The increasing popularity of frusemide treatment in recent years has led to widespread concern over its effect on other drugs. The studies described in this thesis investigate the disposition of frusemide and its effect on the metabolism, pharmacokinetics and "detectability" of an acidic drug and a basic drug. Salicylic acid and theophylline were the two drugs chosen, in view of their therapeutic use and occasional occurrence as drugs of abuse. A new analytical method for the determination of frusemide was developed. The fluorescence properties associated with the ring structure make this drug particularly amenable to this form of detection, permitting the development of a selective method of assay using solid phase extraction and reverse phase HPLC. In the first two experiments three horses received on separate occasions in a randomized cross-over design frusemide (1 mg/kg; i.v.) alone, the radioactive test drug ([14C]-salicylic acid or [14C]-theophylline) alone and the test drug followed by frusemide (1 mg/kg; i.v.). Serial venous blood samples were collected from an indwelling jugular cannula for 24 h and urine was collected as voided for up to 96 h.
