EDUCATIONAL COMMENTARY – TESTING FOR AMPHETAMINES AND RELATED COMPOUNDS Learning Outcomes Upon completion of this exercise, participants will be able to: • list amphetamine-like compounds that may be detected by immunoassay screening tests for amphetamines. • describe the 3 types of amphetamine immunoassays based on their antibody specificity. • discuss common causes of false-positive results when testing for amphetamines. Correct interpretation of testing results for amphetamines and related compounds is dependent on many factors including an understanding of the nomenclature, structure, and metabolism of these compounds. The class of phenethylamine compounds having varying degrees of sympathomimetic activity include amphetamine, methamphetamine, and many other compounds known by several names including amphetamines (as a group), designer drugs often grouped together as ecstasy [3,4- methylenedioxymethamphetamine (MDMA); 3,4-methylenedioxyamphetamine (MDA), and 3,4- methylenedioxy-N-ethylamphetamine (MDEA)], and sympathomimetic amines including ephedrine, pseudoephedrine, and phenylpropanolamine found in over-the-counter (OTC) medications. Amphetamine, a primary amine, and methamphetamine, a secondary amine, have a stereogenic center and have enantiomers or optical isomers designated d (or +) for dextrorotatory and l (or -) for levorotatory. In general, the d isomers are the more physiologically active compounds, but pharmaceutical preparations may consist of either isomer or a mixture of both isomers. When the mixture contains equal concentrations of the 2 enantiomers, it is known as a racemic mixture. The existence of enantiomers for amphetamines creates analytical and interpretative problems. Immunoassay Screening The primary screening methods for detecting amphetamines are immunoassays. The structural similarity to amphetamine or methamphetamine of the compounds previously mentioned makes it difficult to produce antibodies specific for amphetamine, methamphetamine, or both. -

CHEMICAL AND PHYSICAL DATA Khat is used as a stimulant or as a medicine in parts of Africa and the Arabian Peninsula. The plant is thought to have been in cultivation before the coffee plant; historical references date the use of the plant to the fourteenth century. Peter Forsskal, a physician and botanist, collected khat specimens in an expedition organized by the King of Denmark in the eighteenth century. Forsskal assigned the name Catha edulis to the plant. The effects produced by the drug include excitation, hypersensitivity, anorexia, insomnia, euphoria, increased respiration, and hyperthermia. These effects closely parallel the effects of d-amphetamine. Khat is a bush or tree that grows naturally in the humid mountainous regions (elevations of 5000 to 6500 feet) of East and South Africa. The trees can grow naturally to over 60 ft; however, cultivated khat trees are pruned and their height to kept to approximately 16 feet. Khat also grows to a height of 3 feet as a small bush in arid regions. Like opium, the alkaloid content of khat will vary with the soil, climatic conditions, and cultivation. Khat belongs to the genus Catha edulis. It is recognized that the genus consists of only one species; however, the plant exhibits extreme polymorphism. -

Michael LaVoe (1999) When Michael LaVoe observed Lee Morgan, a fellow freshman at Philadelphia's Mastbaum Vocational Technical High School, playing trumpet with members of the school's dance band in the first days of school in September 1953, he could not believe his ears. Morgan, who had just turned fifteen years old the previous July, had remarkable facility on his instrument and displayed a sophisticated understanding of music for someone so young. Other members of the ensemble, some of whom al- ready had three years of musical training and performing experience in the school's vocational music program, experienced similar feelings of dis- belief when they heard the newcomer's precocious ability. Lee Morgan had successfully auditioned into Mastbaum's music program, the strongest of its kind in Philadelphia from the 1930s through the 1960s, and demon- strated a rare ability that begged the title "prodigy." Almost exactly three years later, in November of 1956, Lee Morgan, now a member of die Dizzy Gillespie orchestra, elicited a similar response at the professional level after the band's New York opening at Birdland. Naturforsch. 35 c, 80-87 (1980); received October 24, 1979 (—)-Ephedrine, (—)-Ephedrine: NAD+-oxidoreductase, Pseudomonas putida, Arthrobacter globiformis A bacterium utilizing the alkaloid (-)-ephedrine as its sole source of carbon was isolated by an enrichment-culture technique from soil supplemented with 4-benzoyl-l,3-oxazolidinon-(2). The bacterium was identified as Pseudomonas putida by morphological and physiological studies. The following metabolites were isolated from the culture fluid: methylamine, formaldehyde, methyl- benzoylcarbinol (2-hydroxy-1-oxo-l-phenylpropane), benzoid acid, pyrocatechol and cis, cis- muconic acid. A pathway for the degradation of (-)-ephedrine by Pseudomonas putida is proposed and compared with the degradative pathway in Arthrobacter globiformis. The enzyme, which is responsible for the first step in the catabolism of (-)-ephedrine could be demonstrated in extracts from Arthrobacter globiformis. This enzyme catalyses the dehydrogena tion of (—)-ephedrine yielding phenylacetylcarbinol/methylbenzoylcarbinol and methylamine. It requires NAD+ as cofactor and exhibits optimal activity at pH 11 in 0.1 M glycine/NaOH buffer. The K m value for (—)-ephedrine is 0.02 mM and for N A D + 0.11 mM , respectively. No remarkable loss of activity is observed following treatment
