Thirteen strains of microalgae were isolated from the alpine zone in Canyon Creek, Canterbury, New Zealand. Ten species were characterized by traditional means, including ultrastructure, and subjected to phylogenetic analysis to determine their relationships with other strains. Because alpine algae are exposed to extreme conditions, and such as those that favor the production of secondary metabolites, it was hypothesized that alpine strains could be a productive source of PUFAs. Fatty acid (FA) profiles were generated from seven of the characterized strains and three of the uncharacterized strains. Some taxa from Canyon Creek were already identified from other alpine and polar zones, as well as non-alpine zones. The strains included relatives of species from deserts, one newly published taxon, and two probable new species that await formal naming. All ten distinct species identified were chlorophyte green algae, with three belonging to the class Trebouxiophyceae and seven to the class Chlorophyceae.

    Culturing a novel strain of Haematococcus pluvialis, LSBB612 (ALG_App006) Background The genus Haematococcus is found globally, with reports of isolates from all continents with the exception of Antarctica, with hostile areas of isolation including the artic circle (Klochkova et al., 2013). H. pluvialis is of commercial interest due to its ability to produce copious amounts of astaxanthin, reaching up to 5 % dry weight in the encysted aplanospore state (Wayame et al., 2013). Astaxanthin is sold as a pigment for aquaculture and in animal feed, and is marketed as an antioxidant for the nutraceutical market. The H. pluvialis derived astaxanthin industry is commercially successful; however, several constraints are ever-present including issues of contamination and grazing, high extraction costs, high light requirements for encystment, and conversely, photo-bleaching (Shah et al., 2016). Astaxanthin is produced under high light and nutrient deplete conditions (García-Malea et al., 2008). High temperature is rarely implemented to induce astaxanthin production, as it was reported to severely reduce biomass yield, and thus decrease astaxanthin productivity (Tjahjono et al. 1994). Currently the red stage of astaxanthin production is constrained by biomass production in the green stage, which requires strictly controlled culture conditions. Optimal reported temperatures for the vegetative growth of H. pluvialis are between 20 and 28°C (Wan et al., 2014), with temperatures in excess of 30°C shown to induce transition from the green vegetative stage to the red stage with the formation of aplanospores. Domínguez-Bocanegra et al., (2004) demonstrated optimal growth at an irradiance of 177 µmol photons/m 2/s with higher density cultures achieved under continuous light.

The microalgal cells were exposed to the various concentrations of ZnO NPs (10 to 200 mg/L) from 6 to 96 h to explore the dose- and time-dependent cellular accumulation and the corresponding cytotoxic effects. Scanning electron microscopy -X-ray energy dispersive spectroscopy (SEM EDX) analysis evidenced the cellular accumulation of zinc in the algal biomass. Quantification of cell associated zinc by inductively coupled plasma optical emission spectroscopy (ICP OES) reported a substantial accumulation of zinc in both microalgae with higher zinc accumulation in S.

Amino acid profile were significantly higher (p<0.05) in culture media based on inorganic fertilizer when compared to that in commercial medium (WC). High cell density (4.6 x 105 cells.mL-1) was obtained in NPK 10:10:10 and in WC medium (3.2 x 105 cells.mL-1) after 16 days cultivation. Media inorganic fertilizer (NPK) was adequate and may replace the commercial medium WC for the cultivation of microalgae H. pluvialis, with high nutritional value and biomass concentration.
