Pau (Crassulaceae): an anatomical, cytochemical and ultrastructural analysis Emilia Brzezicka1 & Małgorzata Kozieradzka-Kiszkurno1 Received: 9 July 2018 /Accepted: 4 October 2018 # The Author(s) 2018 Abstract Available documentation about the development of the female gametophyte of Crassulaceae is very limited. The aim of this study was to extend the embryological knowledge of Crassulaceae by analysing the development of the embryo sac in Sedum sediforme. Transmission electron microscopy and light microscopy including Nomarski optics (DIC) were used to observe individual stages of female gametophyte development. Cytochemical staining enabled detection of lipids, insoluble polysaccharides and proteins in gametophyte cells during their formation. Their increased accumulation was observed during nucellar cell and unfunctional cell degeneration in the embryo sac at the coenocytic and cellular stages (megagametogenesis). The female gametophyte develops in anatropous, bitegmic and crassinucellate ovules. The mature embryo sac is built of seven cells but after antipodes degeneration it is formed by the egg apparatus and a central cell. The monosporic Polygonum type was observed. One megaspore mother cell (MMC) formed three cells after meiosis. A triad was formed from a functional megaspore (placed chalazally), one uninucleate megaspore and a binucleate cell located at the micropylar end. Plasmodesmata with adhering electron-dense dome were noticed in walls of the coenocytic embryo sac and in the outer walls of ephemeral antipodes. Moreover, similar to synergids, antipodes form wall ingrowths. Here, we report new structural features of the antipodal cells (the presence of plasmodesmata with an electron-dense dome) which have not been described before.

Dunant 3, I-21100 Varese, Italy ([email protected]) ABSTRACT Foliar trichomes of gray-leaved Tillandsioideae (Bromeliaceae) are highly reflective, suggesting a role in protecting the leaf against direct sunlight in exposed niches. The performance of photosystem II, as denoted by the chlorophyll fluorescence characteristic Fv /Fm , was determined for seven Tilland- sia species and Vriesea barclayana that were exposed to excessive light, with trichomes either present or removed. Additionally, trichome structure and interaction with light was recorded using extended depth-of-field photomicrography, and reflectance quantified using a novel photographic technique. Trichomes of mesomorphic Type IV life forms (T. cryptantha, T. cyanea) and of the intermediate life form V. barclayana conferred reflectance of between 1 and 11%, which did not significantly influence Ϫ2 Ϫ1 Fv /Fm when exposed to a high light intensity of 1500 ␮mol m s (photosynthetically active radiation) for one hour.

    Unexpected Behavior of Some Nitric Oxide Modulators under Cadmium Excess in Plant Tissue Jozef Kova´cˇik1,2*, Petr Babula3, Borˇivoj Klejdus1,2, Josef Hedbavny1, Marke´ta Jarosˇova´ 1 1 Institute of Chemistry and Biochemistry, Faculty of Agronomy, Mendel University in Brno, Brno, Czech Republic, 2 CEITEC – Central European Institute of Technology, Mendel University in Brno, Brno, Czech Republic, 3 Department of Natural Drugs, Faculty of Pharmacy, University of Veterinary and Pharmaceutical Sciences Brno, Brno, Czech Republic Abstract Various nitric oxide modulators (NO donors - SNP, GSNO, DEA NONOate and scavengers – PTIO, cPTIO) were tested to highlight the role of NO under Cd excess in various ontogenetic stages of chamomile (Matricaria chamomilla). Surprisingly, compared to Cd alone, SNP and PTIO elevated Cd uptake (confirmed also by PhenGreen staining) but depleted glutathione (partially ascorbic acid) and phytochelatins PC2 and PC3 in both older plants (cultured hydroponically) and seedlings (cultured in deionised water). Despite these anomalous impacts, fluorescence staining of NO and ROS confirmed predictable assumptions and revealed reciprocal changes (decrease in NO but increase in ROS after PTIO addition and the opposite after SNP application). Subsequent tests using alternative modulators and seedlings confirmed changes to NO and ROS after application of GSNO and DEA NONOate as mentioned above for SNP while cPTIO altered only NO level (depletion). On the contrary to SNP and PTIO, GSNO, DEA NONOate and cPTIO did not elevate Cd content and phytochelatins (PC2,PC3) were rather elevated. These data provide evidence that various NO modulators are useful in terms of NO and ROS manipulation but interactions with intact plants affect metal uptake and must therefore be used with caution.

Box 21692, Kitwe, Zambia Received: 19.05.2020 Accepted: 01.07.2020 ABSTRACT: Industrial activities compromise the ambient air quality at a local, regional and global level through gaseous and dust emissions. This study reviews uptake mechanisms and the associated phytotoxicity of pollutants in plants, focusing on heavy metals and SO2. It further describes detoxification mechanisms and the resultant biochemical and physiological changes in plants. Finally, the morpho-physiological and growth responses to stress-induced biochemical changes are discussed. Heavy metals and SO2 enter the plant tissue through the stomata, cuticular layers, lenticels and root hairs. In 2- 2- the plant cells, SO2 converts to SO3 or SO4 ions upon reacting with water molecules, 2- which in excess are toxic to plants. However, the detoxification process of SO3 increases the production of reactive oxygen species (ROS). ROS are toxic to plants and damages biomolecules such as lipids, proteins, carbohydrates and DNA. On the other hand, heavy metals, such as Cu and Fe catalyse the Fenton/Haber-Weiss reactions, breaking down H2O2 into OH•.
