This article is an open access publication Abstract Many nootropic compounds claim to have positive effects on cognitive performance. In this study, we tested the effects of the nootropic compound CAF+ on cognitive functioning. CAF+ contains a combination of ingredients that has separately shown to boost cognitive performance, including caffeine, l-theanine, vinpocetine, l-tyrosine, and vitamin B6/B12. We examined whether CAF+ would improve cognitive functions in healthy young participants, and whether it would be more effective than caffeine. We used a randomized double-blind placebo-controlled three-way cross-over design to examine the performance of 21 healthy young participants on a test battery aimed to measure memory performance, attention, and sensorimotor speed. Our main outcome measure was participant’s performance on the Verbal Learning Test (VLT). Subjective alertness, heart rate, and blood pressure were also monitored. Participants were tested at 30 and 90 min after treatment. We found that after 90 min, the delayed recall performance on the VLTafter caffeine was better than after CAF+ treatment. Further, caffeine, but not CAF+, improved the performance in a working memory task. In a complex choice reaction task caffeine improved the speed of responding. Subjective alertness was increased as a result of CAF+ at 30 min after administration. Only caffeine increased diastolic blood pressure. We conclude that in healthy young students, caffeine improves memory performance and sensorimotor speed, whereas CAF+ does not affect the cognitive performance at the dose tested. Keywords CAF+ . In this article, we summarize the state of the art on the neurochemistry of the cGMP system and then review the pre-clinical and clinical evidence on the use of cGMP enhancers in Alzheimer’s disease (AD) therapy. Although preclinical data demonstrates the beneficial effects of cGMP on cognitive deficits in AD animal models, the results of the clinical studies carried out to date are not conclusive.

Abstract: Pentoxifylline (PTX) is a methylxanthine derivative that has been developed as an im- munomodulatory agent and an improvement of microcirculation. Osteoradionecrosis (ORN) is a serious complication of radiation therapy due to hypovascularity. Coronavirus disease 2019 (COVID- 19) has spread globally. Symptoms for this disease include self-limiting respiratory tract illness to severe pneumonia and acute respiratory distress. In this study, the effects of PTX on RAW 264.7 cells were investigated to reveal the possibility of PTX as a therapeutic agent for ORN and COVID-19. To reveal PTX effects at the cellular level, protein expression profiles were analyzed in the PTX-treated RAW 264.7 cells by using immunoprecipitation high-performance liquid chromatography (IP-HPLC). PTX-treated RAW 264.7 cells showed increases in immunity- and osteogenesis-related proteins and concurrent decreases in proliferation-, matrix inflammation-, and cellular apoptosis-related pro- teins expressions. The effects of various selective phosphodiesterase (PDE) inhibitors on carbachol (CCh)-induced contraction in the bovine ab- omasum were investigated. Various selective PDE inhibitors, vinpocetine (type 1), erythro-9-(2-hydroxy-3-nonyl) adenine (EHNA, type 2), milrinone (type 3), Ro20-1724 (type 4), vardenafil (type 5), BRL-50481 (type 7) and BAY73-6691 (type 9), inhibited CCh-induced contrac- tions in a concentration-dependent manner. Among the PDE inhibitors, Ro20-1724 and vardenafil induced more relaxation than the other inhibitors based on the data for the IC50 or maximum relaxation. In smooth muscle of the bovine abomasum, we showed the expression of PDE4B, 4C, 4D and 5 by RT-PCR analysis. In the presence of CCh, Ro20-1724 increased the cAMP content, but not the cGMP content. By contrast, vardenafil increased the cGMP content, but not the cAMP content. These results suggest that Ro20-1724-induced relaxation was correlated with cAMP and that vardenafil-induced relaxation was correlated with cGMP in the bovine abomasum.
