Molecular tools have become the reference even though they remain relatively time-consuming and expensive. We propose a new approach for the identification of cercariae using MALDI-TOF mass spectrometry. Snails of different genera (Radix, Lymnaea, Stagni- cola, Planorbis, and Anisus) were collected in the field to perform emitting tests in the laboratory. The cercariae they emitted (Trichobilharzia anseri, Diplostomum pseudospathaceum, Alaria alata, Echinostoma revolutum, Petasiger phalacrocoracis, Tylodelphys sp., Australapatemon sp., Cotylurus sp., Posthodiplostomum sp., Parastrigea sp., Echi- noparyphium sp. and Plagiorchis sp.) were characterized by sequencing the D2, ITS2 and ITS1 domains of rDNA, and by amplification using specific Alaria alata primers. A sample of each specimen, either fresh or stored in ethanol, was subjected to a simple preparation protocol for MALDI-TOF analysis. -

OCCURRENCE OF METACERCARIAE OF DIPLOSTOMUM AND TYLODELPHYS SPECIES (DIPLOSTOMIDAE) IN CLARIAS SPECIES (CLARIIDAE) FROM LAKE VICTORIA MJ Musiba and G Nkwengulila Department of Zoology and Marine Biology P. E-mail: [email protected] ABSTRACT The localisation and abundance of the metacercariae of Diplostomum and Tylodelphys species among the clariids of Lake Victoria were investigated between April and December 2001. Four species viz. Clarias gariepinus (271), C. alluaudi (65), C. werneri (50) and C. liocephalus (47) were examined, but only C. gariepinus was infected in the brain case and along the nasal cavity. Prevalence of metacercariae was high (>70%). Prevalence and mean intensity were similar between male and female fish. Mean intensity increased with fish size and declined in larger fish for D. mashonense; but declined with the increase in fish size for Tylodelphys species. The metacercariae were over-dispersed and the variance to mean ratio curves behaved similarly to mean intensity. Possible factors leading to the observed infection patterns are discussed. [ 1 ]. Members of the superfamilies Schistosomatoidea (Schistosomatidae, Aporocotylidae, and Spirorchiidae) and Diplostomoidea (Diplostomidae and Strigeidae) will be characterized. All these fl ukes have indirect life cycles with cercariae having ability to penetrate body surfaces of vertebrate intermediate or defi nitive hosts. In some cases, invasions of accidental (noncompat- ible) vertebrate hosts (including humans) are also reported. Their presence in a habitat will refect the presence of diferent host species and, as such, they can serve as ecological indicators. Only limited infor‑ mation on the occurrence of trematodes and their link to other trophic levels in the Danish freshwater ecosystems is currently available.Therefore, the main aim of the present study was to increase our knowledge in this feld. Methods: Snails were sampled from 21 freshwater lakes in Denmark, following which shedding procedures were performed, cercariae were recoved and the released parasites were identifed using molecular tools (PCR and sequencing). Results: A total of 5657 snail hosts belonging to ten species were identifed, revealing a highly diverse parasite fauna comprising 22 trematode species. The overall trematode prevalence was 12.6%, but large variations occurred between host species. The snail host Lymnaea stagnalis showed the highest prevalence and also exhibited the highest diversity, accounting for 47.6% of the species richness.
