Anti-MIS18BP1 (phospho T702) polyclonal antibody (CABT-B1900) This product is for research use only and is not intended for diagnostic use. PRODUCT INFORMATION Specificity Specifically reacts with the immunogen peptide, but not the corresponding non-phosphorylated sequence, by dot blot analysis. Phosphorylation site is not present in the spliced isoform 2 of human Mis18-binding protein 1/M18BP1. Immunogen Linear peptide corresponding to a human M18BP1 sequence containing phosphorylated Thr702. Isotype IgG Source/Host Rabbit Species Reactivity Human Purification Affinity Purfied Conjugate Unconjugated Applications ICC, WB, Dot Epitope M18BP1 pThr702 Molecular Weight 129 kDa calculated. Format Liquid Concentration Please refer to lot specific datasheet. Size 100 μg Buffer PBS with 0.05% sodium azide and 30% glycerol. Preservative 0.05% Sodium Azide Storage Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage. 45-1 Ramsey Road, Shirley, NY 11967, USA Email: [email protected] Tel: 1-631-624-4882 Fax: 1-631-938-8221 1 © Creative Diagnostics All Rights Reserved BACKGROUND Introduction Mis18-binding protein 1 (UniProt Q6P0N0; also known as hsKNL-2, Kinetochore null 2 homolog, Kinetochore-associated protein KNL-2 homolog, P243, Putative protein p243 which interacts with transcription factor Sp1) is encoded by the MIS18BP1 (also known as C14orf106, KIAA1903, KNL2, M18BP1) gene (Gene ID 55320) in human. Mis18-binding protein 1 is a component of the Mis18 complex (Mis18α, Mis18β, and M18BP1/KNL2) that plays a role in mediating cell cycle- dependent deposition/incorporation of the histone H3 variant CENP-A at the centromeres. CENP-A incorporation is carried out by at least two sets of events, the Mis18 complex must first localize to centromeres at the onset of anaphase, HJURP-mediated CENP-A deposition then occurs during G1. Polo-like kinase 1 (Plk1) is shown to phosphorylate M18BP1 on Thr702 and plays an important role in promoting Mis18 complex centromere localization.
