ABSTRACT Oxalis Triangularis (A.St.-Hil) Or Commonly Known As
ABSTRACT Oxalis triangularis (A.St.-Hil) or commonly known as ‘Pokok Rama-rama’ in Malaysia is a beautiful ornamental plant which is propagated by bulbs. The plant grows to a height of 0.1 m - 0.2 m and is perfect for cultivating in pots or containers. Nowadays, with the emerging and advanced technologies, an efficient protocol has been established for a rapid multiplication of Oxalis triangularis in a large scale production under aseptic conditions. In vitro plant regeneration of Oxalis triangularis was successfully obtained in the present study via petiole and leaf as explants. The petiole explants cultured on MS medium supplemented with 0.5 mg/l α-Naphthaleneacetic acid (NAA) and 1.0 mg/l Kinetin (KIN) produced maximum number of adventitious shoots (12 shoots) while for leaf explants, the best treatment achieved on MS medium supplemented with 1.0 mg/l NAA and 1.5 mg/l 6- Benzylaminopurine (BAP) which produced a maximum of 14 shoots within 8 weeks. Comparison between in vivo plants and in vitro was observed using a Scanning Electron Microscope. The morphological features for both petiole and leaf samples have no differences. Both contain same structures of stomata and trichomes. In vitro flowering which is very important in order to improve quality and shortened physiological process of flowering was observed when adventitious shoots explants cultured on MS medium supplemented with 0.5 mg/l NAA and 0.5 mg/l BAP (90% in vitro flowering). In the synthetic seeds study, two different storage durations were tested (Day 7 and Day 30). The highest frequency of synthetic seeds production in Oxalis triangularis was recorded on Day 7 with 96.67% of conversion frequency. -

Echaide Abstract: Variation in repeated nucleotide sequences (RNSs) at the level of entire families assayed by Southern blot hybridization is remarkably low within species and is a powerful tool for scrutinizing the origin of allopolyploid taxa. Thirty-one clones from RNSs isolated from different Triticeae genera were used to investigate the genome constitution of South American Elymus. One of these clones, pHch2, preferentially hybridized with the diploid H genome Hordeum species. Hybridization of this clone with a worldwide collection of Elymus species with known genome formulas showed that pHch2 clearly discriminates Elymus species with the H genome (StH, StHH, StStH, and StHY) from those with other genome combinations (Sty, StStY, StPY, and StP). Hybridization with pHch2 indicates the presence of the H genome in all South American Elymus species except Elymus erianthus and Elymus mendocinus. Hybridization with additional clones that revealed differential restriction fragments (marker bands) for the H genome confirmed the absence of the H genome in these species. Hipkins Abstract: Genetic variation in the self-fertile, allotetraploid grass Elymus glaucus Buckley was assessed using isozymes in 133 populations from southwestern Oregon and from the San Francisco Bay area and central Sierra Nevada moun- tains in California. Elymus glaucus was highly (98.5%) homozygous but also highly variable; 77% of loci were poly- morphic, and the mean number of alleles per locus was 2.96. Populations were highly differentiated, with 40% of variation among populations. Geographic and genetic distances among populations were not correlated, except that pop- ulations collected within 5 km were generally more similar than average. Genetic distance among populations could not be predicted from geographic distance, geographic location, foliage pubescence, serpentine substrate, or habitat mois- ture. However, two genetic clusters, associated with elevation, did emerge. The taxonomic status of Elymus glaucus ssp. jepsonii (Burtt Davy) Gould, based on leaf pubescence, was not supported. PALMATIFOLIAE (OXALIDACEAE): MORFOLOGÍA DE LOS GRANOS DE POLEN Y DIVERSIDAD DE LAS ORBÍCULAS ALICIA LÓPEZ1* y SONIA ROSENFELDT2 Resumen: La morfología del polen y las características de las orbículas fueron estudiadas en todas las especies de Oxalis sección Palmatifoliae, endémica del sur de Sudamérica, empleando microscopio óptico (M.O.) y microscopio electrónico de barrido (M.E.B.). Los granos de polen son 3-colpados, de forma prolato esferoidales, oblato esferoidales o esferoidales. La exina es microrreticulada. Lúmen circular a poligonal; disminuyendo de tamaño hacia los polos. Las orbículas están dispersas irregularmente en la superficie interna de las anteras; son generalmente aplanadas, redondeadas o con forma de “dona”, de superficie lisa. La homogeneidad observada en la morfología de granos de polen de las especies de Oxalis sección Palmatifoliae refuerza la hipótesis de su origen monofilético y la variedad de tipos de orbículas podría contribuir a la diferenciación interespecífica. Pollen morphology and orbicules characteristics were studied by means of transmitted light microscopy (LM) and scanning electron microscopy (SEM) within five species of Oxalis section Palmatifoliae endemic from southern South America. Oxalis pollen grain is generally 3-colpate and the shape is prolate spheroidal, oblate spheroidal or spheroidal. The exine is microreticulate. The brochi are circular to polygonal; brochi decrease in size near the colpi.
