Still, it takes ones breath when realizing the huge challenges ahead. Articles and Letters Statistics has not always been considered as being very necessary. In 1848 the Dutch On Bayesian Measures of Ministry of Home Affairs established an ofŮice of statistics. ZCWPW1 Is Recruited to Recombination Hotspots by PRDM9
RESEARCH ARTICLE ZCWPW1 is recruited to recombination hotspots by PRDM9 and is essential for meiotic double strand break repair Daniel Wells1,2†*, Emmanuelle Bitoun1,2†*, Daniela Moralli1, Gang Zhang1, Anjali Hinch1, Julia Jankowska1, Peter Donnelly1,2, Catherine Green1, Simon R Myers1,2* 1The Wellcome Centre for Human Genetics, Roosevelt Drive, University of Oxford, Oxford, United Kingdom; 2Department of Statistics, University of Oxford, Oxford, United Kingdom Abstract During meiosis, homologous chromosomes pair and recombine, enabling balanced segregation and generating genetic diversity. In many vertebrates, double-strand breaks (DSBs) initiate recombination within hotspots where PRDM9 binds, and deposits H3K4me3 and H3K36me3. However, no protein(s) recognising this unique combination of histone marks have been identified. We identified Zcwpw1, containing H3K4me3 and H3K36me3 recognition domains, as having highly correlated expression with Prdm9. Here, we show that ZCWPW1 has co-evolved with PRDM9 and, in human cells, is strongly and specifically recruited to PRDM9 binding sites, with higher affinity than sites possessing H3K4me3 alone. Surprisingly, ZCWPW1 also recognises CpG dinucleotides. Male Zcwpw1 knockout mice show completely normal DSB positioning, but persistent DMC1 foci, severe DSB repair and synapsis defects, and downstream sterility. It traces the growing reliance upon contingent work as the foundation for industrial capitalism along the Pacific Coast; the shaping of urban space according to the demands of workers and capital; the formation of a working class subject through the discourse and social practices of both laborers and intellectuals; and workers’ struggles to improve their circumstances in the face of coercive and onerous conditions. Woven together, these strands reveal the consequences of a regional economy built upon contingent and migratory forms of labor. This workforce was hardly new to the American West, but the Pacific Coast’s reliance upon contingent labor reached its apogee after World War I, drawing hundreds of thousands of young men through far flung circuits of migration that stretched across the Pacific and into Latin America, transforming its largest urban centers and working class demography in the process. The presence of this substantial workforce (itinerant, unattached, and racially heterogeneous) was out step with the expectations of the modern American worker (stable, married, and white), and became the warrant for social investigators, employers, the state, and other workers to sharpen the lines of solidarity and exclusion. -

Statistics provides a special kind of understanding that enables well-informed deci- sions. As citizens and consumers we are faced with an array of choices. Statistics can help us to choose well. Our statistical brains need to be nurtured: we can all learn and practise some simple rules of statistical thinking. To understand how statistics can play a bigger part in our lives today we can draw inspiration from the founders of the Royal Statistical Society. Although in today’s world the information landscape is confused, there is an opportunity for statistics that is there to be seized.This calls for us to celebrate the discipline of statistics, to show confidence in our profession, to use statistics in the public interest and to champion statistical education. Introduction Dictionaries trace the source of the word statistics from the Latin ‘status’, the state, to the Italian ‘statista’, one skilled in statecraft, and on to the German ‘Statistik’, the science dealing with data about the condition of a state or community. The Oxford English Dictionary brings ‘statistics’ into English in 1787. Florence Nightingale held that ‘the thoughts and purpose of the Deity are only to be discovered by the statistical study of natural phenomena:::the application of the results of such study [is] the religious duty of man’ (Pearson, 1924). -
