Official Proceedings Presentation Awards and Eligibility Abstracts submitted are eligible for awards. The George Peters Award recognizes the best presentation by a breast fellow and is awarded $1,000. The Scientific Presentation Award recognizes an outstanding presentation by a resident or fellow and is awarded $500. All presenters are eligible for the Scientific Impact Award. The recipient of the award is selected by the audience. The awards are supported by The American Society of Breast Surgeons Foundation. The George Peters Award was established in 2004 by the Society to honor Dr. George N. Peters, who was instrumental in bringing together the Susan G. Komen Breast Cancer Foundation, The American Society of Breast Surgeons, the American Society of Breast Disease, and the Society of Surgical Oncology to develop educational objectives for breast fellowships. The educational objectives were first used to award Komen Interdisciplinary Breast Fellowships. Subsequently the curriculum was used for the breast fellowship credentialing process that has led to the development of a nationwide matching program for breast fellowships. TABLE OF CONTENTS I. Endocrine therapies that target these nuclear receptors (NRs) resistance provide significant clinical benefit for metastatic patients. Because the vast f estrogen receptor majority of therapy-resistant PCa and BCa remain dependent on the augmented activity f oncogene of their primary NR driver, common mechanisms of resistance involve enhanced NR signaling through overexpression, mutation, or alternative splicing of the receptor, coregulator alterations, and increased intracrine hormonal synthesis. In addition, a significant subset of endocrine therapy-resistant tumors become independent of their primary NR and switch to alternative NR or transcriptional drivers. While these hormone-dependent cancers generally employ similar mechanisms of endocrine therapy resistance, distinct differences between the two tumor types have been observed.

Approximately 70-80% of BC diagnoses are positive for estrogen receptor (ER) alpha (ERα). The steroid hormone estrogen [17β-estradiol (E2)] plays a vital role both in the initiation and progression of BC. The E2-ERα mediated actions involve genomic signaling and non-genomic signaling. The specificity and magnitude of ERα signaling are mediated by interactions coregulators are common during BC progression and they enhance ligand-dependent and ligand-independent between ERα and several coregulator proteins called coactivators or corepressors. Alterations in the levels of proteins function as scaffolding proteins and some have intrinsic or associated enzymatic activities, thus the ERα signaling which drives BC growth, progression, and endocrine therapy resistance.

We have previously reported that in monocytes, estrogen regulates TLR4/NFκB-mediated inflammation via the interaction of the Erα isoform ERα36 with GPER1. We therefore investigated whether a similar mechanism is present in breast cancer epithelial cells, and the effect of ERα36 expression on the classic 66 kD ERα isoform (ERα66) functions. We report that estrogen inhibits LPS-induced NFκB activity and the expression of downstream molecules TNFα and IL-6. In the absence of ERα66, ERα36 and GPER1 are both indispensable for this effect. In the presence of ERα66, ERα36 or GPER1 knock-down partially inhibits NFκB-mediated inflammation. In both cases, ERα36 overexpression enhances the inhibitory effect of estrogen on inflammation. Phthalate exposure can induce reproductive disorders at various regulatory levels. The aim of this review was to compile the evidence concerning the association between phthalates and reproductive diseases, phthalates-induced reproductive disorders, and their possible endocrine and intracellular mechanisms. Phthalates may induce alterations in puberty, the development of testicular dysgenesis syndrome, cancer, and fertility disorders in both males and females. At the hormonal level, phthalates can modify the release of hypothalamic, pituitary, and peripheral hormones. At the intracellular level, phthalates can interfere with nuclear receptors, membrane receptors, intracellular signaling pathways, and modulate gene expression associated with reproduction. To understand and to treat the adverse effects of phthalates on human health, it is essential to expand the current knowledge concerning their mechanism of action in the organism. Description Elagolix (Orilissa™) is a gonadotropin-releasing hormone (GnRH) receptor antagonist. Elagolix/estradiol/norethinedrone; elagolix (Oriahnn™) is a combination of a GnRH receptor antagonist with an estrogen and progestin. FDA Approved Indication(s) Orilissa is indicated for the management of moderate to severe pain associated with endometriosis. Oriahnn is indicated for the management of heavy menstrual bleeding associated with uterine leiomyomas (fibroids) in premenopausal women. Limitation(s) of use: Use of Oriahnn should be limited to 24 months due to the risk of continued bone loss, which may not be reversible. Policy/Criteria Provider must submit documentation (such as office chart notes, lab results or other clinical information) supporting that member has met all approval criteria. Health plan approved formularies should be reviewed for all coverage determinations. Kelly1 1Department of Physiology and Pharmacology, Oregon Health and Science University, Portland, Oregon 97239, and 2Department of Pharmaceutical Chemistry, University of California, San Francisco, San Francisco, California 94143 ␤ Classically, 17 -estradiol (E2 ) is thought to control homeostatic functions such as reproduction, stress responses, feeding, sleep cycles, temperature regulation, and motivated behaviors through transcriptional events. Although it is increasingly evident that E2 can also rapidly activate kinase pathways to have multiple downstream actions in CNS neurons, the receptor(s) and the signal transduction ␮ pathways involved have not been identified. We discovered that E2 can alter -opioid and GABA neurotransmission rapidly through nontranscriptional events in hypothalamic GABA, proopiomelanocortin (POMC), and dopamine neurons. Therefore, we examined the effects of E2 in these neurons using whole-cell recording techniques in ovariectomized female guinea pigs. E2 reduced rapidly the potency ϩ of the GABAB receptor agonist baclofen to activate G-protein-coupled, inwardly rectifying K channels in hypothalamic neurons. These effects were mimicked by the membrane impermeant E2–BSA and selective estrogen receptor modulators, including a new diphenylac- ␣ ␤ rylamide compound, STX, that does not bind to intracellular estrogen receptors or , suggesting that E2 acts through a unique ␣ membrane receptor. We characterized the coupling of this estrogen receptor to a G q-mediated activation of phospholipase C, leading to the upregulation of protein kinase C␦ and protein kinase A activity in these neurons.
