  • Molecular Identification of Commercialized Medicinal Plants in Southern Morocco
Molecular Identification of Commercialized Medicinal Plants in Southern Morocco Anneleen Kool1*., Hugo J. de Boer1.,A˚ sa Kru¨ ger2, Anders Rydberg1, Abdelaziz Abbad3, Lars Bjo¨ rk1, Gary Martin4 1 Department of Systematic Biology, Evolutionary Biology Centre, Uppsala University, Uppsala, Sweden, 2 Department of Botany, Stockholm University, Stockholm, Sweden, 3 Laboratory of Biotechnology, Protection and Valorisation of Plant Resources, Faculty of Science Semlalia, Cadi Ayyad University, Marrakech, Morocco, 4 Global Diversity Foundation, Dar Ylane, Marrakech, Morocco Abstract Background: Medicinal plant trade is important for local livelihoods. However, many medicinal plants are difficult to identify when they are sold as roots, powders or bark. DNA barcoding involves using a short, agreed-upon region of a genome as a unique identifier for species– ideally, as a global standard. Research Question: What is the functionality, efficacy and accuracy of the use of barcoding for identifying root material, using medicinal plant roots sold by herbalists in Marrakech, Morocco, as a test dataset. Methodology: In total, 111 root samples were sequenced for four proposed barcode regions rpoC1, psbA-trnH, matK and ITS. Sequences were searched against a tailored reference database of Moroccan medicinal plants and their closest relatives using BLAST and Blastclust, and through inference of RAxML phylograms of the aligned market and reference samples. Principal Findings: Sequencing success was high for rpoC1, psbA-trnH, and ITS, but low for matK. Searches using rpoC1 alone resulted in a number of ambiguous identifications, indicating insufficient DNA variation for accurate species-level identification. Combining rpoC1, psbA-trnH and ITS allowed the majority of the market samples to be identified to genus level.

CURRICULUM VITAE – ORIANE HIDALGO Research line: The connecting thread of my research is to uncover patterns and processes of genome evolution responsible for plant diversification, through an integrated approach including evolutionary-developmental biology, phylogenetics and cytogenetics. I am particularly interested in studying the origin and diversification of plant reproductive morphologies across angiosperms from the evo-devo and genomic perspectives. My research concentrates on the changes in floral symmetry and inflorescence complexity in Papaveraceae and the Asteraceae representatives displaying secondary heads (=syncephalia). My other main research line focuses on the evolutionary significance of the extraordinary diversity of plant genomes (i.e. size, organisation, composition and dynamics), covering a wide array of plant groups including angiosperms, gymnosperms and pteridophytes. By Wilhelm (Bill) Bischoff / member BC Council of Garden Clubs If you can accept that the island called Santorini in the central Mediterranean, also known as Thira / Tera, is the original Island of Atlantis; if you also can agree that this Island had a terrific volcanic explosion more than 3,000 years ago, than I can share with you an equally fantastic botanical story with you. That today’s Thira is the remnant of an exploded volcano is quite evident when one looks at a map of this region of the Mediterranean. Located as part of the Aegean Islands, just north of Crete, it shows the unmistakable shape of a water filled volcanic caldera with a center-cone island. Scientists have identified volcanic ash taken from the bottom of the Mediterranean Sea, close to the Lebanese coast, as originating from Thira. The time frame of some 3300 years ago also coincides with the beginning of a rather tumultuous time in this part of the ancient world, the end of the “Bronze Age”. The possible cause of that could well have been a natural disaster, in the very heart of the ancient world as we know it. Now that I have your attention and possibly have whetted your curiosity, let me introduce you to one of the small wonders of this very ancient world, the beautiful Cyclamen, all 22 species of them.

Khafagi(3), Nashua A.M. Mostafa(1) (1)Department of Botany and Microbiology, Faculty of Science, El-Arish University, N. Sinai, Egypt; (2)The Herbarium, Botany Department, Faculty of Science, Cairo University, Giza, Egypt; (3)Botany Department, Faculty of Science, Al-Azhar University (Girls Branch), Cairo, Egypt. ANUALLY constructed keys for identification of plants leave much to be desired. Keys Mto the Asteraceae of Egypt are no exception and depend largely on floral minutiae while vegetative morphology is a much richer source of characters suitable for key construction. Inspection of some 3000 specimens showed that the most obvious feature of the plants is the presence or absence of spines on leaves, leaf axils, stem internodes, margins of stem wings and phyllaries. This feature was selected to divide species of this family into two main groups: spiny and spineless. Nomenclature of all taxa was updated and those with names reduced to synonyms of others were eliminated. This article deals only with the 65 species belonging to 20 genera of the first group. A total of 51 characters describing variation in spine distribution and other characters of vegetative morphology were recorded for each of the 65 spiny species and the key-generating program DELTA was applied to the data matrix. The result is a much improved automated key, a detailed description of every species in terms of the entire set of 51 characters, and the same description but in terms of the serial numbers assigned to these characters and their states.

  • New Perspectives of Purple Starthistle (Centaurea Calcitrapa) Leaf Extracts
Dimkić et al. AMB Expr (2020) 10:183 https://doi.org/10.1186/s13568-020-01120-5 ORIGINAL ARTICLE Open Access New perspectives of purple starthistle (Centaurea calcitrapa) leaf extracts: phytochemical analysis, cytotoxicity and antimicrobial activity Ivica Dimkić1*† , Marija Petrović1†, Milan Gavrilović1, Uroš Gašić2, Petar Ristivojević3, Slaviša Stanković1 and Peđa Janaćković1 Abstract Ethnobotanical and ethnopharmacological studies of many Centaurea species indicated their potential in folk medi- cine so far. However, investigations of diferent Centaurea calcitrapa L. extracts in terms of cytotoxicity and antimi- crobial activity against phytopathogens are generally scarce. The phenolic profle and broad antimicrobial activity (especially towards bacterial phytopathogens) of methanol (MeOH), 70% ethanol (EtOH), ethyl-acetate (EtOAc), 50% acetone (Me2CO) and dichloromethane: methanol (DCM: MeOH, 1: 1) extracts of C. calcitrapa leaves and their poten- tial toxicity on MRC-5 cell line were investigated for the frst time. A total of 55 phenolic compounds were identifed: 30 phenolic acids and their derivatives, 25 favonoid glycosides and aglycones. This is also the frst report of the pres- ence of centaureidin, jaceidin, kaempferide, nepetin, favonoid glycosides, phenolic acids and their esters in C. calci- trapa extracts. The best results were obtained with EtOAc extract with lowest MIC values expressed in µg/mL ranging from 13 to 25, while methicillin resistant Staphylococcus aureus was the most susceptible strain. The most susceptible phytopathogens were Pseudomonas syringae pv. syringae, Xanthomonas campestris pv. campestris and Agrobacte- rium tumefaciens. The highest cytotoxicity was recorded for EtOAc and Me2CO extracts with the lowest relative and absolute IC50 values between 88 and 102 µg/mL, while EtOH extract was the least toxic with predicted relative IC 50 value of 1578 µg/mL.
