We demonstrate that in R. prolixus following infection with epimastigotes of Trypanosoma cruzi clone Dm28c and, in comparison with uninfected control insects, the midgut contained (i) fewer bacteria, (ii) higher parasite numbers, and (iii) reduced nitrite and nitrate production and increased phenoloxidase and antibacterial activities. In addition, in insects pre-treated with antibiotic and then infected with Dm28c, there were also reduced bacteria numbers and a higher parasite load compared with insects solely infected with parasites. Furthermore, and in contrast to insects infected with Dm28c, infection with T. cruzi Y strain resulted in a slight decreased numbers of gut bacteria but not sufficient to mediate a successful parasite infection. We conclude that infection of R. prolixus with the T. cruzi Dm28c clone modifies the host gut immune responses to decrease the microbiota population and these changes are crucial for the parasite development in the insect gut. -

Its main reservoir is the domestic dog, especially in rural areas with favorable characteristics for vector establishment and proliferation. The aims of this study were to collect data, survey and map the fauna, and identify T. cruzi infection in triatomines, as well as to assess the presence of anti-T. cruzi antibodies in dogs in rural areas of the municipality of Mossoró, Brazil. Methods: An active entomologic research was conducted to identify adult specimens through an external morphology dichotomous key. The analysis of natural infection by T. cruzi in the insects was performed by isolation in culture and polymerase chain reaction. -

Among these, thrombin inhibitors have most frequently been isolated. In order to study the thrombin inhibitor from Triatoma brasiliensis and its biological significance for the bug, we sequenced the corresponding gene and evaluated its biological function. The T. brasiliensis intestinal thrombin inhibitor, termed brasiliensin, was sequenced and primers were designed to synthesize double strand RNA (dsRNA). Gene knockdown (RNAi) was induced by two injections of 15 lg of dsRNA into fourth instar nymphs. Forty-eight hours after the second injection, bugs from each group were allowed to feed on hamsters. PCR results showed that injections of dsRNA reduced brasiliensin expression in the ante- rior midgut by approximately 71% in knockdown nymphs when compared with controls. The reduction in gene expression was con- firmed by the thrombin inhibitory activity assay and the citrated plasma coagulation time assay which showed activity reductions of 18- and 3.5-fold, respectively. -

In this work, we performed a histological characterization of the fat body of the Chagas’ disease vector, Panstrongylus megistus (Burmeister), subjected to different nutritional conditions. In addition, we addressed the role of the β-chain of ATP synthase (β-ATPase) in the process of lipid transfer from lipophorin to the fat body. Fifth-instar nymphs in either fasting or fed condition were employed in the assays. Histological examination revealed that the fat body was composed by diverse trophocyte phenotypes. -

ISBN 91-554-5756-8 This thesis analyses several ecological factors affecting the control of triatomines in Guatemala. There are three synanthropic triatomines in Guatemala, i. e., Rhodnius prolixus, Triatoma dimidiata and T. nitida. Their distribution is mainly at an altitude between 800 and 1500 m. a. s. l. R. prolixus and T. nitida have localized but scattered distributions while T. dimidiata is present in 21 of the 22 departments in the country. Several investigations have shown that R. prolixus could be relatively easily eradicated while T. dimidiata may be more difficult to control, since it is present in domestic, peridomestic and sylvatic environments showing high diversity and a variety of epidemiological characteristics. Based on the incidence of Trypanosoma cruzi infection in humans in the distributional areas of the triatomines, R. prolixus appears to be a more competent vector than T. dimidiata. This is despite the fact that these vectors have similar infection rates.
