Differences in Sexual Dimorphism and Influences of Sexual Dichromatism on Crypsis Among Populations of the Jumping Spider Habronattus oregonensis by Jason Bazzano A thesis submitted in partial fulfillment of the requirements for the degree of Master of Science in Biology Thesis Committee: Susan Masta, Chair Suzanne Estes Michael Murphy Portland State University 2011 Abstract Crypsis can be an important mechanism of predator avoidance for organisms. However, many species exhibit sexual dichromatism, in which the males possess a suite of colorations in order to attract female attention. The resulting differences in crypsis between the males and females can provide insight into the relative strengths of the sexually and naturally selective forces shaping the coloration of the organism, as well as clues regarding potential sensory biases of the selecting sex. In this study, I examine variation in the coloration of four Pacific Northwest populations of the sexually dimorphic and dichromatic polygynous species of jumping spider Habronattus oregonensis and compare the coloration of different body regions of the spiders to their habitats. I also investigate differences in relative size of a male sexual ornament, the enlarged first leg tibia. Field work for this study was conducted in June and July of 2009. The three main foci of this study are 1) to compare the degree of color matching of females and their habitat to the degree of color matching of males and their habitat, evaluating whether sexual selection on males has reduced their degree of crypsis relative to that of females, 2) if there is indeed a difference in crypsis between the sexes, to gauge whether there are similar divergences from crypsis among the populations – both in the quantitative amount of divergences as well as the colorimetric direction of such divergences, and 3) whether there is any variation in sexual ornament size among populations. Male first leg tibia size is a sexual character that is presumably not influenced by habitat coloration; differences i in male tibia allometry among populations would provide supporting evidence for the hypothesis that sexual selection is indeed maintaining phenotypic differences among the populations, regardless of habitat location and color. I found a high degree of conformity of hue and chroma between male and female spiders and their habitats, with three notable exceptions. The most extreme difference in coloration between spider and habitat was that of the Gorge and Siskiyou population male anteriors. The anteriors had proportionally less green and more ultraviolet reflectance than their habitat. Second, the Mt. Hood and Tillamook population male abdomens diverged from their habitat in a similar, although less pronounced manner to that of the Gorge and Siskiyou population male anteriors: they had proportionally less green and more UV reflectance. Third, female abdomens of all populations were highly variable in chroma, despite having hues that generally matched their habitat. Tibia area relative to body size of Gorge and Siskiyou population males was significantly smaller than that of Mt. Hood and Tillamook population males. The lower level of background hue matching among males compared to females implies that sexual selection has directly conflicted with natural selection, resulting in impaired crypsis. While the reduced crypsis of the Gorge and Siskiyou population males is centered on their anterior (the primary body region presented to the females during courtship), the deviations from crypsis in the Mt. Hood and Tillamook population males are highest on their abdomen, although the degree of ii contrast is lower than that of the Gorge and Siskiyou population anteriors. These differences in coloration between the Mt. Hood and Tillamook population male abdomens and their habitats are in the same colorimetric direction as those of the Gorge and Siskiyou population anteriors and their habitat; this may indicate a sensory bias of the females, conserved in all four populations, selecting for male reflectance with a higher UV to green ratio. The fact that Mt. Hood and Tillamook population male abdomens have a more modest reduction in background matching compared to Gorge and Siskiyou population male anteriors may be due to the search methods of flying predators (e.g., spider wasps); the dorsum would presumably be more conspicuous to predators than the anterior, and would thus be subject to more intense selection for crypsis despite sexual selection to the contrary. The variability of abdomen coloration of females of both morphs may indicate that selection for crypsis is less strong among females than among males. One possible reason for this would be if females spent less time in the exposed courtship habitat than males, a conclusion implied by a highly male-skewed sex ratio encountered during field collections. Like the differences in coloration between different males of different populations, the significant differences in male tibia size also imply variability in the intensity of sexual selection. Relative importance of male coloration and tibia size may be weighted differently among populations, operating under similar constraints on reductions in survival accrued by developing these characters. iii The high degree of variation found among the populations implies that there is a degree of reproductive isolation among the chromatically and morphologically dissimilar populations. However, the similarity of the environments in which the populations existed, the close geographic proximity of some of the dissimilar populations, and the lack of any substantial geographic boundaries between the populations imply that this isolation is not maintained through extrinsic factors. Rather, it would seem that the interpopulational diversity is maintained by sexual selection. Most of 37 the transfer RNA gene sequences are greatly reduced in size and cannot be folded into typical 38 cloverleaf-shaped secondary structures. At least nine of the tRNA sequences lack the potential to 39 form TYC arm stem pairings, and instead are inferred to have TV-replacement loops. 40 Furthermore, sequences that could encode the 3' aminoacyl acceptor stems in at least 10 tRNAs 41 appear to be lacking, because fully paired acceptor stems are not possible and because the 42 downstream sequences instead encode adjacent genes. Hence, these appear to be among the 43 smallest known tRNA genes.

