In mice, nAChR activation by nicotine is anti-aggressive, or ‘serenic,’ an effect which requires α7 nAChRs and is recapitulated by GTS-21, an α7 α nAChR partial agonist. Pharmacotherapies potentiating 7 nAChR signaling have also been shown to reduce aggression in human α 15q13.3DS. These findings identify the 7 nAChR as an important regulator of aggressive behavior, but the underlying neurobiological α substrates remain to be determined. We therefore investigated the brain regions and potential neural circuits in which 7 nAChRs regulate aggressive behavior in male mice. As in 15q13.3DS, mice heterozygous for Chrna7 were significantly more aggressive compared to wild- type controls in the resident-intruder test. We subsequently examined the hippocampus, where α7 nAChRs are highly expressed, particularly in GABAergic interneurons. Resident–intruder interactions strongly activated granule cells in the dentate gyrus (DG). In contrast, GTS-21, which reduces aggression in mice, reduced DG granule cell activity during resident–intruder interactions. Short hairpin RNA knockdown of Chrna7 in the DG enhanced baseline aggression and eliminated the serenic effects of both nicotine and GTS-21 on α α attack latency. These data further implicate 7 nAChRs in regulation of aggression, and demonstrate that hippocampal 7 nAChR signaling is necessary and sufficient to limit aggression. These findings suggest that nAChR-mediated regulation of hippocampal excitatory–inhibitory balance could be a promising therapeutic intervention for aggression arising in certain forms of neuropsychiatric disease. G proteins are specialized proteins with the ability to bind the nucleotides guanosine triphosphate (GTP) and guanosine diphosphate (GDP). In unstimulated cells, the state of G alpha is defined by its interaction with GDP, G beta-gamma, and a GPCR. Upon receptor stimulation by a ligand, G alpha dissociates from the receptor and G beta-gamma, and GTP is exchanged for the bound GDP, which leads to G alpha activation. G alpha then goes on to activate other molecules in the cell. These effects include activating the MAPK and PI3K pathways, as well as inhibition of the Na+/H+ exchanger in the plasma membrane, and the lowering of intracellular Ca2+ levels. Most human GPCRs can be grouped into five main families named; Glutamate, Rhodopsin, Adhesion, Frizzled/Taste2, and Secretin, forming the GRAFS classification system. A series of studies showed that aberrant GPCR Signaling including those for GPCR-PCa, PSGR2, CaSR, GPR30, and GPR39 are associated with tumorigenesis or metastasis, thus interfering with these receptors and their downstream targets might provide an opportunity for the development of new strategies for cancer diagnosis, prevention and treatment. Nevertheless, over 90% of those requiring treatment for depression have yet to receive care,7 even though depression is one of the leading causes of disability in Malawi.8 One recent study estimated the point prevalence of depression in Malawi at 19%.9 Underlying the paucity of depression care is a perception that, relative to treatments for infectious disease, treatments for mental disorders are more time-intensive and less cost- effective.10 However, research to-date has two major shortcomings: First, evaluations often study the cost of stand-alone programs, rather than leveraging existing infrastructure to integrate care, which would reduce costs.11 The HIV care platform in Malawi, comprising a network of 708 facilities, represents one such example.

It has been hypothesized that the time lag between serotonin increase and reduction of MDD symptoms is due to downregulation of inhibitory receptors such as the serotonin 1B receptor (5-HT1BR). The research on 5-HT1BR has previously been hampered by a lack of selective ligands for the receptor. The last extensive review of 5-HT1BR in the pathophysiology of depression was published 2009, and based mainly on findings from animal studies. Since then, selective radioligands for in vivo quantification of brain 5-HT1BR binding with positron emission tomography has been developed, providing new knowledge on the role of 5-HT1BR in MDD and its treatment. The main focus of this review is the role of 5-HT1BR in relation to MDD and its treatment, although studies of 5-HT1BR in obsessive-compulsive disorder, alcohol dependence, and cocaine dependence are also reviewed. The evidence outlined range from animal models of disease, effects of 5-HT1B receptor agonists and antagonists, case-control studies of 5-HT1B receptor binding postmortem and in vivo, with positron emission tomography, to clinical studies of 5-HT1B receptor effects of established treatments for MDD.
