Introduction Mis18-binding protein 1 (UniProt Q6P0N0; also known as hsKNL-2, Kinetochore null 2 homolog, Kinetochore-associated protein KNL-2 homolog, P243, Putative protein p243 which interacts with transcription factor Sp1) is encoded by the MIS18BP1 (also known as C14orf106, KIAA1903, KNL2, M18BP1) (Gene ID 55320) in human. Mis18-binding protein 1 is a component of the Mis18 complex (Mis18α, Mis18β, and M18BP1/KNL2) that plays a role in mediating cell cycle- dependent deposition/incorporation of the histone H3 variant CENP-A at the centromeres. CENP-A incorporation is carried out by at least two sets of events, the Mis18 complex must first localize to centromeres at the onset of anaphase, HJURP-mediated CENP-A deposition then occurs during G1. Polo-like kinase 1 (Plk1) is shown to phosphorylate M18BP1 on Thr702 and plays an important role in promoting Mis18 complex centromere localization.
