The microalgae are cultivated under heterotrophic conditions, which involves a nutrient feed of organic carbon substrate, to prevent light dependency. Glucose yields high cell density and lipid content for biofuel production from Auxenochlorella protothecoides under nitrogen-deplete conditions but leads to chloroplast bleaching. Hence, waste streams from the biodiesel industry, in the form of glycerol, are incorporated in the batch level system in order to modulate the pigment content. The glycerol rich conditions enabled production of significantly high levels of lutein and beta-carotene at about 52.43 mg.L-1 and 6.2 mg.L-1 respectively. High biomass growth of 11.53 g.L-1 was reported for C:N ratio of 10:1 in glucose rich conditions. Since distinct nutrients promote accumulation of specific products, a mathematical model is used to simulate and predict micro-algal response to varying cultivation conditions in the batch level operation. For a reliable model, the biochemical reactions within the micro-algae are reconstructed and calibrated to predict the performance results for varying nutrient conditions. The mathematical model enables scale-up and control of process conditions within the bioreactor system. A sustainable and feasible solution for a micro-algal production system strengthens the bio-economy and renewable energy sector. ii Preface This thesis is original, unpublished, independent work by the author, S.     1 AMINO ACIDS Commonly, 21 L-amino acids encoded by DNA represent the building blocks of animal, plant, and microbial proteins. The basic amino acids encountered in proteins are called proteinogenic amino acids 1.1). Biosynthesis of some of these amino acids proceeds by ribosomal processes only in microorganisms and plants and the ability to synthesize them is lacking in animals, including human beings. These amino acids have to be obtained in the diet (or produced by hydrolysis of body proteins) since they are required for normal good health and are referred to as essential amino acids. The essential amino acids are arginine, histidine, isoleucine, leucine, lysine, methionine, phenylalanine, threonine, tryptophan, and valine. The rest of encoded amino acids are referred to as non-essential amino acids (alanine, asparagine, aspartic acid, cysteine, glutamic acid, glutamine, glycine, proline, serine, and tyrosine). Arginine and histidine are classified as essential, sometimes as semi-essential amino acids, as their amount synthesized in the body is not sufficient for normal growth of children. Although it is itself non-essential, cysteine (classified as conditionally essential amino acid) can partly replace methionine, which is an essential amino acid. Similarly, tyrosine can partly replace phenylalanine. 1.1 The glutamic acid group 1.1.1 Glutamic acid and glutamine Free ammonium ions are toxic to living cells and are rapidly incorporated into organic compounds. One of such transformations is the reaction of ammonia with 2-oxoglutaric acid from the citric acid cycle to produce L-glutamic acid. This reaction is known as reductive amination. Glutamic acid is accordingly the amino acid generated first as both constituent of proteins and a biosynthetic precursor.

Positively-charged inhibitors that mimic the transition states/intermediates formed with these enzymes were synthesized. In chapter two, inhibitor 2 containing a guanidinium moiety appended to a phosphonylphosphinate was designed to mimic the transition state for the dissociation of dimethylallyl diphosphate into an allylic carbocation-pyrophosphate ion-pair. To test for the effectiveness of incorporating a guanidinium functionality into inhibitors of human farnesyl diphosphate synthase, inhibitors 3 and 4 were also prepared. Inhibitor 3 has a positive charge localized onto one atom, and inhibitor 4 is isosteric to inhibitor 2, but lacks positive charge. The inhibitors displayed IC50 values that were significantly higher than the substrate Km value, indicating that the positive charge did not result in tight binding to the enzyme.

Monad Shoal is a standard municipality MPA, and 150 Pesos (US$ 3) per visitor to the island per day is collected by the municipality as a marine park fee. While some of the fees collected have been used to install mooring buoys, how the majority of the funds are used is unclear, and enforcement of the ban on all fishing within the MPA was weak.

A true macro critter playground. LIGHTHOUSE, CABILAO ISLAND Sloping walls full of huge elephant ear sponges and gorgonian fans. This is a great place to dive with large green turtles. Giant frogfish can be spotted resting on sponges and numerous cowries hiding amidst the stunning hard coral garden. Often, we can see schools of surgeon fish and big-mouth mackerel. Drift along the wall and as you ascend towards the shallows a sandy plateau dotted with gorgonian fans is the place the stop and look for pygmy seahorses. The dive ends at a stunning hard coral garden where pipefish, cowries, cuttlefish, commensal shrimps, nudibranchs, squat lobsters and orangutan crabs are amongst a few of the commonly seen critters. Occasionally some stronger currents can be experienced! DIVER'S HEAVEN, BALICASAG ISLAND Typically, with stronger currents than most dives during the trip, Diver’s Heaven is a character site of Balisage with a steep reef wall, many overhangs with soft corals, sponges & sea fans. The main attraction of this beautiful dive site is the green sea and hawksbill turtles in the shallow waters. The reef top edge is a nice habitat for schools of colorful damsels and anthias and when lucky also for giant frogfish.

Found off the northern coast of Bohol Island, central Philippines, Danajon expands over an area of 272 km². The Coastal Conservation and Education Foundation, a Filipino nonprofit, is implementing a project to develop a management plan for Danajon Bank. One of the goals is to increase the economic opportunities for the coastal communities adjacent to the Danajon reefs through marine-related tourism. This study examined the feasibility of expanding dive tourism to this area. The results were derived from two methods: (1) semi-structured interviews with dive business owners and employees; and (2) contingent valuation survey among foreign and local tourists to determine willingness-to-pay an entrance fee to the reefs as a dive destination.
