Human infestation with the dwarf tapeworm has been reported from a wide variety of places in the United States but most of the evidence suggests that it is largely restricted to the southern part of the country. Even in the southern United States the incidence is low according to most of the data. Of the older surveys Greil (1915) re- ports 6 per cent dwarf tapeworm in 665 children examined in Alabama. The Rockefeller Sanitary Commission for the Eradication of Hook- worm Disease Reports for 1914 and 1915 record from 0 to 2.5 per cent dwarf tapeworm in eleven southern states (Alabama, Arkansas, Geor- gia, Kentucky, Louisiana, Mississippi, North and South Carolina, Ten- nessee, Texas, and Virginia) with an average of 1.8 per cent in the 141,247 persons examined. Wood (1912) tabulated the results of 62,786 routine fecal examinations made in the state laboratories of eight southern states (Arkansas, Florida, Georgia, Kentucky, Missis- sippi, North Carolina, Tennessee, and Virginia) and found an average of less than 1 per cent infection with this worm. Of the more recent reports on the frequency of the dwarf tapeworm in the general population, three are based on extensive surveys. Spindler (1929) records that 3.6 per cent of the 2,152 persons of all ages examined in southwest Virginia were positive; Keller, Leathers, and Bishop (1932) report 3.5, 2.9, and 1.5 per cent in eastern, central and western Tennessee; and Keller and Leathers (1934) report 0.4 per cent in Mississippi. -

Tort1* Abstract Background: Small non-coding RNAs, including miRNAs, and gene silencing mediated by RNA interference have been described in free-living and parasitic lineages of flatworms, but only few key factors of the small RNA pathways have been exhaustively investigated in a limited number of species. The availability of flatworm draft genomes and predicted proteomes allowed us to perform an extended survey of the genes involved in small non-coding RNA pathways in this phylum. Results: Overall, findings show that the small non-coding RNA pathways are conserved in all the analyzed flatworm linages; however notable peculiarities were identified. While Piwi genes are amplified in free-living worms they are completely absent in all parasitic species. Remarkably all flatworms share a specific Argonaute family (FL-Ago) that has been independently amplified in different lineages. Other key factors such as Dicer are also duplicated, with Dicer-2 showing structural differences between trematodes, cestodes and free-living flatworms. Similarly, a very divergent GW182 Argonaute interacting protein was identified in all flatworm linages. Contrasting to this, genes involved in the amplification of the RNAi interfering signal were detected only in the ancestral free living species Macrostomum lignano. We here described all the putative small RNA pathways present in both free living and parasitic flatworm lineages. Conclusion: These findings highlight innovations specifically evolved in platyhelminths presumably associated with novel mechanisms of gene expression regulation mediated by small RNA pathways that differ to what has been classically described in model organisms. -

Price Center For Parasite Repository and Education College of Public Health, University of South Florida PRESENTS Sources of Infective Stages and Modes of Transmission of Endoparasites Epidemiology is the branch of science that deals with the distribution and spread of disease. How diseases are transmitted, i.e. how they are passed from an infected individual to a susceptible one is a major consideration. Classifying and developing terminology for what takes place has been approached in a variety of ways usually related to specific disease entities such as viruses, bacteria, etc. The definitions that follow apply to those disease entities usually classified as endoparasites i.e. those parasites that reside in a body passage or tissue of the definitive host or in some cases the intermediate host. When the definition of terms for the “Source of Infection” or “Mode of Infection” relate to prevention and/or control of an endoparasitic disease, they should be clearly described. For the source of infection, the medium (water, soil, utensils, etc.) or the host organism (vector, or intermediate host) on which or in which the infective stage can be found should be precisely identified. For the mode of transmission, the precise circumstances and means by which the infective stage is able to come in contact with, enter, and initiate an infection in the host should be described. SOURCE OF INFECTION There are three quite distinct and importantly different kinds of sources of the infective stage of parasites: Contaminated Sources, Infested Sources, and Infected Sources. CONTAMINATE SOURCES Contaminated Source, in parasitology, implies something that has come in contact with raw feces and is thereby polluted with feces or organisms that were present in it. -

