It is caused by a positive, single stranded RNA virus from the enveloped coronaviruse family. However, the main viral proteinase (Mpro/3CLpro) has recently been regarded as a suitable target for drug design against SARS infection due to its vital role in polyproteins processing necessary for coronavirus reproduction. Objectives: The present in silico study was designed to evaluate the effect of Eucalyptol (1,8 cineole), a essential oil component from eucalyptus oil, on Mpro by docking study. Methods: In the present study, molecular docking studies were conducted by using 1- click dock and swiss dock tools. Protein interaction mode was calculated by Protein Interactions Calculator. Results: The calculated parameters such as RMSD, binding energy, and binding site similarity indicated effective binding of eucalyptol to COVID-19 proteinase. Active site prediction further validated the role of active site residues in ligand binding.

NOTE: 1) The RN or LPN is permitted to delegate ONLY after application of all components of the NCBON Decision Tree for Delegation to UAP and after careful consideration that delegation is appropriate: a) for this client, b) with this acuity level, c) with this individual UAP’s knowledge and experience, and d) now (or in the time period being planned). 2) Successful completion of the “Infection Control” module by the UAP should be documented prior to instruction in medication administration by this or ANY route. Procedure for: SUPPOSITORY 1. Perform skills in General Medication Administration Checklist. 2. Provide privacy. Have client void before procedure. 3. Put on clean gloves. 4. Position the client on their left (preferred) side with the top leg bent slightly. 5. Remove the foil or wrapper from the suppository, if present. 6. Apply a small amount of lubricant to the suppository and your gloved index finger on your dominant hand – the hand holding the suppository. 7. Separate the buttocks with your gloved, non dominant hand. 8. Ask the client to breathe slowly and deeply through the mouth. 9. Place tip of suppository against anus and gently insert the suppository about 4- inches along the rectal wall. Avoid putting the suppository in stool. 10. After removing your finger, squeeze buttocks together for a few minutes to help client hold in the suppository for as long as possible.

    Preparation of Aspirin, Methyl Salicylate and Nylon Experiment #6 Objective: To observe the contrasts in appearance and odor of two different esters of salicylic acid, namely aspirin and methyl salicylate. The preparation of nylon will be demonstrated. Introduction Salicylic acid is a phenolic acid, indicating it has two functional groups on the benzene ring, an -OH group and a -COOH group. Aspirin and oil of wintergreen are esters of salicylic acid. Aspirin is the ester of salicylic acid and acetic acid (acetyl salicylic acid), whereas oil of wintergreen is the ester of salicylic acid and methanol (methyl salicylate). The chemical name for aspirin, acetyl salicylic acid, is not easily recognized as the name of an ester. However, the chemical name for oil of wintergreen, methyl salicylate, should readily be recognized as the name of an ester. In the preparation of aspirin the -OH group of salicylic acid reacts with the acetyl group of acetic acid or acetic anhydride to form acetyl salicylic acid. The formation of aspirin will proceed faster if acetic anhydride is used in place of acetic acid. However, acetic anhydride will hydrolyze in the presence of water to form acetic acid, slowing down the reaction. Hence, the reaction vessel must be very DRY to ensure a complete reaction. Sulfuric acid, H2SO4, is used as a dehydrating agent. The aspirin will be isolated by filtration and washed several times with ice cold deionized water to remove the water soluble impurities (acetic acid and sulfuric acid). Aspirin is quite insoluble in ice cold water, but is relatively soluble in warm water (1 g dissolves in 100 mL water at 37EC).

This study characterizes a novel interactor of TRPM2. Using fusion proteins comprised of the TRPM2 C-terminus we established that tubulin interacted directly with the predicted C-terminal coiled-coil domain of the channel. In vitro studies revealed increased interaction between tubulin and TRPM2 during LPS-induced macrophage activation and taxol-induced microtubule stabilization. We propose that the stabilization of microtubules in activated macrophages enhances the interaction of tubulin with TRPM2 resulting in the gating and/or localization of the channel resulting in a contribution to increased intracellular calcium and downstream production of chemokines. ii Acknowledgments I would like to thank my supervisor Dr. Michelle Aarts for providing me with the opportunity to pursuit a Master’s of Science Degree at the University of Toronto.
