Petr Smýkal, Ph.D. Rok obhajoby práce: 2013 Abstrakt: Poznání fylogenetických vztahů v rámci čeledi bobovitých (Leguminosae), je velmi důleţité pro porozumění původu a rozrůznění této ekologicky a ekonomicky důleţité čeledi. Tato třetí nejrozsáhlejší čeleď vyšších rostlin, zahrnuje více neţ 850 rodů s přibliţně 20 tisíci druhy, včetně domestikovaných druhů. Bakalářská práce si klade za cíl, literární rešerši rozsáhlého dostupného materiálu a syntézu dostupných poznatků na téma vyuţití DNA a morfologických znaků pro účely taxonomických a fylogenetických studií rostlin čeledi Leguminosae se zaměřením na podčeleď Fabeae/Vicieae. Bude studována morfologická variabilita a genetická diverzita pomocí DNA sekvencí tří vybraných druhů, identifikovaných v předchozím studiu jako fylogeneticky nejbliţších příbuzných rodu Pisum. Petr Smýkal, Ph.D. The year of representation: 2013 Abstract: Knowledge of phylogenetic relationships within the legume family (Leguminosae) is very important for understanding the origin and diversification of this ecologically and economically important family. This is the third largest family of vascular plants, including more than 850 genera with about 20 thousand species, including domesticated species. Bachelor thesis aims, an extensive literature review and synthesis of available material available knowledge about the use of DNA and morphological characters for taxonomic and phylogenetic studies of plants of the family Leguminosae subfamily focusing on Fabeae / Vicieae. Morphological variability and genetic diversity was studied using DNA sequences of three selected species identified in previous studies as phylogenetically closest relatives of the genus Pisum. Petru Smýkalovi, Ph.D., který mi vţdy ochotně pomáhal a zasvěcoval do problematiky související s touto prací. Naučil mě orientaci v laboratoři a práci potřebné k vykonávání experimentů. Během naší spolupráce jsme sice narazili na nějaké překáţky, ale vše jsme vţdy vyřešili - dá se říci s úsměvem. Děkuji Vám. vii 1 ÚVOD 1.1 Čeleď Leguminosae Taxonomie této čeledi je dosud ne zcela jednotná a existující tři alternativní moţnosti zařazení: 1) Leguminosae je chápána jako čeleď a Mimosoideae a Caesalpinioideae jako podčeledi, nebo 2) Leguminosae = Fabaceae, zahrnující tři podčeledi: Faboideae (= Papilionoidae), Mimosoideae a Caesalpinioieae a nebo 3) Fabaceae (= Papilionaceae) je chápána jako čeleď stejně jako Mimosaceae a Caesalpiniaceae (Lewis et al. 2005). Čeleď Leguminosae zahrnuje byliny a dřeviny, s výjimkou rostlin adaptovaných na vodní prostředí. Tato čeleď má hned několik zajímavostí. Rostliny patřící do Leguminosae (Fabaceae) rostou na půdách ochuzených o dusík, proto na svých kořenech mají speciální útvary - hlízky, které jim vyrůstají po infekci nitrogenními bakteriemi rodu Rhizobium. Jedná se o symbiózu, kde bakterie metabolizují atmosférický dusík na amonný iont, který vyuţívá hostitelská rostlina, za úplatek v podobě jednoduchých cukrů, které si naopak bakterie berou od rostliny. Rostliny čeledi Leguminosae mají listy střídavé, nejčastěji sloţené, jednoduše zpeřené nebo dlanitě sloţené, vzácněji jednoduché, palistnaté, výjimečně palisty chybějí. Květenství je hroznovité (rod Vicia), méně často jsou květy jednotlivě (rody Lathyrus, vzácněji Vicia nebo Pisum). Květy jsou oboupohlavné, souměrné, kalich srůstá z pěti lístků, korunu tvoří pět lístků volných rozlišených na pavézu, křídla a člunek, kde pavéza je v rovině souměrnosti. V pupenu pavéza obaluje ostatní části květu (sestupná estivace). Dva sousedící lístky - křídla, a člunek srůstající ze dvou zbylých lístků. Tyčinek

