It is an opportunistic intracellular pathogenic microorganism that has become crucial reason for human food borne infections worldwide. It causes Listeriosis, the disease that can be serious and fatal to human and animals. Listeria outbreaks are often linked to dairy products, raw vegetables, raw meat and smoked fish, raw milk. The most effected country by Listeriosis is United States. CDC estimated that 1600 people get Listeriosis annually and regarding 260 die. It additionally contributes to negative economic impact because of the value of surveillance, investigation, treatment and prevention of sickness. The analysis of food products for presence of pathogenic microorganisms is one among the fundamental steps to regulate safety of food. This article intends to review the status of its introduction, characteristics, outbreaks, symptoms, prevention and treatment, more importantly to controlling the Listeriosis and its safety measures. Eight phages were selected for further characterization to determine the most appropriate phage for use in a detection assay. They were characterized for host range, TEM, stability to air dying for 24 h at 25 °C, and restriction endonuclease pattern. L. monocytogenes strain C716 was very resistant to all phages, however; a mutated phage, AG13M, was able to infect L. monocytogenes strain C716. AG20 & AG23 phages were high specificity against their host (Listeria spp). AG20 phage was immobilized on ColorLok paper and used in the to detect L. monocytogenes C519 in broth and FCS. AG20 phage was able to detect as few as 50 CFU/mL of L. monocytogenes in TSB and 40 CFU/cm2 on FCS using a plaque assay to detect progeny phage within 24 h. Pettengill and Hugh Rand Biostatistics and Bioinformatics Staff, Office of Analytics and Outreach, US Food and Drug Administration, College Park, MD, United States of America ABSTRACT Obtaining human population level estimates of the prevalence of foodborne pathogens is critical for understanding outbreaks and ameliorating such threats to public health. Estimates are difficult to obtain due to logistic and financial constraints, but citizen science initiatives like that of the American Gut Project (AGP) represent a potential source of information concerning enteric pathogens. With an emphasis on genera Listeria and Salmonella, we sought to document the prevalence of those two taxa within the AGP samples. The results provided by AGP suggest a surprising 14% and 2% of samples contained Salmonella and Listeria, respectively. However, a reanalysis of those AGP sequences described here indicated that results depend greatly on the algorithm for assigning taxonomy and differences persisted across both a range of parameter settings and different reference databases (i.e., Greengenes and HITdb). These results are perhaps to be expected given that AGP sequenced the V4 region of 16S rRNA gene, which may not provide good resolution at the lower taxonomic levels (e.g., species), but it was surprising how often methods differ in classifying reads—even at higher taxonomic ranks (e.g., family). This highlights the misleading conclusions that can be reached when relying on a single method that is not a gold standard; this is the essence Submitted 8 February 2017 of Segal's Law: an individual with one watch knows what time it is but an individual Accepted 31 May 2017 with two is never sure. -

Embora os tratamentos convencionais para combater infeções microbianas sejam muito eficazes, geram subpopulações bacterianas resistentes. O controlo da virulência bacteriana e a modulação da resposta do hospedeiro surgem como alternativas terapêuticas promissoras para combater as infeções. Listeria monocytogenes é uma ameaça recorrente para a saúde pública e para a indústria alimentar. É um patogénio Gram-positivo e intracelular de origem alimentar e um excelente modelo para o estudo da interação do hospedeiro com o patogénio. Esta bactéria tem a capacidade de atravessar as barreiras intestinal, hematoencefálica e materno-fetal e colonizar tecidos do hospedeiro podendo causar listeriose. Esta capacidade é alcançada através da expressão de inúmeros fatores de virulência que permite L. monocytogenes invadir, sobreviver e multiplicar dentro de células fagocíticas e não-fagocíticas. A análise do modo como este patogénio manipula as funções celulares do hospedeiro, leva à identificação de novos mecanismos que podem ser alargados a outros patogénios relevantes e ajuda na criação de novas estratégias terapêuticas. Deste modo, este trabalho teve como objetivo identificar e caracterizar novos mecanismos de virulência de L.
