E-mail: [email protected] ABSTRACT. Glycoalkaloids are important secondary metabolites accumulated by plants as protection against pathogens. One of them, α-tomatine, is found in high concentrations in green tomato fruits, while in the ripe fruits, its aglycone form, tomatidine, does not present a protective effect, and it is usual to find parasites of tomatoes like Phytomonas serpens in these ripe fruits. To investigate the sensitivity of trypanosomatids to the action of α-tomatine, we used logarithmic growth phase culture of 20 trypanosomatids from insects and plants and Trypanosoma cruzi. The lethal dose 50% (LD50) was determined by mixing 107 cells of the different isolates with α-tomatine at concentrations ranging from 10-3 to 10-8 M for 30 min at room temperature. The same tests performed with the tomatidine as a control showed no detectable toxicity against the same trypanosomatid cultures. The tests involved determination of the percentage (%) survival of the protozoan cultures in a Neubauer chamber using optical microscopy. -

Levin Food Safety and Health Research Unit, Western Regional Research Center, Agricultural Research Service, U.S. Department of Agriculture, 800 Buchanan Street, Albany, California 94710 Tomato plants (Lycopersicon esculentum) synthesize the glycoalkaloid a-tomatine, possibly as a defense against insects and other pests. As part of an effort to improve the safety of plant foods, the usefulness of a new HPLC pulsed amperometric detection (PAD) method for the direct analysis of a-tomatine in different parts of the tomato plant; in store-bought and field-grown, including transgenic, tomatoes; in a variety of commercial and home-processed tomato products; and in eggplant and tomatillos was evaluated. The method was found to be useful for analysis of a variety of products including high-tomatine calyxes, flowers, leaves, roots, and stems of the tomato plant (14-130 mg/100 g of fresh weight), low-tomatine red tomatoes (0.03-0.08 mg/100 g), intermediate- tomatine tomatoes (0.1-0.8 mg/100 g), and high-tomatine fresh and processed green, including pickled and fried, tomatoes (0.9-55 mg/100 g). No experimental difficulties were encountered with extraction and analysis of tomatine in complex foods such as tomato juice, ketchup, salsa, sauce, and sun-dried tomatoes. Microwaving and frying did not significantly affect tomatine levels of tomato foods. The tomatine content of fresh market and transgenic delayed-ripening varieties was not different from the range ordinarily seen in tomato. -

The glycoalkaloid tomatine, derived from the wild tomato, can act as a powerful adjuvant to elicit an antigen-specific cell-mediated immune response to the circumsporozoite (CS) protein, a major pre-erythrocytic stage malaria vaccine candidate antigen. Using a defined MHC-class-I-restricted CS epitope in a Plasmodium berghei rodent model, antigen-specific cytotoxic T lymphocyte activity and IFN-γ secretion ex vivo were both significantly enhanced compared to responses detected from similarly stimulated splenocytes from naive and tomatine-saline-immunized mice. Further, through lymphocyte depletion it is demonstrated that antigen-specific IFN-γ is produced exclusively by the CD8+ T cell subset. -

(May 2008) Sherif Mohamed Hassan, B.S.; M.S., Suez Canal University Chair of Advisory Committee: Dr. Aubrey Lee Cartwright Three saponin-rich extracts (20, 60, 100% methanol), four 100% methanol sub- fractions and seven independently acquired fractions (A-G) from guar meal, Cyamopsis tetragonoloba L. (syn. C. psoraloides), were evaluated for antimicrobial and hemolytic activities. These activities were compared against quillaja bark (Quillaja saponaria), yucca (Yucca schidigera), and soybean (Glycine max) saponins in 96-well plates using eight concentrations (0.01 to 1.0 and 0.1 to 12.5 mg extract/mL). Initial guar meal butanol extract was 4.8 ± 0.6% of the weight of original material dry matter (DM). Butanol extract was purified by preparative reverse-phase C-18 chromatography. Two fractions eluted with 20, and one each with 60, and 100% methanol with average yields of 1.72 ± 0.47, 0.88 ± 0.16, 0.91 ± 0.16 and 1.55 ± 0.15% of DM, respectively. -

Japan 60: 288-294 (1994) Induction of Tomatine in Tomato Plant by an Avirulent Strain of Pseudomonas solanacearum Triwidodo ARWIYANTO*,**, Kanzo SAKATA***, Masao GOTO***, Shinji TSUYUMU*** and Yuichi TAKIKAWA*** Abstract Tomato plants inoculated with an avirulent strain of Pseudomonas solanacearum (strain Str-10 op type) produced a growth inhibitory substance at a higher level than those of uninoculated control in root system but not in stem. The substance was identified to be tomatine by Mass Spectrometry, TLC, UV spectrometry and chemical component analysis. The tomatine contents five days after inoculation were 113 and 152ƒÊg/g fresh root when Str-10 op type was inoculated at the inoculum concentrations of 108 and 109cfu/ml, respectively. Tomatine content in the uninoculated control plants was 65.9ƒÊg/g. Tomatine content nine days after inoculation reached 450ƒÊg/g. In an in vitro experiment, 100ƒÊg/ disk of authentic tomatine and approximately 150ƒÊg/disk of extracted tomatine were sufficient for growth inhibition of P. solanacearum. Both extracted and authentic tomatine exhibited bacteriostatic activity against P. solanacearum. Treatment with heat-killed cells or culture filtrate of Str-10 op type failed to increase tomatine content in the root tissue. Our current knowledge gained from the chemistry and analysis of these compounds in fresh and processed tomatoes and from studies on their bioavailability, bioactivity, and mechanisms of action against cancer cells and other beneficial bioactivities including antibiotic, anti-inflammatory, antioxidative, cardiovascular, and immunostimulating effects in cells, animals, and humans is discussed and interpreted here. Areas for future research are also suggested. The collated information and suggested research might contribute to a better understanding of the agronomical, biochemical, chemical, physiological, molecular, and cellular bases of the health-promoting effects and facilitate and guide further studies needed to optimize the use of lycopene and α-tomatine in pure form and in fresh tomatoes and processed tomato products to help prevent or treat human disease.
