Zürich, 1990 ZUSAMMENFASSUNG
Zellsuspensionskulturen von Vanilla planifolia: Eine Suspensionskultur von Vanilla planifolia (Andr.), etabliert aus oberflächensterilisierten Kapseln, wurde in MS-
Medium, versetzt mit 1 mg/l 2,4-D, charakterisiert. Die geringe Aktivität des
Phenylpropanoid Stoffwechsels der in diesem Medium wachsenden Zellen konnte
durch eine Veränderung der Zusammensetzung der Phytohormone gesteigert werden.
In einem MS-Medium, versetzt mit 1 mg/l NAA, zeigten die Zellen eine etwa 60-
fach erhöhte Konzentration löslicher Phenole.
Fütterung verschieden substituierter Zimtsäuren: Zwischenprodukte des allge¬
meinen Phenylpropanoid Stoffwechsels, wie Zimt- und Ferulasäure, wurden von den
Zellen aus dem Medium aufgenommen und vor allem in ligninartiges Material
eingebaut. Demgegenüber wurden 4-Methoxy-substituierte Zimtsäuren (4-Methoxy-,
3,4-Dimethoxy- und 3,4,5-Trimethoxyzimtsäure), die als schlechte Substrate der 4-
Hydroxycinnamat:CoA-Ligase gelten, nicht in ligninartiges Material eingebaut. Sie
wurden in den Zellen zu den entsprechenden C6-C,-Säuren abgebaut (4-Hydroxy-
benzoe-, Vanillin- und Syringasäure). Isoferulasäure hingegen wurde nicht direkt zur
entsprechenden C6-C,-Säure abgebaut (Isovanillin- oder Protocatechusäure), sondern
zu 3,4-Dimethoxyzimtsäure methyliert und dann zu Vanillinsäure umgesetzt. Die
Isoferulasäure, die in den Zellen durch eine Methylierung der Kaffeesäure gebildet
werden kann, war also eine Vorstufe der Vanillinsäure.
Inhibition der Ligninbiosynthese: 3,4-(Methylendioxy)-zimtsäure (MDCA) zeigte
in Fütterungsexperimenten eine inhibierende Wirkung auf die Ligninbiosynthese. Als
Ursache konnte in zellfreien Extrakten eine Inhibition der 4-Hydroxycinnamat:CoA-
Ligase durch MDCA gezeigt werden (Kj = 100 /_M). Bedingt durch die Inhibition der
Ligninbiosynthese wurde der Stoffluss in den MDCA-behandelten Zellen in die C6-
C,-Biosynthese geleitet und bis zu 10.5 mg Vanillinsäure pro Liter Suspension konnten nachgewiesen werden. Radioaktiv markierte Zimtsäure, nicht aber Ferula¬
säure, wurde in MDCA-behandelten Zellen in die Vanillinsäure eingebaut.
Induktion verschiedener Produkte und Enzyme: Die Suspensionskulturen wurden
mit Chitosan und Kinetin induziert. Chitosan induzierte zwei Enzyme des allgemeinen Phenylpropanoid Stoffwechsels (Phenylalanin-Ammonium-Lyase und 4-Hydroxycin- namat:CoA-Ligase), sowie zwei Enzyme der Ligninbiosynthese (4-Hydroxycinna- moyl-CoA Reduktase und Coniferylalkohol-Dehydrogenase). Bis zu 300 mg lignin¬ artiges Material pro Liter Suspension konnte in diesen Zellen nachgeweisen werden.
In kinetininduzierten Zellen wurden zusätzlich zu den zwei Enzymen des allgemeinen
Phenylpropanoid Stoffwechsels auch Enzyme der Vanillinsäurebiosynthese induziert.
Zwei Isoenzyme der O-Methyltransferase (OMT) konnten aus diesen Zellen durch
DEAE-Sepharose Chromatographie aufgetrennt werden. Sie methylierten in vitro
Kaffeesäure zu Ferulasäure (30MT) beziehungsweise zu Isoferulasäure (40MT).
Die 40MT, die Kaffee- zu Isoferulasäure methyliert, konnte nur in kinetininduzierten
Zellen nachgewiesen werden und stellt wahrscheinlich das Schlüsselenzym der C6-C,- Biosynthese dar. Es konnte gezeigt werden, dass die Vanillinsäurebiosynthese in den
Suspensionskulturen von V. planifolia ausgehend von Zimtsäure über 4-Cumar-,
Kaffee-, Isoferula-, 3,4-Dimethoxyzimtsäure und 3,4-Dimethoxybenzoesäure verläuft.
