Claviceps purpurea is one of the most important species, being a major producer of EAs that infect more than 400 species of monocotyledonous plants. Rye, barley, wheat, millet, oats, and triticale are the main crops affected by EAs, with rye having the highest rates of fungal infection. The 12 major EAs are ergometrine (Em), ergotamine (Et), ergocristine (Ecr), ergokryptine (Ekr), ergosine (Es), and ergocornine (Eco) and their epimers ergotaminine (Etn), egometrinine (Emn), egocristinine (Ecrn), ergokryptinine (Ekrn), ergocroninine (Econ), and ergosinine (Esn). Given that many food products are based on cereals (such as bread, pasta, cookies, baby food, and confectionery), the surveillance of these toxic substances is imperative. Although acute mycotoxicosis by EAs is rare, EAs remain a source of concern for human and animal health as food contamination by EAs has recently increased. Environmental conditions, such as low temperatures and humid weather before and during flowering, influence contamination agricultural products by EAs, contributing to the Citation: Agriopoulou, S. The contribution by the numerous grape-must-associated non-Saccharomyces yeasts to wine fermentation has been debated extensively. These yeasts, naturally present in all wine fermentations, are metabolically active and their metabolites can impact on wine quality. Although often seen as a source of microbial spoilage, there is substantial contrary evidence pointing to a positive contribution by these yeasts. The role of non-Saccharomyces yeasts in wine fermentation is therefore receiving increasing attention by wine microbiologists in Old and New World wine producing countries. Species that have been investigated for wine production thus far include those from the Candida, Kloeckera, Hanseniaspora, Zygosaccharomyces, Schizosaccharomyces, Torulaspora, Brettanomyces, Saccharomycodes, Pichia and Williopsis genera. In this review the use and role of non-Saccharomyces yeast in wine production is presented and research trends are discussed. Deep dermatophytosis may be a rare skin infection, but late diagnosis or ineffective treatment may lead to mortality in some cases. This issue of the AFWG newsletter focuses on this fungal infection that usually occurs in immunosuppressed individuals. Dr Pei-Lun Sun takes us through the basics of deep dermatophytosis, presenting data from published studies, and emphasizes the importance of treating superficial tinea infections before starting immunosuppressive treatment. Dr Ruojun Wang and Professor Ruoyu Li share a case of deep dermatophytosis caused by Trichophyton rubrum. In this issue, we also feature a new fungus, Fereydounia khargensis, first discovered in 2014. Ms Ratna Mohd Tap and Dr Fairuz Amran present 2 cases of F. khargensis and show how PCR sequencing is crucial to correct identification of this uncommon yeast. -
Introduction to Mycology
INTRODUCTION TO MYCOLOGY The term "mycology" is derived from Greek word "mykes" meaning mushroom. Therefore mycology is the study of fungi. The ability of fungi to invade plant and animal tissue was observed in early 19th century but the first documented animal infection by any fungus was made by Bassi, who in 1835 studied the muscardine disease of silkworm and proved the that the infection was caused by a fungus Beauveria bassiana. In 1910 Raymond Sabouraud published his book Les Teignes, which was a comprehensive study of dermatophytic fungi. He is also regarded as father of medical mycology. Importance of fungi: Fungi inhabit almost every niche in the environment and humans are exposed to these organisms in various fields of life. Beneficial Effects of Fungi: 1. Decomposition - nutrient and carbon recycling. 2. Biosynthetic factories. The fermentation property is used for the industrial production of alcohols, fats, citric, oxalic and gluconic acids. 3. Important sources of antibiotics, such as Penicillin. 4. Model organisms for biochemical and genetic studies. Eg: Neurospora crassa 5. Saccharomyces cerviciae is extensively used in recombinant DNA technology, which includes the Hepatitis B Vaccine. 6. Some fungi are edible (mushrooms). 7. Yeasts provide nutritional supplements such as vitamins and cofactors. 8. Penicillium is used to flavour Roquefort and Camembert cheeses. 9. Ergot produced by Claviceps purpurea contains medically important alkaloids that help in inducing uterine contractions, controlling bleeding and treating migraine. 10. Fungi (Leptolegnia caudate and Aphanomyces laevis) are used to trap mosquito larvae in paddy fields and thus help in malaria control. Harmful Effects of Fungi: 1. -

