Identification of Apocynaceae Climbers in the Sangha Trinational, Central Africa Identification of Apocynaceae climbers in the Sangha Trinational, Central Africa Melanie Bayo August 2020 Thesis submitted in partial fulfilment for the MSc in the Biodiversity and Taxonomy of Plants 2 Abstract Apocynaceae climbers are easy to distinguish from other plant families due to the white latex and the opposite leaves. Identification at lower taxon levels, however, is more difficult as fertile material is rarely available in the field. This study aimed to facilitate the identification of Apoc- ynaceae climbers in the Sangha Trinational, Central Africa. For this, a morphological study on 42 species was conducted by examining digital specimens from across their distribution. An identification key to the species based on sterile characters is presented. Results showed that Apocynaceae climbers can be grouped based on sterile traits, however, for a few species, de- limitation without fertile characters has proved to be difficult. The key is the first of that kind to be produced for central African Apocynaceae climbers, yet it needs to be subject to further revision and testing. Secondly, this study aimed to assess the utility of rbcL as barcode marker for plants of the Sangha Trinational. For this, 87 rbcL sequences of 50 species in 28 families were subjected to two BLAST searches. Results showed that rbcL performs well at discriminating families (97 % identification matches) and is satisfactory at genus-level (77 %). Potentials and perils of DNA barcoding for identification are discussed. Under good management, these forests will continue to supply mankind with products long after mined resources are exhausted. The vast treeless areas elsewhere in Africa give added emphasis to the economic significance of the forests of Liberia and its neighboring countries in West Africa. The mature forests of Liberia are composed entirely of broadleaf or hardwood tree species. These forests probably covered more than 90 percent of the country in the past, but only about one-third is now covered with them. Another one-third is covered with young forests or reproduction referred to as low bush. The mature, or "high," forests are typical of tropical evergreen or rain forests where rainfall exceeds 60 inches per year without pro­ longed dry periods. Certain species of trees in these forests, such as the cotton tree, are deciduous even when growing in the coastal area of heaviest rainfall, which averages about 190 inches per year. Endress2 (APOCYNACEAE) BASED ON MOLECULAR AND MORPHOLOGICAL EVIDENCE1 ABSTRACT To elucidate deeper relationships within Rauvolfioideae (Apocynaceae), a phylogenetic analysis was conducted using sequences from five DNA regions of the chloroplast genome (matK, rbcL, rpl16 intron, rps16 intron, and 39 trnK intron), as well as morphology. Bayesian and parsimony analyses were performed on sequences from 50 taxa of Rauvolfioideae and 16 taxa from Apocynoideae. Neither subfamily is monophyletic, Rauvolfioideae because it is a grade and Apocynoideae because the subfamilies Periplocoideae, Secamonoideae, and Asclepiadoideae nest within it. In addition, three of the nine currently recognized tribes of Rauvolfioideae (Alstonieae, Melodineae, and Vinceae) are polyphyletic. We discuss morphological characters and identify pervasive homoplasy, particularly among fruit and seed characters previously used to delimit tribes in Rauvolfioideae, as the major source of incongruence between traditional classifications and our phylogenetic results. Based on our phylogeny, simple style-heads, syncarpous ovaries, indehiscent fruits, and winged seeds have evolved in parallel numerous times. A revised classification is offered for the subfamily, its tribes, and inclusive genera.
