Ancak çok sayıda ilaçla etkileşime girmesi nedeniyle kullanırken dikkatli olunması gereken türlerdir. Bu çalışmada Hypericum türlerinin halk arasında kullanımları, sahip oldukları biyolojik aktiviteler ve etkileşime girdiği ilaçlar araştırılmıştır. Bu çalışmanın konusunun belirlenmesinde ve hazırlanma sürecinin her aşamasında değerli bilgilerini ve zamanını benden esirgemeyen, bana yol gösteren danışman hocam Sayın Prof. Dr. Hüsniye KAYALAR’a teşekkürlerimi özellikle belirtmek istiyorum. İZMİR – 2019 GİZEM ELBAY Özet Hypericum L. Türlerinin Biyolojik Aktiviteleri, Tıbbi Kullanımları ve İlaçlarla Etkileşimleri Antarktika dışında dünyanın her kıtasında yayılış gösteren 484 Hypericum L. (Hypericaceae) türü bulunmaktadır. Ülkemizde bulunan 96 Hypericum türünün 47’si endemiktir. St John’s wort olarak tanınan Hypericum bitkileri, çeşitli tıbbi özellikleri nedeniyle, geleneksel tıptaki kullanımlarıyla iyi bilinmektedir. Bazı Hypericum türleri pek çok ülkede (Avrupa ülkeleri, ABD) yerel halk tarafından hastalıkların tedavisinde kullanılmaktadır. Halk arasında dahilen antidepresan, spazm giderici, kurt düşürücü olarak, haricen antiseptik ve özellikle yanık yaraları başta olmak üzere yara iyileştirici olarak kullanılmaktadır. Hypericum cinsi Türkiye'de “kantaron” olarak adlandırılır. Bu familyanın en çok bilinen ve kullanılan türü Hypericum perforatum L. Türkiye’de “sarı kantaron, binbirdelik otu, kanotu, kılıçotu, mayasılotu, yaraotu, koyunkıran, batof’’ adlarıyla, uzun yıllardan beri çok iyi bilinen ve kullanılan tıbbi bir bitkidir. St John’s wort’un çiçekli kısımları, geleneksel Avrupa tıbbında, nevralji, anksiyete, nevroz ve depresyon tedavisinde yüzyıllardır kullanılmaktadır. Hayvan deneyleri ve klinik deneyler sonucunda antidepresan etkinliği kanıtlanan Hypericum perforatum L. bugün hafif ve orta şiddetteki depresyonların tedavisinde başarıyla kullanılmaktadır. Literatürde, Hypericum ektstrelerinin, ilaç metabolizmasından sorumlu hepatik enzimleri indüklediğini ve ilaçların serum düzeylerini ve terapötik etkinliğini azaltabileceğini gösteren çok sayıda rapor bulunmaktadır. Bu çalışmada Hypericum L. türlerinin biyolojik aktivitelerinin, yapılan çalışmalarda gösterilen tıbbi etkilerinin, halk arasındaki kullanımlarının ve ilaçlarla etkileşimlerinin derlenmesi amaçlanmıştır. Species, Their Medicinal Uses and Their Interactions with Drugs There are 484 Hypericum L. (Hypericaceae) species spreading in every continent of the world except Antarctica. 47 of the 96 Hypericum species found in our country are endemic. Hypericum plants, known as St John's wort, are well known for their use in traditional medicine because of their various medicinal properties. Some Hypericum species are used in many countries (European countries, USA) by local people to treat diseases. As an internal antidepressant, antispasmodic, anthelmintic, externally used as antiseptic and for wound healing, especially in burn wounds. The genus Hypericum is called as “kantaron” in Turkey. The most commonly known and used species of this genus in Turkey is Hypericum perforatum L. which is called as ‘‘sarı kantaron, binbirdelik otu, kanotu, kılıçotu, mayasılotu, yaraotu, koyunkıran, batof’’ is very well known medicinal herb used in traditional medicine for many years. The flowering parts of St John’s wort have been used in traditional European medicine for centuries to treat neuralgia, anxiety, neurosis and depression. Hypericum perforatum L., which has proven antidepressant efficacy as a result of animal and clinical trials, is used successfully in the treatment of mild and moderate depressions today. There are numerous reports in the literature that Hypericum extracts induce hepatic enzymes responsible for drug metabolism and may