Dr. Abdullah KAYA Haziran, 2014, 129 Sayfa Bu çalışma Yavuzeli ve Şehitkâmil (Gaziantep) yörelerinde yetişen makromantarlar üzerinde gerçekleştirilmiştir. Bölgede periyodik olarak gerçekleştirilen arazi çalışmaları sürecinde 298 makromantar örneği toplanarak, laboratuvar ortamına taşınmış ve fungaryum materyali haline getirilmiştir. Gerekli tür tanı işlemleri sonucunda Ascomycota ve Basidiomycota bölümleri içinde yer alan 4 sınıf, 11 takım, 35 familya ve 57 cinse ait 73 takson tanımlanmıştır. Araştırma bölgesinde Leotiomycetes sınıfı 2 (Helotiaceae 1, Rhytismataceae 1), Pezizomycetes sınıfı 8 (Ascobolaceae 1, Helvellaceae 3, Morchellaceae 1, Pezizaceae 1, Pyronemataceae 1, Valsaceae 1), Agaricomycetes sınıfı 63 (Agaricaceae 8, Auriculariaceae 1, Bolbitiaceae 2, Cyphellaceae 1, Diplocystidiaceae 1, Ganodermataceae 2, Gomphidiaceae 1, Hygrophoraceae 1, Hymenochaetaceae 2, Inocybaceae 2, Marasmiaceae 3, Meruliaceae 2, Mycenaceae 2, Niaceae 2, Omphalotaceae 4, Physalacriaceae 1, Pleurotaceae 1, Pluteaceae 2, Polyporaceae 4, Psathyrellaceae 8, Rhizopogonaceae 1, Schizophyllaceae 1, Stereaceae 1, Strophariaceae 5, Suillaceae 1, Tricholomataceae 5), Dacrymycetes sınıfı (Dacrymycetaceae 1) ise 1 taksonla temsil edilmektedir. Prof. Dr. Abdullah KAYA June, 2014, 129 pages This study was carried out on macofungi growing in Yavuzeli and Şehitkâmil (Gaziantep) districts. During periodical field trips, 298 macrofungi samples were collected and transferred to the laboratory, and prepared as fungarium material. As a result of necessary identification processes, 73 taxa belonging to 4 classes, 35 families and 57 genera within the divisions Ascomycota and Basidiomycota were identified. In research area Leotiomycetes is represented by 2 taxa (Helotiaceae 1, Rhytismataceae 1), Pezizomycetes is represented by 8 taxa (Ascobolaceae 1, Helvellaceae 3, Morchellaceae 1, Pezizaceae 1, Pyronemataceae 1, Valsaceae 1), Agaricomycetes is represented by 63 taxa (Agaricaceae 8, Auriculariaceae 1, Bolbitiaceae 2, Cyphellaceae 1, Diplocystidiaceae 1, Ganodermataceae 2, Gomphidiaceae 1, Hygrophoraceae 1, Hymenochaetaceae 2, Inocybaceae 2, Marasmiaceae 3, Meruliaceae 2, Mycenaceae 2, Niaceae 2, Omphalotaceae 4, Physalacriaceae 1, Pleurotaceae 1, Pluteaceae 2, Polyporaceae 4, Psathyrellaceae 8, Rhizopogonaceae 1, Schizophyllaceae 1, Stereaceae 1, Strophariaceae 5, Suillaceae 1, Tricholomataceae 5) and Dacrymycetes is represented by 1 taxon (Dacrymycetaceae 1).     THE CULTURE-INDEPENDENT ANALYSIS OF FUNGAL ENDOPHYTES OF WHEAT GROWN IN KWAZULU-NATAL, SOUTH AFRICA By Richard Jörn Burgdorf Submitted in partial fulfillment of the requirements for the degree of DOCTOR OF PHILOSOPHY In Microbiology School of Life Sciences College of Agriculture, Engineering and Science Pietermaritzburg South Africa December 2016 Thesis summary Fungal endophytes are of interest due to their diverse taxonomy and biological functions. A range of definitions exists based on their identity, morphology, location and relationship with their host. Fungal endophytes belong to a wide range of taxa and they are categorized by a variety of characteristics. The detection and identification of these fungal endophytes can be performed using culture-dependent and culture-independent methods. These organisms have a range of application in pharmaceutical discovery and agriculture. Agricultural applications include the exploitation of the growth promoting and protective properties of fungal endophytes in crops such as wheat. This important crop is grown in South Africa where biotic and environmental stresses pose a challenge to its cultivation. Fungal endophytes have demonstrated potential to ameliorate these challenges. Future research will reveal how they can be harnessed to fight food insecurity brought about by stress factors such as climate change. Extraneous DNA interferes with PCR studies of endophytic fungi. A procedure was developed with which to evaluate the removal of extraneous DNA. Wheat (Triticum aestivum) leaves were sprayed with Saccharomyces cerevisiae and then subjected to physical and chemical surface treatments. The fungal ITS1 products were amplified from whole tissue DNA extractions. ANOVA was performed on the DNA bands representing S. cerevisiae on the agarose gel.

