Losses due to pneumonia and associated expenses are estimated to be higher in Ethiopia with limited information about the distribution of capsular serotypes. Hence, this study was designed to determine the phenotypic and capsular serotypes of P. multocida from pneumonic cases of cattle. Methods: A cross sectional study with purposive sampling method was employed in 400 cattle from April 2018 to January 2019. Nasopharyngeal swabs and lung tissue samples were collected from clinically suspected pneumonic cases of calves (n = 170) and adult cattle (n = 230). Samples were analyzed using bacteriological and molecular assay. Results: Bacteriological analysis revealed isolation of 61 (15.25%) P. multocida subspecies multocida. Incidence was higher in calves 35 (57.38%) compared to adult cattle 26 (42.62%) at P < 0.5. PCR assay targeting KMT1 gene (~460 bp) conrmed P. -

Gupta L8N 3Z5, Canada [email protected] Understanding of the phylogeny and interrelationships of the genera within the order ‘Enterobacteriales’ has proven difficult using the 16S rRNA gene and other single-gene or limited multi-gene approaches. In this work, we have completed comprehensive comparative genomic analyses of the members of the order ‘Enterobacteriales’ which includes phylogenetic reconstructions based on 1548 core proteins, 53 ribosomal proteins and four multilocus sequence analysis proteins, as well as examining the overall genome similarity amongst the members of this order. The results of these analyses all support the existence of seven distinct monophyletic groups of genera within the order ‘Enterobacteriales’. In parallel, our analyses of protein sequences from the ‘Enterobacteriales’ genomes have identified numerous molecular characteristics in the forms of conserved signature insertions/deletions, which are specifically shared by the members of the identified clades and independently support their monophyly and distinctness. Many of these groupings, either in part or in whole, have been recognized in previous evolutionary studies, but have not been consistently resolved as monophyletic entities in 16S rRNA gene trees. The work presented here represents the first comprehensive, genome- scale taxonomic analysis of the entirety of the order ‘Enterobacteriales’. -

Pasteurellosis in backyard poultry and other birds Shane R. Raidal1 Case 1 An investigation was requested to determine the cause of sudden mortalities in a group of 3 week old goslings bought from a local market. Within 4 days of purchase 2 of 12 birds were found very weak. Both died within 12 hours. One comatose and 2 dead goslings were submitted for necropsy examination. They were in good physical condition. Haematology results on blood collected from the live bird demonstrated a marked leucocytosis (37.3 × 109 cells/mL) due to a heterophilia (30.4 × 109 heterophils/mL) with a left shift. Necropsy examination demonstrated multifocal abscess throughout the lungs, mild hepatomegaly and a mild thickening of the airsacs. Histological examination demonstrated a severe multifocal necrosuppurative airsacculitis and pneumonia with pulmonary abscessation. A heavy growth of Pasteurella multocida was cultured from the lungs and liver. Further cases did not occur in remaining birds which were treated with doxycycline (5 g/L drinking water). Case 2 A backyard poultry flock of 29 Rhode Island red chickens consisting of 27 hens and 2 roosters all 1 year of age with a 3 month history of illness and deaths was investigated. Affected birds reportedly developed swollen eyes, lethargy, and inappetance and death within 2 days of the onset of clinical signs. One affected hen was submitted for necropsy. Haematology results were normal. Necropsy demonstrated pale, swollen periorbital sinuses, nares and beak and a caseous exudate from the choana. The larynx, trachea, lungs, airsacs and other visceral organs appeared normal. A moderate burden of Heterakis gallinarum was present in the caecae. -

This disease is one of an economically important disease in Ethiopia. Losses due to mortality and associated expenses are estimated to be higher in the country. Studies revealed that limited information is available regarding the capsular types, genotypes, and antimicrobial sensitivity of P. multocida isolates circulating in the country. This suggests, further molecular advances to understand the etiological diversity of the pathogens representing severe threats to the cattle population. Results: Bacteriological analysis of nasopharyngeal swab and pneumonic lung tissue samples collected from a total of 400 samples revealed isolation of 61 (15.25%) P. multocida subspecies multocida. 35 (20.59%) were isolated from calves and 26 (11.30%) from adult cattle. Molecular analysis using PCR assay targeting KMT1 gene (~460 bp) amplication was shown in all presumptive isolates. Capsular typing also conrmed the presence of serogroup A (hyaD-hyaC) gene (~1044 bp) and serogroup D (dcbF) gene (~657 bp) from 56 (91.80%) and 5 (8.20%) isolates, respectively.