reduce serum levels and therapeutic efficacy of drugs. In this study, it was aimed to compile the biological activities of Hypericum L. species, their medicinal effects shown in the studies, their use among the public and their interactions with drugs. Bu çalışmada Hypericum cinsinin 2000 yılı öncesi ile 2001 yılına kadar belirlenen tehlike kategorileri araştırılıp veriler karşılaştırılmıştır. Yeni verilere göre 2000 yılından sonra Türkiye Florası'na eklenen yeni türler ile tehlike durumları da belirlenip durum değerlendirmesi yapılmıştır. Güncel verilerde bitkinin özellikle Ege ve Akdeniz Bölgesi’ndeki birçok türünün tehlike altında olduğu, çoğu endemik olan 15 Hypericum türünün ise tehlike (EN) kategorisine alındığı tespit edilmiştir. ABSTRACT he chemical composition of the essential oils of aerial parts of Hypericum lanuginosum var. lanuginosum TLam. and Hypericum perforatum L. were analyzed by GC and GC-MS. Forty one compounds were identified in the essential oils of H. lanuginosum with spathulenol (17.3%), caryophyllene oxide (13.1%), α-pinene (11.7%) and undecane (6.2%) as main constituents. Forty components were identified in the oil of H. perforatum with β-selinene (19.4%), bicyclogermacrene (15.3%), 2 tetradecene (8.2%) and α-amorphene (8.1%) as the most abundant components. ÖZET ypericum lanuginosum var. lanuginosum Lam. ve Hypericum perforatum L. taksonlarının toprak üstü Hkısımlarının uçucu yağlarının kimyasal birleşimi GC ve GC-MS (Gaz kromatografisi-kütle spektrometresi) ile analiz edildi. H. lanuginosum’da 41 bileşen saptandı. Bu türde spathulenol (17.3%), caryophyllene oxide (13.1%), α-pinene (11.7%) ve undecane (6.2%) major bileşenleri tespit edildi. H. perforatum taksonunda 40 bileşen tanımlandı. Bu bileşenlerden β-selinene (19.4%), bicyclogermacrene (15.3%), 2 tetradecene (8.2%) ve α-amorphene (8.1%) en çok bulunan bileşenlerdir. The Dadia–Lefkimi–Soufli National Park in North-Eastern Greece is a protected area distin- guished as particularly significant not only at the national but also at the European level. The long-term but mild human exploitation coupled with the heterogeneity of the habitats and the maintenance of nature have resulted in the conservation of an important biodiversity of biota, characterized by the existence of unique and rare species of flora and fauna. The present study provides an initial recording of the flora in the area based on fieldwork, and consisting of a total of 351 vascular taxa. Collective data on the chorology, life-form and the habitats of plant taxa are presented. Introduction The Dadia–Lefkimi–Soufli National Park is situated on the North - Eastern tip of Greece, close to the Turkish and Bulgarian borders. It is a representative example of preser- vation of the natural environment in a healthy state, as it has undergone only a small degree of ecosystems mismanagement and low levels of human annoyance. We examined the possible antigenotoxic/genotoxic properties of methanolic extract of H. olympicum subsp. olympicum (HOE) on human lymphocytes by employing sister chromatid exchange, micronucleus and comet assay and analyzed its chemical composition by GCxGC-TOF/MS. The anti-growth activity against MCF-7 and MDA-MB-231 cell lines was assessed by using the ATP viability assay. Cell death mode was investigated with fluorescence staining and ELISA assays. The major components of the flower and trunk were determined as eicosane, heptacosane, 2-propen-1-ol, hexahydrofarnesyl acetone and α-muurolene. HOE caused significant DNA damage at selected doses (250–750 µg/ml) while chromosomal damage was observed at higher concentrations (500 and 750 µg/ml). HOE demonstrated anti-growth activity in a dose-dependent manner between 3.13–100 µg/ml.
