While the collection techniques were demonstrated and discussed last year in Madison, this year the seminars will be focused on the laboratory procedures following the collection of embryos and oocytes. The ‘in vivo’ seminar will cover every aspect from the time the embryos/collection fluids arrive at the lab to the time when they are put in the freezer. The ‘in vitro’ seminar will cover every aspect from the time the collection tube arrives at the lab to the time oocytes are put in the incubator to mature. And the third seminar will cover ‘embryo cryopreservation’. The seminars will include presentation and demonstration. The goal of these seminars is to give participants an opportunity to be introduced or to review techniques and exchange tips with the instructors and participants. Participants will be divided into three (3) groups and will rotate after 50 minutes. The number of places is limited, first come, first served!

SESSIONS: Using blood tests to better manage donors 8:00 AM – 9:00 AM The Anti-Mullerian Hormone (AMH) and embryo production in the bovine species: where are we at? Presented by: Dr. Charlene Rico The Anti-Mullerian hormone (AMH) is currently used in Human as a good marker of the ovarian reserve. Recently, it has been shown that in the adult bovine, the endocrine AMH is also a marker of the folliculogenesis activity and a good predictive tool of the ovarian response to a stimulatory treatment. This presentation will focus on the AMH production by granulosa cells, its regulation by different factors, in order to understand how this hormone could help in practice to predict ovarian response in bovine donors and its limits.

Presented by: Dr. Elodie Kara Synchronisation and insemination in due time in superovulation protocols are important for the number and quality of collected viable embryos. We demonstrate here that LH and progesterone are key factors to be monitored in MOET protocols for donors as well as for recipients.

BREAKOUT SESSION: Equine Embryo Transfer 3:30 PM – 5:00 PM Equine Embryo Transfer: Why do horses have to be different? Presented by: Dr. Tracey Chenier Doing bovine ET and wondering about adding equine services? Do you have lots of questions about how different horse embryos really are? This seminar will cover the general aspects of equine embryo transfer for those new to the technique, including donor and recipient management, morphology of the equine embryo, embryo flushing techniques, and transfer. Recent developments in embryo cryopreservation and biopsy will be briefly covered.

SESSIONS: Insemination, fertilization and sexed semen 3:30 PM – 4:30 PM A novel insemination system for superovulated beef donors that increases the percent of total ova collected that are viable embryos. Presented by: Dr. Brad Stroud Roughly 45% or more of ova collected from superovulated beef donors are either unfertilized or degenerate. We have looked at many methods to some of that population to the viable side. The novel Maximate™ insemination system provides an alternative approach using a high volume diluent. When mixed with thawed semen, this method immediately floods the uterus with sperm thereby by-passing the physiological sperm transport mechanisms of the cow. This presentation will reveal the results of Maximate™ inseminated beef donors on over 21,000 total ova collected over a three year period compared to 18,000 ova collected from donors inseminated by traditional AI methods (controls) during the same period of time.

4:30 PM – 5:00 PM Reverse Sorted, Sexed Frozen, or Conventional - Effects of Semen Type on IVF Outcomes Presented by: Dr. Shantille Kruse Emerging technologies such as the use of sexed frozen and reverse sorted semen in IVF offer more specific options to breeders. However, each semen type has specific challenges associated with using it. Here, we’ll discuss the benefits and drawbacks of each semen type and share real-world IVF outcomes.

One must understand how the endocrine system regulates the functions of the reproductive tract in order to successfully manipulate the donor cows to develop additional oocytes in a reproductive cycle as well as to prepare the recipient cows for implantation of an embryo. This session will provide a general overview of reproductive physiology and endocrinology as it pertains to embryo transfer.

SESSIONS: The early embryo – evaluation, metabolism and imprinting 10:30 AM - 11:00 AM IVP vs IVD embryo evaluation Presented by: Dr. Jennifer Barfield This presentation will explore variations in appearance and quality between in vivo derived and in vitro produced bovine embryos. This information will be presented in the context of grading embryos using IETS standards and impacts on cryopreservation success and pregnancy rates.

11:00 AM – 12:00 PM Fatty acids in relation to oocyte and embryo quality: not that straight forward! Presented by: Dr. Jo Leroy An altered maternal metabolic condition or a fatty acid rich diet can lead to a changed micro- environment of the maturing oocyte or developing embryo. Especially a higher load of fatty acids may directly alter the oocyte’s metabolism resulting in a different embryo physiology. Also a long term exposure of the developing pre-anthral follicle to elevated fatty acid conditions can have long lasting effects. But research results tend to vary and it remains a challenge to come up with clear cut recommendations.

SESSIONS: The early embryo – evaluation, metabolism and imprinting CONTINUED... 1:00 PM – 2:00 PM Impact of assisted reproductive technologies on genomic imprinting Presented by: Dr. Mellissa Mann This talk will define epigenetics and genomic imprinting, describe DNA methylation changes that occur normally during gametogenesis and early embryogenesis and discuss the effects of assisted reproductive technologies on genomic imprinting during early mouse and human development.

SESSIONS: Recipient management – optimizing offspring quality 2:00 PM – 3:00 PM Nutrition-reproduction interactions and developmental programming in recipient beef cattle Presented by: Dr. Kara Stewart Embryo transfer is desirable because it allows for multiple calves to be created from superior genetics in a short generation interval. Management of the recipient females goes beyond successful implantation of the transferred embryo and birthing of a viable calf. Today, Kara will discuss how the nutrition of the recipient beef cow during gestation and lactation can impact the reproductive potential of the developing offspring.

This challenging and breathtaking course features thick stands of deciduous trees, wetland hazards, knee-high fescue rough, and impeccable conditioning from tee to green. These all combine to make Ussher’s Creek a classic parkland course. Thomas McBroom set the course softly on the land with flowing lines, moderately sized greens with slight folds, and strategic bunkering to create targets and intricate turns. First, you'll ride an elevator down 70 metres, then walk along a 73 metre tunnel to the boardwalk--305 metres (1,000 feet or ¼ mile) with stairs that lead to two observation areas at the edge of the river. As the water travels through the Whirlpool Rapids at White Water Walk, it is traveling at about 48 km/hr or 30 mph, creating the Class 6 Whirlpool Rapids. Dr. Paolete Soto Dr. Soto is the CEO and senior embryologist at Repro

Solutions. She received her MSc (University of Florida) and PhD (University of Montreal) degrees in animal reproduction and embryology. Followed by a post- doctorate in reproductive physiology at the McGill University. Dr. Soto research has focused on improving the development of early embryo. Dr. Soto has a great deal of experience with in vitro fertilization, Ovum Pick Up and general reproductive management. She is directly involved in carrying out the services provided by Repro Solutions.

------Dr. Kara Stewart Kara earned her bachelor’s degree from Purdue University and her master’s and doctoral degrees in Animal Sciences from North Carolina State University. Kara currently is an Assistant Professor in the Animal Science department at

Purdue University with a teaching and extension position in reproductive physiology. Her research interests include methods to improve reproductive outcomes in swine and beef cattle. Specifically for beef cattle, she explores variations in the dam’s nutrition during gestation and lactation on the reproductive performance of her offspring. Currently, she is investigating the impacts of feeding high levels of dried distiller’s grains during various stages of gestation and lactation on heifer and bull reproductive development as well as the re-breeding capacity of the dam.
