Sicikeh-cikeh forest includes three forest areas, namely Adian Tinjoan Customary Forest, Adian Tinjoan Protection Forest, and Taman Wisata Alam Sicikeh-cikeh. Typical vegetation of this area is mountain forest with large diversity of plant species. Among the species, orchid is one of the potential plants found diversely in this location and has not yet been recorded. The exploration activities were conducted at Sicikeh-cikeh forest. Orchid exploration was conducted to collect living plants for ex situ conservation purpose. Orchid inventory, to record orchid diversity in this area, was based on plant collection by purposive random sampling. The results of the study recorded approximately 102 different species from 30 genera of orchids in this area. Typical epiphytic and terrestrial highland orchids were found and very common in Sumatra. Among the species found are endemic Sumatra, such as Coelogyne brachygyne, Coelogyne salmonicolor, Dendrobium kruiense, Epigeneium pulchellum, Thrixspermum gombakense, Corybas stenotribonos and Paphiopedilum tonsum. Interesting terrestrial orchids include Kuhlhasseltia javanica, Neuwiedia zollingeri var. javanica, Paphiopedilum tonsum, Phaius callosus, Corybas stenotribonos, Calanthe aurantiaca, Calanthe chrysoglossoides, and Calanthe pulchra.

Due absence of appropriate pollinator in our area the seed setting was absent or was very low. Artificial pollination (Hand pollination) in S. plicata was studied and seed setting was observed by self and cross pollination. Flowers of S. plicata were pollinated and subsequent capsule development was carefully monitored. Hand pollination of orchid flowers provides an opportunity to discuss floral morphology and associated reproductive biology. Capsules were harvested 60 days after the pollination prior to capsule dehiscence. In vitro asymbiotic germination was 87.3% when seeds were cultured on MS media. The effect of BA and NAA hormones were also observed on seed germination. Seedlings were healthy and stronger when cultured on MS media supplemented with hormones. PM medium with PGRs combination gave the maximum percentage response (93.34%) followed by PGRs supplemented MS (80.00%), VW (73.34%) and KC (73.34%) media. Minimum percentage of seed germination was observed on half strength PGRs free VW (26.67%) medium. Minimum time needed for initiation of germination in full strength PGRs supplemented PM (8.67 ± 0.36a weeks) medium, whereas, maximum time required (20.17 ± 0.38h weeks) in PGRs free half strength KC medium.

The pods obtained through selfing were harvested after 30 days and cultured in different media supplemented with individual and combinations of 6-benzylamino purine (BAP) and 1-naphthalene acetic acid (NAA) Seed germination was higher in MS medium (93%) and lower in Mitra medium (87%). The seedlings were grown well in the MS medium supplemented with 1.5 ppm BAP and 0.5 ppm NAA. The plantlets obtained in this medium were healthy and vigorous with 16.8 cm height at the time of deflasking for hardening. The matured rooted plantlets were acclimatized by using potting mixture at 1:1:1 proportion for hardening. The survived plantlets were transferred to field establishment and its growth was successful. The aim of this study was to reinvestigate the embryological characters using classical paraffin embedding and sectioning, along with ovule clearing and scanning electron microscopy analyses. The simultaneous cytokinesis results in decussate, isobilateral, linear, T-shaped and tetrahedral microspore tetrads. Mature pollen grains are two-celled with perforate sculpturing. The anther wall is comprised of an epidermis, two to three layers of endothecium, one layer each of middle layer and one layer of binucleate glandular tapetal cells. At anthesis, the mature anther wall is composed of epidermis and fibrous endothecium. At 9 d after pollination (DAP), the mature ovules are anatropous, bitegmic and tenuinucellate. The megaspore tetrad is linear. Development of the embryo sac conformed to the Polygonum type. The pollen tube enters the embryo sac through the micropyle (porogamy), and double fertilization is observed at 15 DAP. The zygote divides transversely to form a two-celled proembryo.
