learned of Palade’s work during my first year of graduate school in the Stanford biochemistry department. -

Cover image: The Starry Night, Vincent van Gogh Abstract Today’s leading projections of climate change predicate on atmospheric General Circulation Models (GCMs). Since the atmosphere consists of a staggering range of scales that impact global trends, but computational constraints prevent many of these scales from being directly represented in numerical simulations, GCMs require “parameterisations” - models for the influence of unresolved processes on the resolved scales. State- of-the-art parameterisations are commonly based on combinations of phenomenological arguments and physics, and are of considerably lower fidelity than the resolved simulation. In particular, the parameter- isation of low-altitude stratocumulus clouds that result from small-scale processes in sub-tropical marine boundary layers is widely considered the largest source of uncertainty that remains in contemporary GCMs’ prediction of the temperature response to a global increase in CO2. Improvements in the capacity of machine learning algorithms and the increasing availability of high- fidelity datasets from global satellite data and local Large Eddy Simulations (LES) have identified data-driven parameterisations as a high-potential option to break the deadlock. -

The purpose of the reception is to honor and recognize outgoing AGU president Marcia McNutt and her new role as President of the National Academy of Sciences (NAS). Approximately 70 guests have been invited to the reception from academia, Federal agencies, AGU, NAS, and colleagues of Dr. McNutt. The estimated cost per person of the reception is $75. Attendance at the reception will allow NASA employees an opportunity to discuss with other guests issues concerning NASA's programs and missions. Based on the above, I find that the event meets the requirements of a ''widely attended gathering" as defined in 5 CFR 2635.204(g). I further find that there is an Agency interest in having NASA employees attend this event. Accordingly, NASA employees whose duties do not substantially affect AGU or a majority of its members, including NASA employees who are in non-career positions for which they are required to sign an ethics pledge under Executive Order 13490, may accept an invitation for free attendance to the reception for themselves and a guest. However, NASA employees whose duties substantially affect AGU or a majority of its members, for instance by way of procurement duties, should seek an individual determination regarding participation in ~ event from tl,leir local ethics counselor. -

Peterson with research assistance from Paul White Version 1, June 2010 Safe laboratory practices are an important element in the responsible conduct of research. They protect scientists, research assistants, laboratory technicians, and others from hazards that can arise during experimentation. The particular hazards that might arise vary according to the type of experimentation: the explosions or toxic gases that could result from chemical experiments are prevented or contained by different kinds of measures then are used to limit the health hazards posed by working with radioactive isotopes. In most areas, scientists are allowed – and even encouraged – to decide for themselves what experiments to undertake and how to construct and run their labs. In 1970, there were three main exceptions, and they applied more to questions of how to carry out research than to selecting topics for research: use of radioactive materials, use of tumor-causing viruses, and use of human subjects. In these areas, levels of risk to persons inside and outside the lab were perceived as great enough to justify having the government, either through the regular bureaucracy or through scientific agencies, set and enforce mandatory safety standards. When genetics was poised at the threshold of recombinant DNA research and genetic manipulation techniques in the 1960s, the hazards such work might pose to lab personnel or to others were unknown. Many scientists and others believed they were likely to be significant. The primary concern in public discussion was whether hybrid organisms bred from recombinant DNA would cause new diseases or create varieties of plants or animals that would overwhelm and irretrievably change the natural environment. -

Emanuel | Climate Science and Climate Risk: A Primer 1 Considerably more than 90% percent of climate scientists attribute the bulk of the increase in global mean temperature over the past three to four decades to the anthropogenic increase in atmospheric greenhouse gases that commenced with the Industrial Revolution.1 The great majority of these scientists hold that continued warming presents significant risks to humankind over the coming centuries. What scientific evidence led the scientific community to these conclusions? How robust is that evidence? To what extent should we trust uncertain projections of future climate change based on complicated global climate models? How do we deal with climate change as a problem of risk assessment and management? his essay summarizes the most important lines of not fit within the existing scientific framework.