Beug: Chair, NAMA Toxicology Committee Assessing the degree of amatoxin mushroom poisoning in North America is very challenging. Understanding the potential for various treatment practices is even more daunting. Although I have been studying mushroom poisoning for 45 years now, my own views on potential best treatment practices are still evolving. While my training in enzyme kinetics helps me understand the literature about amatoxin poisoning treatments, my lack of medical training limits me. Fortunately, critical comments from six different medical doctors have been incorporated in this article. All six, each concerned about different aspects in early drafts, returned me to the peer reviewed scientific literature for additional reading. There remains no known specific antidote for amatoxin poisoning. There have not been any gold standard double-blind placebo controlled studies. There never can be. When dealing with a potentially deadly poisoning (where in many non-western countries the amatoxin fatality rate exceeds 50%) treating of half of all poisoning patients with a placebo would be unethical. Using amatoxins on large animals to test new treatments (theoretically a great alternative) has ethical constraints on the experimental design that would most likely obscure the answers researchers sought. We must thus make our best judgement based on analysis of past cases. Although that number is now large enough that we can make some good assumptions, differences of interpretation will continue. Nonetheless, we may be on the cusp of reaching some agreement. Towards that end, I have contacted several Poison Centers and NAMA will be working with the Centers for Disease Control (CDC).

ABSTRACT Bioluminescence is a biological process through which light is produced and emitted by a living organism resulting from a chemical reaction within the body of the organism. The mechanism behind this phenomenon is an oxygen-dependent reaction involving substrates generally termed luciferin, which is catalyzed by one or more of an assortment of unrelated enzyme called luciferases. The history of bioluminescence in fungi can be traced far back to 382 B.C. when it was first noted by Aristotle in his early writings. It is the nature of bioluminescent mushrooms to emit a greenish light at certain stages in their life cycle and this light has a maximum wavelength range of 520-530 nm. Luminescence in mushroom has been hypothesized to attract invertebrates that aids in spore dispersal and testing for pollutants (ions of mercury) in water supply. The metabolites from luminescent mushrooms are effectively bioactive in anti-moulds, anti-bacteria, anti-virus, especially in inhibiting the growth of cancer cell and very useful in areas of biology, biotechnology and medicine as luminescent markers for developing new luminescent microanalysis methods. Luminescent mushroom is a novel area of research in the world which is beneficial to mankind especially with regards to environmental pollution monitoring and biomedical applications. Bioluminescence in fungi is a beautiful phenomenon to observe which should be of interest to Scientists of all endeavors.

Area of study: The study was carried out in Gaziantep province which is located in Southeastern Anatolian Region of Turkey. The study area takes place at the transition region of Mediterranean and Irano- Turanean phytogeographic regions and falls in C6 according to Davis’ grid square system. Material and method: The fungal specimens were collected from different localities within the boundaries of Gaziantep province between 2009 and 2015. After they were photographed at their natural habitats, they were transferred to the fungarium and the descriptive data related to their macroscopy and microscopy were obtained. Then the samples were identified by comparing the obtained descriptive data with literature data. Main results: Besides the taxa reported from the region before, 217 macrofungi species belonging to 129 genera, 63 families, 19 orders and 6 classes within Ascomycota and Basidiomycota were determined from Gaziantep. Research highlights: The mycota of Turkey has not prepared yet. That is why biodiversity studies on the macromycota of Turkey is an important requirement to present the overall macrofungal diversity of the country.
