MUITO OBRIGADO! iv RESUMO Os transtornos de ansiedade e a doença de Parkinson são patologias que atingem uma grande parcela da população. Grande parte das alternativas terapêuticas para essas patologias não contribui para melhora de todos os aspectos clínicos e/ou acarreta efeitos colaterais indesejáveis, havendo assim uma grande demanda para o desenvolvimento de novos fármacos. A Passiflora cincinnata Mast., planta conhecida popularmente por “maracujá do mato”, “maracujá tubarão” ou “maracujá mochila”, é uma espécie nativa presente em diversos estados brasileiros. As espécies do gênero Passiflora são muito conhecidas por suas flores vistosas, seus frutos comestíveis de sabor marcante e por suas propriedades sedativas, tranquilizantes e ansiolíticas relatadas pela medicina popular. São plantas que apresentam compostos orgânicos importantes como fenóis, glicosídeos cianogênicos, alcalóides e flavanóides, sendo estes responsáveis pelas atividades ansiolítica, antioxidante, antiinflamatória, anti- hiperglicêmica, entre outras quando testadas em mamíferos. Apesar disto, poucos são os estudos realizados no sentido de investigar os possíveis efeitos biológicos in vivo do extrato da espécie Passiflora cincinnata Mast. Então, neste estudo avaliamos o efeito do extrato alcoólico desta planta na ansiedade e em um modelo animal da doença de Parkinson. O extrato etanólico de P. cincinnata não acarretou alterações significativas nos parâmetros clássicos relacionados com ansiedade no labirinto em cruz elevado, tanto para o tratamento agudo como para o crônico. Em relação ao potencial neuroprotetor, como demonstrado anteriormente por nosso grupo, a administração de injeções repetidas de reserpina induz prejuízos motores progressivos, como o aumento no tempo de catalepsia em barra, aumento da frequência de tremor de mandíbula e mastigação no vácuo, e redução na velocidade média dos animais no campo aberto, bem como inibiu a produção de tirosina hidroxilase em células do SNpc. Além disso, este tratamento promoveu prejuízo na evocação da memória aversiva nos camundongos submetidos a esquiva discriminativa no labirinto em cruz elevado. De forma inesperada, a administração de passiflora também acarretou prejuízo na evocação da memória. Em contrapartida, a injeção concomitantemente com extrato etanólico de P. cincinnata, foi capaz de retardar o surgimento de déficits motores avaliados pela catalepsia em barra, e mais, reverteu o déficit na produção de TH na SNpc. Palavras-chave: Produtos naturais, Passiflora cincinnata, Ansiedade, Doença de Parkinson. v Abstract Anxiety disorders and Parkinson’s disease (PD) affect a large portion of the world population. Indeed, therapeutic alternatives available do not contribute to improve most clinical conditions and/or are linked with undesirable side effects. Thus, there is a great demand for the development of new drugs to treatment of these diseases. Passiflora cincinnata Mast. is a native species present in several Brazilian states, popularly known as “maracujá do mato”, “maracujá tubarão” or “maracujá mochila”. Additionally, species of Passiflora genus are traditionally known for their exotic flowers, edible fruits with pronounced flavor and for their sedative, tranquilizer and anxiolytic properties reported by folk medicine. These plants possess important organic compounds such as phenols, cyanogenic glycosides, flavonoids and alkaloids, which are responsible for the anxiolytic, antioxidant, anti-inflammatory, anti- hyperglycemic, among others activities when tested in mammals. Despite this fact, only a few studies have been conducted to investigate the possible in vivo biological effects of Passiflora cincinnata Mast extracts. Thereby, in this study we evaluated the effects of the alcoholic extract of this plant in anxiety and PD animal model. Mice acutely or chronically administered with ethanolic extract of P. cincinnata do not showed any anxiogenic- or anxyolitic-like effect in elevated plus maze (EPM). In order to reproduce PD symptom’s in mice, we administered repeated injections of reserpine which progressively induced motor impairments such as increase in catalepsy, oral movements, and reduction of the average speed of the animals in the open field, as well as depleted dopamine prodution in SNpc cells. Furthermore, this treatment resulted in the loss of aversive memory recall in mice when undergoing PMDAT. Yet, passiflora group also show this amnesic profile. However, animals treated concomitantly with the alcoholic extract of Passiflora cincinnata Mast. showed higher latency for the onset of motor impairment evaluated by catalepsy. Thus, our results shows that the alcoholic extract of the plant P. cincinnata was able to delay the onset of the catalepsy induced by reserpine administration, plus reverted the depletion of dopamine production in SNpc cells. The genetic diversity and characteristics of commercial interest of Passiflora species make it useful to characterize wild germplasm, because of their potential use for fruit, ornamental and medicinal purposes. We evaluated genetic diversity, using RAPD markers, of 32 genotypes of Passiflora cincinnata collected from the wild in the region of Vitória da Conquista, Bahia, Brazil. Thirteen primers generated 95 polymorphic markers and only one monomorphic marker. The mean genetic distance between the genotypes estimated by the complement of the Dice index was 0.51 (ranging from 0.20-0.85), and genotype grouping based on the UPGMA algorithm showed wide variability among