It is 13,865 bp long and featured by 22 transfer RNA genes (tRNAs), and two ribosomal RNA genes (rRNAs), 13 protein-coding genes (PCGs), and a control region (CR). Most of the PCGs used ATN start codon except cox3, and nad4 with TTG. Comparative studies indicated the use of TTG, TTA, TTT, GTG, CTG, CTA as start codons by few PCGs. Most of the tRNAs were truncated and do not fold into the typical cloverleaf structure. Further, the motif (CATATA) was detected in CR of nine species including L. crotalus. The gene arrangement of L. crotalus compared with ancestral arthropod showed the transposition of fve tRNAs and one tandem duplication random loss (TDRL) event. Five plesiomophic gene blocks (A-E) were identifed, of which, four (A, B, D, E) retained in all taxa except family Salticidae. However, block C was retained in Mygalomorphae and two families of Araneomorphae (Hypochilidae and Pholcidae). Out of 146 derived gene boundaries in all taxa, 15 synapomorphic gene boundaries were identifed. TreeREx analysis also revealed the transposition of trnI, which makes three derived boundaries and congruent with the result of the gene boundary mapping. Maximum likelihood and Bayesian inference showed similar topologies and congruent with morphology, and previously reported multi-gene phylogeny. Spiders are the seventh most diverse order of arthropods globally and are prominent predators in all prairie habitats. In this chapter, a checklist for the spiders of the Prairie Provinces (767 recorded species and 44 possible species) is presented along with an overview of all 26 families that occur in the region. Eighteen of the species from the region are adventive. Linyphiidae is by far the dominant family, representing 39% of all species in the three provinces. Gnaphosidae and Lycosidae each represent 8% and three other families (Salticidae, Dictynidae, and Theridiidae) each account for 7%. A summary of biodiversity studies conducted in the Prairies Ecozone and from transition ecoregions is also provided. The Mixed Grassland Ecoregion has the most distinctive assemblage; Schizocosa mccooki and Zelotes lasalanus are common only in this ecoregion. Other ecoregions appear to harbour less distinctive assemblages, but most have been poorly studied. Lack of professional opportunities for spider systematists in Canada remains a major barrier to the advancement of the taxonomy and ecology of spiders. Résumé. The data matrix used for the analyses includes 84 taxa and 3918 nucleotides representing six mitochondrial protein-coding genes (atp6 and 8, cox1–3, and nad2). The analyses of nucleotide composition show that a reverse strand-bias, i.e., characterized by an excess of T relative to A nucleotides and of G relative to C nucleotides, was independently acquired in six different lineages of Arthropoda: (1) the honeybee mite (Varroa), (2) Opisthothelae spiders (Argiope, Habronattus, and Ornithoctonus), (3) scorpions (Euscorpius and Mesobuthus), (4) Hutchinsoniella (Cephalocarid), (5) Tigriopus (Copepod), and (6) whiteflies (Aleurodicus and Trialeurodes). Phylogenetic analyses confirm that these convergences in nucleotide composition can be particularly misleading for tree reconstruction, as unrelated taxa with reverse strand-bias tend to group together in MP, ML, and Bayesian analyses. However, the use of a specific model for minimizing effects of the bias, the ‘‘Neutral Transition Exclusion’’ (NTE) model, allows Bayesian analyses to rediscover most of the higher taxa of Arthropoda.

    Phylogenetic Utility and Evidence for Multiple Copies of Elongation Factor-1a in the Spider Genus Habronattus (Araneae: Salticidae) Marshal C. Hedin1 and Wayne P. Maddison Department of Ecology and Evolutionary Biology, University of Arizona In the continuing quest for informative genes for use in molecular systematics, the protein-coding gene Elongation factor±1a (EF-1a) has rapidly become one of the most prevalent ``single-copy'' nuclear genes utilized, particularly in arthropods. This paper explores the molecular evolutionary dynamics and phylogenetic utility of EF-1a in the salticid spider genus Habronattus. As has been reported for other arthropod lineages, our studies indicate that multiple (two) copies of EF-1a exist in Habronattus. These copies differ in intron structure and thus in size, making it possible to easily separate PCR ampli®cation products. We present data for an intronless EF-1a copy for three Habronattus species. The presence of nonsense mutations and generally elevated rates of amino acid change suggest that this copy is evolving under relaxed functional constraints in Habronattus. A larger taxon sample (50 species plus outgroups) is presented for an EF-1a copy that includes both intron and exon regions. Characteristics of both regions suggest that this is a functional, orthologous copy in the species sampled. Maximum-likelihood relative- rate comparisons show that exon third codon sites are evolving more than 100 times as fast as second codon sites in these sequences and that intron sites are evolving about twice as fast as exon third sites. In combination, the EF- 1a data provide robust, species-level phylogenetic signal that is largely congruent with morphologically well sup- ported areas of Habronattus phylogeny.
