S. Moore) METODE DPPH (1,1-diphenyl-2-picrylhidrazil) ABSTRAK Latar Belakang: Tumbuhan sintrong merupakan herba yang tumbuh liar di kebun, tepi sungai, tanah lembab di wilayah tropis dan sub tropis. Tumbuhan ini mengandung senyawa flavonoid, saponin, tanin, glikosida, dan steroid/triterpenoid. Tujuan: Untuk mengetahui karakterisasi simplisia daun tumbuhan sintrong, skrining fitokimia serbuk simplisia dan ekstrak etanol daun tumbuhan sintrong, dan uji aktivitas antioksidan ekstrak etanol daun tumbuhan sintrong. Metode: Ekstraksi dilakukan secara perkolasi menggunakan pelarut etanol, pengujian aktivitas antioksidan ekstrak etanol daun tumbuhan sintrong dilakukan menggunakan metode DPPH (1,1-diphenyl-2-picryhydrazyl) sebagai pembanding Vitamin C dan diukur menggunakan alat spektrofotometer UV-Visibel. Hasil: Hasil karakterisasi simplisia daun tumbuhan sintrong diperoleh kadar air 7,98%; kadar sari larut dalam air 23,02%; kadar sari larut dalam etanol21,24%; kadar abu total 4,08%; dan kadar abu tidak larut asam 0,91%. Hasil skrining fitokimia serbuk simplisia dan ekstrak etanol daun tumbuhan sintrong memberikan hasil yang sama yaitu flavonoid, saponin, tanin, glikosida, dan steroid/triterpenoid. Hasil uji aktivitas antioksidan ekstrak etanol daun tumbuhan sintrong diperoleh nilai 5 sebesar 51,3792 mcg/ml dan Vitamin C diperoleh nilai IC50 sebesar 3,2179 mcg/ml. Kesimpulan: Karakterisasi simplisia daun tumbuhan sintrong memenuhi persyaratan mutu simplisia. Hasil skrining fitokimia yang terdapat pada serbuk simplisia dan ekstrak etanol daun tumbuhan sintrong memberikan hasil yang sama yaitu flavonoid, saponin, tanin, glikosida, dan steroid/triterpenoid. Ekstrak etanol daun tumbuhan sintrong memiliki aktivitas antioksidan kuat sedangkan Vitamin C memiliki aktivitas antioksidan sangat kuat. Kata Kunci: Tumbuhan sintrong, karakterisasi, antioksidan, DPPH vii UNIVERSITAS SUMATERA UTARA CHARACTERIZATION OF SIMPLICIA AND ANTIOXIDE ACTIVITY TESTS OF ETHANOL EXTRACT SINTRONG LEAF (Crassocephalum crepidioides (Benth.) S. Moore) DPPH METHOD (1,1-diphenyl-2-picrylhidrazil) ABSTRACT Background: Sintrong plants are herba that grow wild in gardens, river banks, moist soils in tropical and sub-tropical regions. This plant contains tannin compounds, glycosides, saponins, flavonoids, and steroids / triterpenoids. Objective: To determine the charactesrictics of simplex plants leaves of sintrong, phytochemical screening of simplicia powders and ethanol extracts of sintrong plant leaves, and antioxidant activity of ethanol extracts of sintrong plant leaves. Method: Extraction was carried out by percolation using ethanol solvent, antioxidant activity of ethanol extract of sintrong plant leaves was carried out using the DPPH (1,1-diphenyl-2-picryhydrazyl) method and comparred with Vitamin C, and measured using a UV-Visible spectrophotometer. Results: The results of the characterization of the simplex powder of the sintrong plant leaves obtained with moisture content of 7.98 %; water soluble extract content 23.02 %; soluble extract content in ethanol 21.24 %; total ash content of 4.08 %; and acid insoluble ash content of 0.91 %. The results of phytochemical simplicia screening and ethanol extract of sintrong plant leaves gave the same results, namely flavonoids, saponins, tannins, glycosides, and steroids/triterpenoids. The antioxidant activity test results of the ethanol extract of sintrong plant leaves obtained IC50 values of 51,3792 mcg/ml and Vitamin C obtained IC50 value is 3.2179 mcg/ml. Conclusion: Simplicia characterization of sintrong plant leaves meets the quality requirements of simplicia. The results of phytochemical screening found in simplicia powder and ethanol extract of sintrong plant leaves gave the same results, namely flavonoids, saponins, tannins, glycosides, and steroids/triterpenoids. The ethanol extract of the leaves of the sintrong plant has a strong antioxidant activity while Vitamin C has a very strong antioxidant activity. While the intermediates were well defined by feeding experiments, only one enzyme involved in PA biosynthesis has been characterized so far, the homospermidine synthase catalyzing the first committed step in PA biosynthesis. This review gives an overview about structural diversity of PAs, biosynthetic pathways of necine base, and necic acid formation and how PA accumulation is regulated. Furthermore, we discuss their role in plant ecology and their modes of toxicity towards humans and animals.

