A system of specific 16 primers and a Minor Groove Binding (MGB) TaqMan probe based on 17 sequences of the mitochondrial cytochrome b region was designed. The 18 method was successfully tested in 81 specimens of Scomber scombrus and 19 related species and validated in 26 different commercial samples. An average 20 Threshold cycle (Ct) value of 15.3 was obtained with Scomber scombrus DNA. 21 With the other species tested fluorescence signal was not detected or Ct was 22 significantly higher (P<0.001). The efficiency of the assay was estimated to be 23 92.41%, with 100% specificity, and no cross reactivity was detected with any 24 other species. These results reveal that the developed method is a rapid and 25 efficient tool to unequivocally identify S. -

Knowledge of reproductive parameters is necessary to understand the ecology, the population dynamics and to enable rational management of fish of economic interest. This work is a contribution to the study of some aspects of the reproductive biology of the Atlantic mackerel Scomber colias (Gmelin, 1789) in the Mediterranean Moroccan coast. The study is based on samples taken from commercial catches in M'diq Bay. The spawning period was determined following both the monthly changes of the gonadosomatic index and the histological maturity stages. The Atlantic chub mackerel spawn between November and March, with maximum activity in December. 1 Introduction The Atlantic chub mackerel Scomber colias (Gmelin, 1789) is a costal pelagic species. It is frequent over the continental slope from the surface to 300 m depth. The S. Colias is a gregarious and migratory species. It is widely distributed in the warm waters of the Atlantic Ocean, the Mediterranean Sea and the Black Sea. In the eastern Atlantic, chub mackerel occurs between the Bay of Biscay and South Africa [1]. Recently, genetic divergence and phenotypic variation has been demonstrated that Scomber japonicus and Scomber colias were distinct. -
