Arife Merve KAHRAMAN Kayseri, Haziran 2017 vi AKKUYU NÜKLEER SANTRALİ (GÜLNAR,İÇEL) CİVARINDA YAYILIŞ GÖSTEREN BAZI MANTAR TÜRLERİNİN MORFOLOJİK , ANATOMİK VE MOLEKÜLER MARKIRLAR İLE BELİRLENMESİ Arife Merve KAHRAMAN Erciyes Üniversitesi Fen Bilimleri Enstitüsü Yüksek Lisans Tezi, Haziran 2017 Danışman: Prof. Dr. Mehmet Gökhan HALICI ÖZET Bu çalışmada; Türkiye'nin Akdeniz kıyı kesimindeki İçel İli'nin 150 km batısında bulunan Gülnar İlçesi'nin Akkuyu mevkiinde yayılış gösteren bazı likenleşmiş mantar türlerinin morfolojik, anatomik ve moleküler markırlar ile belirlenmesi hedeflenmiştir. Çalışmada Caloplaca erytrocarpha, Circinaria calcarea türleri ve Diplotomma venustım, Flavaploca havasii, Flavoplaca communis, Lecania rabenhorstii, Lobothallia radiosa, Megaspora verrucosa, Pertusaria rubefacta, Physcia stellaris, Psora taurensis, Roccella elisabethea ve Xanthocarpia marmorata türleri kullanılmıştır. Bu türlerden Roccella elisabethae ve likenikol mantar olan Arthonia follmaniana Türkiye için yeni kayıt iken, Psora taurensis bilim dünyası için tanımlanan yeni bir tür olmuştur. Moleküler markır olarak tüm örnekler için ITS ve bazı örnekler için ek olarak mtSSU primerleri kullanılmıştır ve bu sekanslar için filogenetik analizleri çalışılan örnekler ile gen bankasından alınan örnekler kullanılmıştır. Çalışılan örnekler arasındaki benzerliği belirlemek için ise Neighbour Joining ve Maksimum Likelihood yöntemleri kullanılmıştır. Ayrıca bu çalışmanın Türkiye Likenleşmiş mantar biyoçeşitliliğine katkıda bulunmasının yanı sıra, Akkuyu'ya kurulacak olan Nükleer Santralin etkilerinin ortaya koyulmasına bir nevi katkıda bulunması hedeflenmiştir. Bu bölgede kurulacak olan nükleer enerji santralinin etkilerinde kullanılabilecek bir biyomonitor olarak bu likenleşmiş mantarların verileri yardımcı olacaktır. Anahtar Kelimeler: Akkuyu, Nükleer Santral, Filogenetik analiz, ITS, PCR, Likenleşmiş mantar, Psora taurensis vii DETERMINATION BY MORPHOLOGICAL, ANATOMICAL AND MOLECULAR MARKERS OF SOME LICHENIZED FUNGI SPECIES DISTRUBITED NEARBY AKKUYU NUCLEAR POWERPLANT (GULNAR, ICEL) Arife Merve KAHRAMAN Erciyes University, Graduate School of Natural and Applied Sciences M.Sc.Thesis, June 2017 Supervisor: Prof. Dr. Mehmet Gökhan HALICI ABSTRACT In this study; we aimed to determine some lichenized fungi species distributed in Akkuyu region of Gülnar District 150 km west of İçel in the Mediterranean coast of Turkey by morphological and anatomical properties as well as by molecular markers. Arthonia follmaniana, Caloplaca erytrocarpia, Circinaria calcarea species and Diplotomma venustum, Flavaploca havasii, Flavoplaca communis, Lecania rabenhorstii, Lobothallia radiosa, Megaspora verrucosa, Pertusaria rubefacta, Physcia stellaris, Psora taurensis, Roccella elisabethea and Xanthocarpia marmorata species were determined in species level. Arthonia follmaniana and Roccella elisabethae has been a new record for Turkey, Psora taurensis has been a new species defined for the scientific world. ITS for all samples and mtSSU primers for some samples were used as molecular markers and the phylogenetic analyzes for these sequences and the samples taken from the gene bank were used. Neighbor Joining and Maximum Likelihood methods were used to determine the similarity between the studied samples. And also this study has been made to aim some kind of effects of the Nuclear Powerplant which will be establish in Akkuyu soon, As well as contribute to lichenized fungal biodiversity in Turkey. Many recent studies hypothesize global generality of trait patterns, which would allow for comparison among ecosystems and biomes and provide a foundation for general rules and theories, the so-called “Holy Grail” of ecology. However, the majority of these studies focus on functional trait patterns of vascular plants, with a minority examining the patterns of cryptograms such as lichens. Lichens are an important component of many ecosystems due to their contributions to biodiversity and their key ecosystem services, such as contributions to mineral and hydrological cycles and ecosystem food webs. Lichens are also of special interest because of their reliance on atmospheric deposition for nutrients and water, which makes them particularly sensitive to air pollution. Therefore, they are often used as bioindicators of air pollution, climate change, and general ecosystem health. This thesis examines the functional trait patterns of lichens in two contrasting regions with fundamentally different kinds of data. To better understand the patterns of lichen functional traits, we examined reproductive, morphological, and chemical trait variation along precipitation and temperature gradients in Oregon.

It harbors valuable regional biodiversity with vital ecosystem and are under anthropogenic threats. Sacred groves are fairly well-studied for socio-cultural and ecological aspects, and evaluation of higher flora and fauna and their conservation. However, there are no or scarce studies on cryptogams available on sacred groves. The lichens have long been recognized as biologically sensitive indicators of environmental conditions. The present study reveals the occurrence of 129 species of lichens, represented by 52 genera and 25 families in ten selected sacred groves. Shorea robusta exhibited the maximum diversity of lichens by 74 species. Since this is the first study of lichens in the district there is ample scope for further studies in South Bengal region. This study will help to understand the diversity of lichens better and give the correct status of the biodiversity of West Bengal. From a morphological point of view the species belongs to the M. griseoatra group, in which it is, so far, the only constantly lichen- icolous taxon. Chemically it is characterized by the regular presence of miriquidic acid, stictic acid, and constictic acid (in variable amounts). It starts its ontogenetic life cycle as an obligate youth para- site on Sporastatia, usually S. polyspora. Morpho-anatomical treatments of other species of the Miriquidica griseoatra group occurring in Europe, based on the examination of type material, are also presented. According to the characters exhibited by the lectotype of Lecidea contigua var. griseoatra Flot., M. griseoatra (Flot.) Hertel & Rambold is the correct name for a montane species with subdivided areoles and immersed, laterally adnate apothecia with a pigmented hypothecium. Specimens exhibiting this set of characters were usually named M. obnubila auct. According to the characters observed on the lectotype of Lecidea (Biatora) obnubila Th. Fr. & Hellb., M. obnubila (Th. Fr. & Hellb.) Hertel & Rambold is a species with sessile apothecia with a hyaline hypothecium. Lecidea subplumbea Anzi does not constitute a heterotypic synonym of Lecidea griseoatra (Flot.) Schaer. but is the legitimate name at the species level for Miriquidica griseoatra sensu auct., and therefore M.