the genotypes. Acevedo-Rodríguez (Feb 2020) A predominantly tropical family with few species reaching warm-temperate regions, of about 15-17 genera and 850 species of tendrilled lianas or vines, or sometimes shrubs, small trees, or annuals with a perennial rootstock or a fleshy caudex. Represented in the Neotropics by 4 genera and about 600 species, occupying diverse habitats, from savanna to flooded forests, but most abundant in tropical rain forests on terra firme. Most species occur at low to middle elevations, but some grow above the tree line on Andean slopes. Diagnostics: Distinguished by the flowers with Dilkea sp., photo by L. Marinho an extrastaminal corona and usually a gynophore, and by the common presence of petiolar nectaries. Sterile collections of Passifloraceae may be confused with members of Cucurbitaceae as both families may have simple, alternate leaves, axillary tendrils, and petiolar nectaries. However, Passifloraceae is differentiated by the presence of stipules, unbranched axillary tendrils (trifid in Dilkea) [vs. exstipulate and axillary-lateral tendrils (forming a 90º angle with the petiole) that are commonly branched in Cucurbitaceae]. Also, resembles Vitaceae but tendrils and inflorescence in this family are opposite to the leaves, not axillary. 1 General Characters 1. STEMS. Stems are woody or herbaceous depending on the species. The study involved six treatments with four replicates of eight plants per plot and was arranged in a completely randomized block design. The seedlings were produced on a non-sterile substrate composed of a mixture of soil and bovine manure at the ratio of 3:1. The percentage of germination was high for all studied species, and the rate of graft development and survival was higher than 70 and 85.71%, respectively, within 60 days after grafting. The objective of the study was to evaluate the pollen morphology of 18 Passiflora species in order to identify informative pollinic characteristics to contribute to the taxonomic classification of the genus. The morphology of the pollen grains and the exine structure were investigated using light microscopy and scanning electron microscopy. Differences in the pollen morphology were found in the studied species, mainly in terms of shape, pollen aperture and ornamentation pattern of the exine. Most of the species belonging to the subgenus Passiflora presented ornamented 6-syncolpate pollen grains with an oblate-spheroidal shape and an exine with large lumens. In the subgenus Decaloba the pollen grains were 6-colporate, 12-colpate and 12-colporate, with subprolate to prolate-spheroidal shape, as well as an exine with smaller lumen size and few ornamented.

The aim of this study was to karyotypically characterize the chromosome heritance patterns of the progeny resultant from a cross of P. gardneri and P. gibertii using classical cytogenetics, chromosome banding, and molecular cytogenetics. Results: All analyzed genotypes showed the same diploid chromosome number as the genitor species: 2n = 18. Classical and CMA3 and DAPI staining allowed for chromosome counting and satellite identification (secondary constrictions). Fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH) were used to characterize subgenomes by either identifying rDNA-specific genome patterns or parental genomes, respectively. Conclusions: The heritance of chromosomal markers presenting rDNA sites from each parent for genome identification confirmed that all obtained plants were hybrids. These results will improve breeding programs involving the species of this genus. Apart from confirming hybridization, GISH allowed the visualization of recombination between the homeologous chromosome and the introgression of sequences of interest. Our intent was to expand knowledge about native and introduced vegetation in environments with anthropogenic changes. The survey occurred over 12 months and 266 species, distributed into 53 botanical families were identified. This flora was composed of native and exotic plants, especially from Americas (82.7%) and Old World and Australia (17.3%). There were 220 species native to the America’s, but the highest number (60%) was from Brazil. A small percentage of these Brazilian plant species (28.6%) have originated from Cerrado, suggesting that ruderal vegetation was the high represented by native species.

In this context, an anatomical study of the pericarp may help in understanding the changes which occur during the postharvest period. The objective of this work was to anatomically characterize the P. cincinnata pericarp, focusing on the structural changes which occur during refrigerated storage. The selected fruits were kept in net-like packaging at 5°C and evaluated every 15 days for a period of 90 days. Pericarp samples were hand cut on these dates for structural description and to detect starch, phenolic compounds, alkaloids, polysaccharides, proteins, lipids and lignin through histochemical tests. Other samples were analyzed by scanning electron microscope. The fruit is composed of three anatomically distinct regions: exocarp, mesocarp and endocarp. The mesocarp stands out for the presence of sclerified cells and the lax parenchyma, which increased its number of layers during the evaluated period. Idioblasts with prismatic crystals, drusen, silica crystals and raphides were identified, but the amounts reduced over time. This reduction and enlargement of the lax parenchyma were the main changes observed in the postharvest period of P. cincinnata pericarps.
