As electron transfer (ET)-based total antioxidant capacity (TAC) assays generally measure the reducing ability of antioxidant compounds, thiols and phenols cannot be differentiated since they are both responsive to the probe reagent. In this study, three of the most common TAC determination methods, namely cupric ion reducing antioxidant capacity (CUPRAC), 2,20-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt/trolox equivalent antioxidant capacity (ABTS/TEAC), and ferric reducing antioxidant power (FRAP), were tested for the assay of phenolics in the presence of selected thiol and protein compounds. Although the FRAP method is almost non-responsive to thiol compounds individually, surprising overoxidations with large positive deviations from additivity were observed when using this method for (phenols + thiols) mixtures. Among the tested TAC methods, CUPRAC gave the most additive results for all studied (phenol + thiol) and (phenol + protein) mixtures with minimal relative error. -

The main objective of this review was to elucidate the reaction pathways that underlie the ABTS/potassium persulfate decolorization assay of antioxidant capacity. Comparative analysis of the literature data showed that there are two principal reaction pathways. Some antioxidants, at least of phenolic nature, + can form coupling adducts with ABTS• , whereas others can undergo oxidation without coupling, thus the coupling is a specific reaction for certain antioxidants. These coupling adducts can undergo further oxidative degradation, leading to hydrazindyilidene-like and/or imine-like adducts with 3-ethyl-2-oxo-1,3-benzothiazoline-6-sulfonate and 3-ethyl-2-imino-1,3-benzothiazoline-6-sulfonate as marker compounds, respectively. The extent to which the coupling reaction contributes to the total antioxidant capacity, as well as the specificity and relevance of oxidation products, requires further in-depth elucidation. -

Accepted 10 May, 2011 In the present study, the phylogenetic analysis of the Streptomyces strain Eri11 isolated from the rhizosphere of Rhizoma Curcumae Longae and the antioxidant activity of the broth cultured with Eri11 were investigated. Analysis of 16S rDNA gene sequences demonstrated that the strains Eri11 was most closely related to representatives of the genera Streptomyces. The total phenols and flavonoids contents in cultured broth were detected to be13.59 ± 0.17 mg gallic acid equivalent/g and 9.93 ± 0.83 mg rutin equivalent/g, respectively. The cultured broth showed the antioxidant activity against the ABTS (2, 2’-Azinobis-3-ethyl benzthiazoline-6-sulfonic acid) free radicals and hydroxyl free radicals with IC50 (The half-inhibitory concentration) of 223.81 ± 24.50 μg/ml and 582.42 ± 83.10 μg/ml respectively. So, it was suggested that the isolated Streptomyces strain Eri11 could be a candidate for the nature resource of the antioxidants. -

Bacteriostatic and bactericidal effects of free nitrous acid on model microbes in wastewater treatment Shuhong Gao Master of Science Harbin Institute of Technology, Harbin, China A thesis submitted for the degree of Doctor of Philosophy at The University of Queensland in 2016 School of Chemical Engineering Advanced Water Management Centre Abstract There is great potential to use free nitrous acid (FNA), the protonated form of nitrite (HNO2), as an antimicrobial agent due to its bacteriostatic and bactericidal effects on a range of microorganisms. However, the antimicrobial mechanism of FNA is largely unknown. The overall objective of this thesis is to elucidate the responses of two model bacteria, namely Psuedomonas aeruginosa PAO1 and Desulfovibrio vulgaris Hildenborough, in wastewater treatment in terms of microbial susceptibility, tolerance and resistance to FNA exposure. The effects of FNA on the opportunistic pathogen P. aeruginosa PAO1, a well-studied denitrifier capable of nitrate/nitrite reduction through anaerobic respiration, were determined. It was revealed that the antimicrobial effect of FNA is concentration-determined and population-specific. By applying different levels of FNA, it was seen that 0.1 to 0.2 mg N/L FNA exerted a temporary inhibitory effect on P. aeruginosa PAO1 growth, while complete respiratory growth inhibition was not detected until an FNA concentration of 1.0 mg N/L was applied. The FNA concentration of 5.0 mg N/L caused complete cell killing and likely cell lysis. Differential killing by FNA in the P. aeruginosa PAO1 subpopulations was detected, suggesting intra-strain heterogeneity. A delayed recovery from FNA treatment suggested that FNA caused cell damage which required repair prior to P. -

They are a source of small reactive species, including reactive nitrogen species (RNS). Wound healing is a major application and, as its physiology involves RNS signaling, a correlation between clinical effectiveness and the activity of plasma-derived RNS seems evident. To investigate the type and reactivity of plasma-derived RNS in aqueous systems, a model with tyrosine as a tracer was utilized. By high-resolution mass spectrometry, 26 different tyrosine derivatives including the physiologic nitrotyrosine were identified. The product pattern was distinctive in terms of plasma parameters, especially gas phase composition. By scavenger experiments and isotopic labelling, gaseous nitric dioxide radicals and liquid phase peroxynitrite ions were determined as dominant RNS. -

It can react with metal centers and oxidized thiol products such as disulfides (RSSR) and sulfenic acids (RSOH). Reactions with oxidized thiol products form persulfides (RSSH/RSS–). Persulfides have been proposed to transduce the signaling effects of H2S through the modification of critical cysteines. They are more nucleophilic and acidic than thiols and, contrary to thiols, also possess electrophilic character. In this review, we summarize the biochemistry of hydrogen sulfide and persulfides, focusing on redox aspects. We describe biologically relevant one- and two-electron oxidants and their reactions with H2S and persulfides, as well as the fates of the oxidation products. Box 12065, The Hebrew University of Jerusalem, Jerusalem 91120, Israel Sicilian cultivars of prickly pear (Opuntia ficus indica) produce yellow, red, and white fruits, due to the combination of two betalain pigments, the purple-red betanin and the yellow-orange indicaxanthin. The betalain distribution in the three cultivars and the antioxidant activities of methanolic extracts from edible pulp were investigated. In addition, the reducing capacity of purified betanin and indicaxanthin was measured. According to a spectrophotometric analysis, the yellow cultivar exhibited the highest amount of betalains, followed by the red and white ones. Indicaxanthin accounted for about 99% of betalains in the white fruit, while the ratio of betanin to indicaxanthin varied from 1:8 (w:w) in the yellow fruit to 2:1 (w:w) in the red one. Polyphenol pigments were negligible components only in the red fruit. When measured as 6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylic acid (Trolox) equivalents per gram of pulp, the methanolic fruit extracts showed a marked antioxidant activity.