This paper provides a brief overview of the history and the main achievements of archaeobotanical work in Greece to date, with the aim of highlighting its potential and creating a framework in which future work can be contextualised. The term ‘archaeobotany’ is used here in its narrow sense, referring to the study of plant macroremains, such as seeds, fruits and other plant parts, and excluding charcoal studies or ‘anthracology’ and analyses of microremains (e.g. pollen, phytoliths), which have developed to become separate sub-disciplines. From the first finds to a science Plant remains in the form of large concentrations of seeds, or individual finds of large specimens (known as spot finds), such as fruit stones, have been reported in the archaeological literature since the end of the nineteenth century. Botanical specimens that were occasionally unearthed caught the attention of archaeologists and site directors, who would either invite botanists or other experts to identify the species, or would simply rely on the expertise of the archaeological team, including that of local workers. A rather widely reported case is that of the early excavations at Knossos, where local workmen identified seeds found in a pithos as ‘Egyptian beans’, a variety of small fava beans imported to Crete from Alexandria at the end of the nineteenth century (Evans 1901, 20–21). ABSTRACT Lathyrus species are well placed to meet the increasing global demand for food and animal feed, at the time of climate change. Conservation and sustainable use of the genetic resources of Lathyrus is of significant importance to allow the regain of interest in Lathyrus species in world. A comprehensive global database of Lathyrus species originating from the Mediterranean Basin, Caucasus, Central and West Asia Regions is developed using accessions in major genebanks and information from eight herbaria in Europe. This Global Lathyrus database was used to conduct gap analysis to guide future collecting missions and in situ conservation efforts for 37 priority species. The results showed the highest concentration of Lathyrus priority species in the countries of the Fertile Crescent, France, Italy and Greece.

The paper shows the results of floristic investigations of 12 islands and several skerries of the inner part of SW Finnish archipelago, situated within a square of 11.56 km2. The research comprised all vascular plants – growing spontaneously and cultivated, and the results were compared to the present flora of a square 10 × 10 km from the Atlas of Vascular Plants of Finland, in which the studied area is nested. The total flora counted 611 species, among them, 535 growing spontaneously or escapees from cultivation, and 76 exclusively in cultivation. The results showed that the flora of Seili and adjacent islands was almost as rich in species as that recorded in the square 10 × 10 km. This study contributed 74 new species to this square. The hitherto published analyses from this area did not focus on origin (geographic-historical groups), socioecological groups, life forms and on the degree of threat of recorded species. Spontaneous flora of the studied area constituted about 44% of the whole flora of Regio aboënsis.

Nesmím opomenout i další pracovníky univerzity, s kterými jsem se potkávala v laboratoři a kteří mi vţdy ochotně pomáhali, kdyţ bylo potřeba. V roce 2014 jsem mohla díky finanční podpoře z projektu IGA_Prf_2014001 vycestovat do Maďarska na přirozenou lokalitu hrachu, coţ pro mě bylo velikým přínosem. The phylogenetic position of V. incisa is uncertain because various DNA markers have provided contradictory results. Isozymes of V. incisa encoded by 15 loci and resolved with the use of polyacrylamide gel electrophoresis (PAGE) are described and compared with those of seven related species belonging to sections Vicia, Sepium, Lathyroides and Pseudolathyrus in order to get new evidence about its taxonomic rank and phylogenetic position. Phylogenetic relationships are analyzed with maximum parsimony and neighbour joining methods. Vicia incisa is shown to differ from all three subspecies of V. sativa including, sativa, cordata and nigra, by alternate variants of ten isozymes out of 15 analysed. Instead, V. incisa has much more similarity to V. grandiflora and V. sepium by sharing eight isozyme variants which differ from the subspecies of V. sativa. The most parsimony and neighbour joining analyses of the isozyme variation placed V. incisa as basally linked to the V. grandiflora and V. sepium species couple in the clade of section Sepium (= sect.
