Phylogeny and Systematics of the Rauvolfioideae
PHYLOGENY AND SYSTEMATICS Andre´ O. Simo˜es,2 Tatyana Livshultz,3 Elena OF THE RAUVOLFIOIDEAE Conti,2 and Mary E. Endress2 (APOCYNACEAE) BASED ON MOLECULAR AND MORPHOLOGICAL EVIDENCE1 ABSTRACT To elucidate deeper relationships within Rauvolfioideae (Apocynaceae), a phylogenetic analysis was conducted using sequences from five DNA regions of the chloroplast genome (matK, rbcL, rpl16 intron, rps16 intron, and 39 trnK intron), as well as morphology. Bayesian and parsimony analyses were performed on sequences from 50 taxa of Rauvolfioideae and 16 taxa from Apocynoideae. Neither subfamily is monophyletic, Rauvolfioideae because it is a grade and Apocynoideae because the subfamilies Periplocoideae, Secamonoideae, and Asclepiadoideae nest within it. In addition, three of the nine currently recognized tribes of Rauvolfioideae (Alstonieae, Melodineae, and Vinceae) are polyphyletic. We discuss morphological characters and identify pervasive homoplasy, particularly among fruit and seed characters previously used to delimit tribes in Rauvolfioideae, as the major source of incongruence between traditional classifications and our phylogenetic results. Based on our phylogeny, simple style-heads, syncarpous ovaries, indehiscent fruits, and winged seeds have evolved in parallel numerous times. A revised classification is offered for the subfamily, its tribes, and inclusive genera. This project presents work on Voacanga africana (Apocynaceae) a local Ghanaian plant, in which the leaves and stem barks were screened for antioxidant and resistant modulatory properties. The antioxidant assay was determined using DPPH scavenging, total phenolic count and total antioxidant capacity. The antimicrobial and bacterial resistance modulation activity was determined using the broth dilution and agar well diffusion methods using S. aureus, E. coli, S. typhi, P. auregenosa, B. subtilis and Candida albicans. Further work was performed by looking at the phytochemical constituents of the extracts and isolates. The antioxidant activity as seen from the DPPH assay was concentration dependent with the methanolic extract of the stem bark showing more activity than the leaf, with Ic50 of 70 and 1605 respectively, whilst that of the standard (ascorbic acid) was 6.980. -

Another winter has passed and I 2005 trust that everyone survived. Spring is here and for Lela and I, the last couple of weekends have been beautiful and full of those early spring landscaping and gardening chores. Around our house, I have designated this year as the year of the wasp. We have a bumper crop of wasps buzzing around, looking for places to build homes. As I am somewhat allergic, much of my time has been spent urging them to move on. Our Spring Meeting and Natural Bridge State Resort Park’s 25th annual wildflower weekend is coming up and it looks to be great one as usual. If you’ve never made it to one of our annual Spring meetings, I’d encourage you to do so. Everyone has a great time, multiple wildflower walks to choose from and informative presentations on Friday and Saturday nights. Showy Goldenrod Our Native Plant Certification program at Northern Kentucky University is doing great. We have a Solidago speciosa var. speciosa number of people taking every course, targeting see page 7 for more information the certification program and we have also had our share of people just interested in one or two courses.
