www.nature.com/scientificreports OPEN Establish axenic cultures of armored and unarmored marine dinofagellate species using density separation, antibacterial treatments and stepwise dilution selection Thomas Chun‑Hung Lee1, Ping‑Lung Chan1, Nora Fung‑Yee Tam2, Steven Jing‑Liang Xu1 & Fred Wang‑Fat Lee1* Academic research on dinofagellate, the primary causative agent of harmful algal blooms (HABs), is often hindered by the coexistence with bacteria in laboratory cultures. The development of axenic dinofagellate cultures is challenging and no universally accepted method suit for diferent algal species. In this study, we demonstrated a promising approach combined density gradient centrifugation, antibiotic treatment, and serial dilution to generate axenic cultures of Karenia mikimotoi (KMHK). Density gradient centrifugation and antibiotic treatments reduced the bacterial population from 5.79 ± 0.22 log10 CFU/mL to 1.13 ± 0.07 log10 CFU/mL. The treated KMHK cells were rendered axenic through serial dilution, and algal cells in diferent dilutions with the absence of unculturable bacteria were isolated. Axenicity was verifed through bacterial (16S) and fungal internal transcribed spacer (ITS) sequencing and DAPI epifuorescence microscopy. Axenic KMHK culture regrew from 1000 to 9408 cells/mL in 7 days, comparable with a normal culture. The established methodology was validated with other dinofagellate, Alexandrium tamarense (AT6) and successfully obtained the axenic culture. The axenic status of both cultures was maintained more than 30 generations without antibiotics. This efcient, straightforward and inexpensive approach suits for both armored and unarmored dinofagellate species. ABSTRACT The origins and status of the non-endemic termite species established in Florida are re- viewed including Cryptotermes brevis and Incisitermes minor (Kalotermitidae), Coptotermes formosanus, Co. gestroi, and Heterotermes sp. (Rhinotermitidae), and Nasutitermes corniger (Termitidae). A lone colony of Marginitermes hubbardi (Kalotermitidae) collected near Tam- pa was destroyed in 2002. A mature colony of an arboreal exotic, Nasutitermes acajutlae, was destroyed aboard a dry docked sailboat in Fort Pierce in 2012. Records used in this study were obtained entirely from voucher specimen data maintained in the University of Flori- da Termite Collection. Current distribution maps of each species in Florida are presented. Invasion history suggests that established populations of exotic termites, without human intervention, will continue to spread and flourish unabatedly in Florida within climatically suitable regions. Foundations of Science https://doi.org/10.1007/s10699-020-09688-8 Superorganisms of the Protist Kingdom: A New Level of Biological Organization Łukasz Lamża1 © The Author(s) 2020 Abstract The concept of superorganism has a mixed reputation in biology—for some it is a conveni- ent way of discussing supra-organismal levels of organization, and for others, little more than a poetic metaphor. Here, I show that a considerable step forward in the understand- ing of superorganisms results from a thorough review of the supra-organismal levels of organization now known to exist among the “unicellular” protists. Limiting the discussion to protists has enormous advantages: their bodies are very well studied and relatively sim- ple (as compared to humans or termites, two standard examples in most discussions about superorganisms), and they exhibit an enormous diversity of anatomies and lifestyles. This allows for unprecedented resolution in describing forms of supra-organismal organiza- tion. Here, four criteria are used to diferentiate loose, incidental associations of hosts with their microbiota from “actual” superorganisms: (1) obligatory character, (2) specifc spatial localization of microbiota, (3) presence of attachment structures and (4) signs of co-evolu- tion in phylogenetic analyses. Three groups—that have never before been described in the philosophical literature—merit special attention: Symbiontida (also called Postgaardea), Oxymonadida and Parabasalia. Specifcally, it is argued that in certain cases—for Bihos- pites bacati and Calkinsia aureus (symbiontids), Streblomastix strix (an oxymonad), Joe- nia annectens and Mixotricha paradoxa (parabasalids) and Kentrophoros (a ciliate)—it is fully appropriate to describe the whole protist-microbiota assocation as a single organism (“superorganism”) and its elements as “tissues” or, arguably, even “organs”. -