Die verschiedenen enzymatischen Schritte von Zimt- zu 3,4-Dimethoxyzimtsäure konnten in zellfreien Extrakten nachgewiesen werden.
Vier Tage nach der Induktion mit Kinetin konnten 7.5 mg Vanillinsäure pro
Liter Suspension aus den Zellen isoliert werden. In Kombination mit dem Lignin- inhibitor (MDCA) stieg die Vanillinsäurekonzentration auf 15.5 mg/l Suspension. ABSTRACT
CeU Suspension cultures of Vanilla planifolia: A Suspension culture of Vanilla planifolia
(Andr.), established from surface sterilised green Vanilla pods, cultivated in MS- medium, supplemented with 1 mg/l 2,4-D, has been eharaeterized. The low activity of the phenylpropanoid metabolism observed in this medium could be increased by altering the phytohormone composition. In MS-medium, supplemented with 1 mg/l
NAA, the cells showed a 60-fold increase in the accumulating soluble phenolics. Feeding of substituted cinnamic acids: Intermediates of the general phenyl¬ propanoid metabolism (e.g. cinnamic and ferulic acid) added to the growth medium, were taken up by the cells and were mainly incorporated into ligneous material. In contrast, 4-methoxy substituted cinnamic acids (i.e. 4-methoxy-, 3,4-dimethoxy- and
3,4,5-trimethoxy cinnamic acid) known to be poor Substrates for the 4-hydroxy cin- namate:CoA-ligase, were not incorporated into ligneous material. They were converted to the corresponding C6-C,-acids (i.e. 4-hydroxy benzoie-, vaniUic- and syringic acid). Isoferulic acid was not converted directly to a C6-C,-acid, such as isovanillic- or protocatechuic acid. It was first methylated to dimethoxy cinnamic acid and then degraded to vanillic acid. Isoferulic acid, which can be formed within the cells by methylation of caffeic acid, is thus a possible precursor of vanülic acid.
Inhibition of lignin biosynthesis: In feeding experiments, 3,4-(me.hylened_oxy)c_n- namic acid (MDCA) showed an inhibitoty effect on lignin biosynthesis. An inhibition of the 4-hydoxy cinnamate:CoA-ligase could be demonstrated in cell free extracts by
MDCA (K| = 100 /_M). Due to this inhibition, the metabolic flow was channelled into the Cg-C,-biosynthesis and up to 10.5 mg vanillic acid per litre Suspension could be isolated from these cells. Radiolabelled cinnamic acid, but not ferulic acid, was incorporated into vanillic acid in MDCA treated cells.
Induction of different products and enzymes: Chitosan and kinetin were used for elicitation of the Suspension eultured cells. Chitosan induced two enzymes of general phenylpropanoid metabolism (Phenylalanine ammonia-lyase and 4-hydoxy cinnamate:
CoA-ligase), as well as two enzymes of ligninbiosynthesis (4-hydroxy cinnamoyl-CoA
reduetase and coniferyl alcohol dehydrogenase). Up to 300 mg ligneous material per litre Suspension could be isolated from these cells. In addition to the two enzymes of general phenylpropanoid metabolism, enzymes involved in vanillic acid biosynthesis were induced by kinetin. Two isoenzymes of O-methyltransferase (OMT) could be separated from these cells by DEAE-Sepharose chromatography. They methylated caffeic acid in vitro to ferulic- (30MT) and isoferulic acid (40MT), respectively. The
40MT, catalysing the methylation of caffeic- to isoferulic acid, was only detectable in kinetin induced cell extracts and it represents most likely the key enzyme of
Cg-C,-biosynthesis. The biosynthesis of vanillic acid in cell Suspension cultures of V planifolia has been shown to proeeed via cinnamic-, 4-coumaric-, caffeic-, isoferulic-, dimethoxy cinnamic acid and dimethoxy benzoie acid. The partieular enzymatic steps from cinnamic- to dimethoxy cinnamic acid have been shown in cell free extracts.
Four days after induction with kinetin, 7.5 mg vanillic acid per litre Suspension could be isolated from these cells. In combination with the inhibitor of lignin biosynthesis (MDCA), the vanillic acid concentration increased to 15.5 mg/l Suspension.
