Authenticity of the hair is established by optic and electronic microscopy. Sequences of hypervariable regions of the mitochondrial DNA (extracted from two different hairs) show five mutations: 16298C; 72C, 152C, 195C and 263G; the corresponding mtDNA haplogroup is the sub-haplogroup HVO. Consequently, Naundorff cannot be excluded to be considered as being Louis XVII on the basis of mtDNA sequence of his humerus (as affirmed in Jehaes et al., 1998). Comparisons of mtDNA sequences between mtDNA extracted from Naundorff’s hair reported here and those (Jehaes et al., 2001) of the living Anna of Roumania (of Habsburg descent) shows that both correspond to mtDNA haplogroups of the general HV cluster. This review will discuss the nature of mitochondrial DNA (mtDNA) and its levels in the cell, which are the key elements to consider when trying to achieve molecular identification in ancient and degraded samples. mtDNA sequence analysis has been appropriately validated and is a consistent molecular target for the examination of biological evidence encountered in forensic cases—and profiling, in certain conditions—especially for burnt bodies and degraded samples of all types. Exceptional cases and samples will be discussed in this review, such as mtDNA from leather in Beethoven’s grand piano, mtDNA in mummies, and solving famous historical criminal cases. In addition, this review will be discussing the use of ancient mtDNA to understand past human diet, to trace historical civilizations and ancient trade routes, and to uncover geographical domestication origins and lineage relationships.

His genomic DNA was extracted from his authentic hairs, that were studied by optic and electronic microscopy. Louis XVI’s Y-STRs profile is very similar to those of three living Bourbons previously published , differing from them by three Y-STRs allele values only. Dating estimates of the divergence time of the common ancestor (Louis XIII) between Louis XVI and the living Bourbons correspond to the observed genealogical time. K.W.