INTRODUCTION Aspergillus niger is a member of the genus Aspergillus which includes a set of fungi that are generally considered asexual, although perfect forms (forms that reproduce sexually) have been found. Aspergilli are ubiquitous in nature. They are geographically widely distributed, and have been observed in a broad range of habitats because they can colonize a wide variety of substrates. A. niger is commonly found as a saprophyte growing on dead leaves, stored grain, compost piles, and other decaying vegetation. The spores are widespread, and are often associated with organic materials and soil. History of Commercial Use and Products Subject to TSCA Jurisdiction The primary uses of A. niger are for the production of enzymes and organic acids by fermentation. While the foods, for which some of the enzymes may be used in preparation, are not subject to TSCA, these enzymes may have multiple uses, many of which are not regulated except under TSCA. Fermentations to produce these enzymes may be carried out in vessels as large as 100,000 liters (Finkelstein et al., 1989). A. niger is also used to produce organic acids such as citric acid and gluconic acid. The history of safe use for A. niger comes primarily from its use in the food industry for the production of many enzymes such as a-amylase, amyloglucosidase, cellulases, lactase, invertase, pectinases, and acid proteases (Bennett, 1985a; Ward, 1989). Numerous phytopathogens decimate crop production by causing various diseases. During July-August 2018, leaf spot symptoms were recurrently observed on cotton leaves in Rahim Yar Khan, Pakistan and adjacent areas. Infected leaf samples were collected and plated on potato dextrose agar (PDA) media. Causal agent of cotton leaf spot was isolated, characterized and identified as Aspergillus tubingensis based on morphological and microscopic observations. Conclusive identification of pathogen was done on the comparative molecular analysis of CaM and β-tubulin gene sequences. BLAST analysis of both sequenced genes showed 99% similarity with A. tubingensis. Koch’s postulates were followed to confirm the pathogenicity of the isolated fungus. Healthy plants were inoculated with fungus and similar disease symptoms were observed. Fungus was re-isolated and identified to be identical to the inoculated fungus. To our knowledge, this is the first report describing the involvement of A. tubingensis in causing leaf spot disease of cotton in Pakistan and around the world. -

The disease symptoms, the morphology of fruiting bodies and fungal culture, and DNA barcoding were combined for determining the fungi at the genus or species level. DNA barcoding is a standardized and automated identification of species based on recognition of highly variable DNA sequences. DNA barcoding has a wide application in the diagnostic purpose of fungi in biological specimens. -

Cairns*, Corrado Nai* and Vera Meyer* Abstract In 1917, a food chemist named James Currie made a promising discovery: any strain of the flamentous mould Aspergillus niger would produce high concentrations of citric acid when grown in sugar medium. This tricarboxylic acid, which we now know is an intermediate of the Krebs cycle, had previously been extracted from citrus fruits for applications in food and beverage production. Two years after Currie’s discovery, industrial-level production using A. niger began, the biochemical fermentation industry started to fourish, and industrial biotechnology was born. A century later, citric acid production using this mould is a multi-billion dollar industry, with A. niger additionally produc- ing a diverse range of proteins, enzymes and secondary metabolites. In this review, we assess main developments in the feld of A. niger biology over the last 100 years and highlight scientifc breakthroughs and discoveries which were infuential for both basic and applied fungal research in and outside the A. niger community. We give special focus to two developments of the last decade: systems biology and genome editing. We also summarize the current interna- tional A. niger research community, and end by speculating on the future of fundamental research on this fascinating fungus and its exploitation in industrial biotechnology. Genetic engineering of fungal strains based on regulatory network of transcriptional factors (TFs) and their targets is an efcient strategy to achieve the above described aim. Talaromyces pinophilus produces integrative amylolytic and cellulolytic enzymes; however, the regulatory mechanism associated with the expression of amylase and cellulase genes in T. pinophilus remains unclear. In this study, we screened for and identifed novel TFs regulating amylase and/or cellulase gene expression in T. pinophilus 1-95 through comparative transcriptomic and genetic analyses. Results: Comparative analysis of the transcriptomes from T. pinophilus 1-95 grown on media in the presence and absence of glucose or soluble starch as the sole carbon source screened 33 candidate TF-encoding genes that regu‑ late amylase gene expression. Thirty of the 33 genes were successfully knocked out in the parental strain T. pinophilus ∆TpKu70, with seven of the deletion mutants frstly displaying signifcant changes in amylase production as compared with the parental strain. Among these, ∆TpRfx1 (TpRfx1: Talaromyces pinophilus Rfx1) showed the most signifcant decrease (81.5%) in amylase production, as well as a 57.7% reduction in flter paper cellulase production.
