STRESEMANN From the Asthmapoliklinik, Free University, West Berlin, Germany (Received May 14, 1963) The dose/response curves for the protective effects of the new antihistamine compounds trimeprazine, 10-(3-diethylamino-2-methylpropyl)phenothiazine 1,1-dioxide hydrochloride (oxomemazine hydrochloride), cyproheptadine, homochlorcyclizine and methotrimeprazine against the anaphylactic microshock of the guinea-pig were similar to that of promethazine. The first three compounds, however, protected at lower doses than promethazine (5 to 10 ug/kg). The protective effect of cyproheptadine lasted longer than 24 hr. The antianaphylactic effects of promethazine, mepyramine, chlorcyclizine, tri- pelennamine and diphenhydramine have been investigated by Armitage, Herxheimer & Rosa (1952). In this paper we describe investigations of a few of the newer antihistamines. METHODS The microshock method (Herxheimer, 1952) was used and the protection against anaphylactic shock was calculated according to Armitage et al. (1952) with the formula p=100(1-c/t), in which p is the percentage of full protection, c the control preconvulsion time and t the preconvulsion time of the animal under the influence of the drug. The compounds investigated were trimeprazine, methotrimeprazine, cyproheptadine, homo- chlorcyclizine and 10(-3-diethylamino-2-methylpropyl)phenothiazine 1,1-dioxide hydrochloride (oxomemazine hydrochloride, 6847 R.P.). They were injected intramuscularly in aqueous solution; the exposure to the antigen (an aerosol of 5% albumen solution) was done 1 hr later. For cyproheptadine, the exposure was delayed by between 1 and 48 hr in order to investigate the duration of the effect. -

Second-generation antipsychotic drugs can also result in fewer extrapyramidal side effects, but can induce weight gain. The authors' conclusions reflected the evidence presented, but some potential methodological flaws in the review process meant that the extent to which those conclusions were reliable was unclear. The response to nicotine was antagonized by the nicotinic receptor antagonist, hexamethonium but was insensitive to tetrodotoxin. 2 The nicotine-induced contraction was potentiated by the cholinesterase inhibitor, physostig- mine, and was reduced to 50% and 70% by the muscarinic cholinoceptor antagonist, atropine and the sympathetic neurone blocking drug, guanethidine, respectively. Chemical denervation with 6- hydroxydopamine abolished the inhibitory effect of guanethidine. Simultaneous treatment with atropine and guanethidine did not abolish the response to nicotine, but the degree of inhibition was comparable to that obtained with atropine alone. 3 The nicotine-induced contraction was insensitive to bunazosin and yohimbine (al- and Cc2-adrenoceptor antagonists, respectively), and exogenously applied noradrenaline did not cause a contraction even in the presence of blockade of noradrenaline uptake mechanisms with desipramine and normetanephrine and of fi-adrenoceptors with propranolol, suggesting a non-adrenergic nature of the sympathomimetic effect of nicotine in this tissue. 4 The nicotine-induced contraction in the presence of atropine was abolished after desensitization of P2-purinoceptors with a, ,B-methylene adenosine 5'-triphosphate, a slowly degradable ATP ana- logue selective for P2-purinoceptors. By this desensitization, the response to ATP, but not to hista- mine, was also abolished. 5 A cyclo-oxygenase inhibitor flurbiprofen partially inhibited the nicotine-induced contraction.