It has been described as one species, R. nana, found in both humans and rodents. Others have identified a subspecies; R. nana var. fraterna, describing it as morphologically identical to the human form but only found in rodents. The species present in Australian communities has never been identified with certainty. Fifty one human isolates of Rodentolepis (= Hymenolepis) nana were orally inoculated into Swiss Q, BALB/c, A/J, CBA/ CAH and nude (hypothymic) BALB/c mice, Fischer 344 and Wistar rats and specific pathogen free (SPF) hamsters. Twenty four human isolates of R. nana were cross-tested in flour beetles, Tribolium confusum. No adult worms were obtained from mice, rats or hamsters, even when immunosuppressed with cortisone acetate. Only one of the 24 samples developed to the cysticercoid stage in T. confusum; however, when inoculated into laboratory mice the cysticercoids failed to develop into adult worms. The large sample size used in this study, and the range of techniques employed for extraction and preparation of eggs provide a comprehensive test of the hypothesis that the human strain of R. -

We identified six helminth endoparasite taxa, being Phylum Acantocephala and Family Trichuridae the most prevalent. The high prevalence of the Family Ancylostomatidae indicates a possible transmission via domestic dogs, which are abundant in the study area. Nevertheless, our results indicate that the endoparasite species found are not different from those observed in protected or least disturbed areas, suggesting a high resilience of maned wolf and their parasites to human impacts, or a common scenario of disease transmission from domestic dogs to wild canid whether in protected or unprotected areas of southeastern Brazil. The goal of the present study was to identify the parasite in the specimens collected from NorthWestern regions of Iran using PCR-sequencing method. Methods: A total of 1521 stool samples were collected from the study individuals. Initially, the identification of hymenolepis nana was confirmed by parasitological method including direct wet-mount and formalin-ethyl acetate concentration methods. Afterward, PCR-sequencing analysis of ribosomal ITS2 fragment was targeted to investigate the molecular identification of the parasite. Results: Overall, 0.65% (10/1521) of the isolates were contaminated with H. nana in formalin-ethyl acetate concentration. All ten isolates were succefully amplified by PCR and further sequenced. The determined sequences were deposited in GenBank under the accession numbers MH337810 -MH337819. Conclusion: Our results clarified the presence ofH. nana among the patients in the study areas. In addition, the molecular tech- nique could be accessible when the human eggs are the only sources available to identify and diagnose the parasite. Keywords: Hymenolepis nana, rDNAITS2, PCR, Iran. -

Identification of Immunogenic Proteins of the Cysticercoid of Hymenolepis

Reports indicating the proteins in the cestode-host interactions are limited predominantly to taeniids, with no previous data available for non-taeniid species. A non-taeniid, Hymenolepis diminuta, represents one of the most important model species in cestode biology and exhibits an exceptional developmental plasticity in its life-cycle, which involves two phylogenetically distant hosts, arthropod and vertebrate. Results: We identified H. diminuta cysticercoid proteins that were recognized by sera of H. diminuta-infected rats using two-dimensional gel electrophoresis (2DE), 2D-immunoblotting, and LC-MS/MS mass spectrometry. Proteomic analysis of 42 antigenic spots revealed 70 proteins. The largest number belonged to structural proteins and to the heat-shock protein (HSP) family. These results show a number of the antigenic proteins of the cysticercoid stage, which were present already in the insect host prior to contact with the mammal host. These are the first parasite antigens that the mammal host encounters after the infection, therefore they may represent some of the molecules important in host-parasite interactions at the early stage of infection. Conclusions: These results could help in understanding how H. diminuta and other cestodes adapt to their diverse and complex parasitic life-cycles and show universal molecules used among diverse groups of cestodes to escape the host response to infection. -

Since praziquantel was first introduced as a broadspectrum anthelmintic in 1975, innumerable articles describ- ing its successful use in the treatment of the majority of human-infecting trematodes and cestodes have been published. The target trematode and cestode diseases include schistosomiasis, clonorchiasis and opisthorchiasis, paragonimiasis, het- erophyidiasis, echinostomiasis, fasciolopsiasis, neodiplostomiasis, gymnophalloidiasis, taeniases, diphyllobothriasis, hyme- nolepiasis, and cysticercosis. However, Fasciola hepatica and Fasciola gigantica infections are refractory to praziquantel, for which triclabendazole, an alternative drug, is necessary. In addition, larval cestode infections, particularly hydatid disease and sparganosis, are not successfully treated by praziquantel. The precise mechanism of action of praziquantel is still poorly understood. There are also emerging problems with praziquantel treatment, which include the appearance of drug resis- tance in the treatment of Schistosoma mansoni and possibly Schistosoma japonicum, along with allergic or hypersensitivity