Pollen sizes were obtained from polar and equatorial diameter measurements. Pollen fertility was accessed through the staining property in cot- ton blue in lactophenol reagent. The structural form of pollen grains was examined by means of a light microscope (X100 magnification). There was no significant difference in pollen size, structure and fertility among species investigated. C. crepidioides had relatively the largest size of pollen grains (28.10±1.67 µm) (media), while C. rubens pollen were relatively most fertile (98.7%). However, pollen grains of the species were tricolporate with three germ pores except in C. crepidioides which had up to six germ pores (multiporate). All pollens in the species were fenestrate type with high fertility. The study suggests that number of germ pores in addition to pollen morphology be used in species separa- tion. Accepted 30 August, 2013 The morphotaxonomic identity of Crassocephalum biafrae (Oliv. & Hiern.) S. Moore was compared with other three taxa namely: Crassocehalum crepidioides (Benth.) S. Moore, Crassocephalum rubens (Juss.ex.Jacq.) S. Moore and, Crassocephalum togoense C.D. Adams in the genus Crassocephalum tribe Senecioneae; with the intent of establishing some delimiting features truly uniting (or otherwise) the taxa. C. biafrae was clearly separable from C. crepidioides, C. rubens and C. togoense on the basis of growth habit, stem texture, leaf shape, size, apex, and texture. Other morphological evidences separating these are well documented. Morphological characters considered being of taxonomic significance were widely manifested by C. biafrae as differing from the other three species. There was evidence of more similarities and overlaps in morphological attributes of these other three species. However, the somewhat narrow morphological similarities exhibited by C. biafrae to the other three species suggest infrageneric segregation in the genus. We hereby suggest the infrageneric group for C. biafrae from the genus Crassocephalum based on its strikingly distinctive morphological characteristics.

Abstract: Somatic chromosome numbers of four species in the genus Crassocephalum were counted. Basic chromosome number of n=10 which was tentatively suggested for the genus by previous workers was confirmed. Three species, Crassocephalum rubens (Juss.ex.Jacq) S. Moore, C. biafrae (Oliv.& Hiern) S. Moore and C. togoense C.D. Adams had chromosome numbers of 2n=20 while 2n=40 was reported for the C. crepidioides (Benth.) S. Moore. The occurence of higher chromosome number in C. crepidioides suggests the species to be polyploid. Basic chromosome number of n=10 was confirmed for the genus. Key words: Crassocephalum, Basic chromososme number. I. Introduction Crassocephalum (Moench.) S. Moore occur throughout tropical Africa with 24 species. In West Africa, the genus is represented by fifteen species, with nine species reported in Nigeria. Of these, four species collected for this study are endemic to Southwestern Nigeria. (Hutchinson and Dalziel, 1963). The four species are Crassocephalum crepidioides, C. biafrae, C. rubens and, C. togoense (Hutchinson and Dalziel, 1963; Olorode, 1974; Olorode and Okoli, 1978). Crassocephalum constitutes special delicacies in the Southwestern, Nigeria and feature prominently among numerous edible plants common to the Yoruba culture (Bankole et al., 2003).

All over the world, plant domestication is continually being carried out by local communities to support their needs for food, fibre, medicine, building materials, etc. Using participatory rapid appraisal approach, 150 households were surveyed in 5 villages selected in five ethnic groups of Benin, to investigate the local communities’ motivations for plant domestication and the contributions of this process to in situ conservation of genetic resources. The results indicated differences in plant domestication between agroecological zones and among ethnic groups. People in the humid zones give priority to herbs mainly for their leaves while those in dry area prefer trees mostly for their fruits.

MOORE (COMPOSITAE) IN NIGERIA Kemka, C.I.1 & Nwachukwu, C.U.2 Department of Biology, Alvan Ikoku Federal College of Education Owerri, Imo State, Nigeria ABSTRACT Detailed studies on eight Crassocophalum species ( biafrae, C. montuosum, C. mannii, C. crepidioides C. vitellinum, C. rubens, C.togoense and C. sarcobasis) found in Nigeria, which are important leaf vegetables were undertaken with the aid of a microscope to ascertain their taxonomic relationship. The micromorphological features discovered include the hypostomatic distribution of stomata in C. mannii while the other species had hypoamphistomatic. Sinuous anticlinal wall was present on adaxial (upper) surfaces of C. togoense and C.crepidioides, the clusters of trichomes on mid-vein of the adaxial surfaces of C.vitellinum and C. crepidioides.The high density of trichomes on the veins of the adaxial surface of C.mannii is of taxonomic interest and could be used to demilit the taxon from the other species. C.vitellinum, C.crepidioides and C. togoense which are densely hairy could also be delimited from C.biafrae and C.montuosum that are sparsely hairy.A mixture of sinuous and straight-arcuate anticlinal wall was present in both the abaxial and adaxial surfaces of all crassocephalum species studied except in C.togoense and C.crepidioides that has sinuous anticlinal wall in both adaxial and abaxial surfaces.
