Martens2* Abstract Parkinson’s disease (PD) is a devastating and highly prevalent neurodegenerative disease for which only symptomatic treatment is available. In order to develop a truly effective disease-modifying therapy, improvement of our current understanding of the molecular and cellular mechanisms underlying PD pathogenesis and progression is crucial. For this purpose, standardization of research protocols and disease models is necessary. As human dopaminergic neurons, the cells mainly affected in PD, are difficult to obtain and maintain as primary cells, current PD research is mostly performed with permanently established neuronal cell models, in particular the neuroblastoma SH-SY5Y lineage. This cell line is frequently chosen because of its human origin, catecholaminergic (though not strictly dopaminergic) neuronal properties, and ease of maintenance. However, there is no consensus on many fundamental aspects that are associated with its use, such as the effects of culture media composition and of variations in differentiation protocols. Here we present the outcome of a systematic review of scientific articles that have used SH-SY5Y cells to explore PD. We describe the cell source, culture conditions, differentiation protocols, methods/approaches used to mimic PD and the preclinical validation of the SH-SY5Y findings by employing alternative cellular and animal models. Thus, this overview may help to standardize the use of the SH-SY5Y cell line in PD research and serve as a future user’sguide.

    ICP-PR Honey Bee Protection Group 1980 - 2017 The ICP-PR Bee Protection Group held its rst meeting in Wageningen in 1980 and over the subsequent 38 years it has become the established expert forum for discussing the risks of pesticides to bees and developing solutions how to assess and manage this risk. In recent years, the Bee Protection Group has enlarged its scope of interest from honey bees to many other pollinating insects, such as wild bees including bumble bees. 462 The group organizes international scienti c symposia, usually once in every three years. These are open to Julius-Kühn-Archiv everyone interested. The group tries to involve as many countries as possible, by organizing symposia each time in another European country. A wide range of international experts active in this eld drawn from regulatory authorities, industry, universities and research institutes participate in the discussions. Histone H3 lysine 79 (H3K79) methylation at Myc-responsive elements of target gene promoters is a strict prerequisites for Myc-induced transcriptional activation. DOT1L is the only known histone methyltransferase that catalyses mono-methylation (me), di- methylation (me2) and tri-methylation (me3) at the histone H3K79 position, which have been linked to gene transcriptional activation. JMJD6 is a bi-functional arginine demethylase and lysyl-hydroxylase. The JMJD6 gene is located on the chromosome 17q25 position. 17q21-gter gain has been identified as the most frequent chromosome alternation in neuroblastoma and an indicator of poor patient prognostic. Here, I investigated the roles of DOT1L and JMJD6 in N-Myc over-expressing neuroblastoma. I found that N-Myc up-regulated DOT1L mRNA and protein expression, by binding to an E-box at the DOT1L gene promoter.
